Evaluation of drought indices to identify tolerant genotypes in common bean bush(Phaseolus vulgaris L.)

drought is one of the major abiotic stresses often causing negative impacts on bean crops in the Andean region in Colombia. An experiment under the greenhouse conditions was carried out to assess the effect of a prolonged drought period(15 days) at two different phenological stages(vegetative or reproductive) on grain yield and yield components of five bush bean cultivars(ICA-Cerinza, Bachue, NUA35, Bianca, and Bacatá). Nine tolerance indices including stress susceptibility index(SSI), tolerance(TOL), mean productivity(MP), geometric mean productivity(GMP), stress tolerance index(STI), yield stability index(YSI), yield index(YI), Harmonic mean(HM), and drought sensitivity index(dSI) were calculated based on grain yield under non-stressed(YP) and drought(YS) conditions. Based on the different drought indices, genotypes ICA-Cerinza and NUA35 had the best performance under drought conditions in both studied phases, which reflected in a reduction of grain yield ~≤40%. The biplot analysis also showed a clear superiority of these two genotypes at both phenological stages. Results also showed that SSI was more effective to identify genotypes less affected by drought. The above results allowed us to conclude that ICA-Cerinza and NUA35 may be considered for agricultural areas where long periods of water deficit are expected and can be used in breeding programs for drought tolerance.

An enhancement in the wheel-rail contact model used in a nonlinear vehicle-structure interaction(VSI)methodology for railway applications is presented,in which the detection of the contact points between wheel and rail in the concave region of the thread-flange transition is implemented in a simplified *** presenting the enhanced formulation,the model is validated with two numerical applications(namely,the Manchester Benchmarks and a hunting stability problem of a sus-pended wheelset),and one experimental test performed in a test rig from the Railway Technical Research Institute(RTRI)in *** its finite element(FE)nature,and contrary to most of the vehicle multibody dynamic commercial software that cannot account for the infrastructure flexibility,the proposed VSI model can be easily used in the study of train-bridge systems with any degree of *** validation presented in this work proves the accuracy of the proposed model,making it a suitable tool for dealing with different railway dynamic applications,such as the study of bridge dynamics,train running safety under different scenarios(namely,earthquakes and crosswinds,among others),and passenger riding comfort.

In this paper we present a study into the cleaning properties of synthetic dry adhesives. We have manufactured the adhesive micro-fibres through a low-cost, high yield fabrication method using Sylgard 184 Polydimethylsiloxane (PdMS) as the structural material. We deliberately contaminated the adhesive samples with different sized particles in the micro and macro scales and tested different cleaning methods for their efficacy with respect to each particle size. We investigated different cleaning methods, which included the use of wax moulding, vibration and pressure sensitive adhesives. For adhesion testing we used a custom system with a linear stage and a force sensor indenting a hemispherical probe into the adhesive surface and measuring the pull-off force. To characterize the cleaning efficacy we visually inspected each sample in a microscope and weighed the samples with a microgram-accuracy analytical balance. Results showed that the moulding method induced adhesion recovery in a greater percentage than the other cleaning methods and even helped with the recovery of collapsed posts in some cases. On the other hand pressure sensitive adhesives seem to have the upper hand with regards to certain particle sizes that can potentially pose problems with the moulding method.

We demonstrate improved peptide linkers which allow both conjugation to biomolecules such as dNA and self-assembly with luminescent semiconductor quantum dots.A hexahistidine peptidyl sequence was generated by standard solid phase peptide synthesis and modified with the succinimidyl ester of iodoacetamide to yield a thiol-reactive iodoacetyl polyhistidine *** reactive peptide was conjugated to dye-labeled thiolated dNA which was utilized as a model target *** gel electrophoresis and fluorescence resonance energy transfer analysis confirmed that the linker allowed the dNA to self-assemble with quantum dots via metal-affinity driven *** contrast to previous peptidyl linkers that were based on disulfide exchange and were thus labile to reduction,the reactive haloacetyl chemistry demonstrated here results in a more stable thioether bond linking the dNA to the peptide which can withstand strongly reducing environments such as the intracellular *** thiol groups occur naturally in proteins,can be engineered into cloned proteins,inserted into nascent peptides or added to dNA during synthesis,the chemistry demonstrated here can provide a simple method for self-assembling a variety of stable quantum dot bioconjugates.
