Salt stress can be alleviated by straw layer burial in the soil, but little is known of the appropriate form of the straw layer for optimal regulation of soil water and salinity because of the uncontrollability of field tests. Here, the following four straw forms with compaction thickness of 5 cm buried 40–45 deep were studied: no straw layer(CK), segmented straw(SL, 5 cm in length), straw pellet(SK), and straw powder(sf). The three straw forms(SL, SK and sf) significantly delayed the infiltration of irrigation water down the column profile by 71.20–134.3 h relative to CK and the migration velocity of the wetting front under sf was the slowest. It took longer for the wetting front to transcend SK than SL but shorter for it to reach the bottom of soil column after water crossed the straw layer. Compared with CK, the average volumetric water content in the 0–40 cm soil layer increased by 6.45% under SL, 1.77% under SK and 5.39% under sf. The desalination rates at the 0–40 and 0–100 cm soil layers increased by 5.85 and 3.76% under SL, 6.64 and 1.47% under SK and 5.97 and 4.82% under sf. However, there was no significant difference among straw forms in the 0–40 cm soil layer. Furthermore, the salt leaching efficiency(SLE, g mm^–1 h^–1) above the 40 cm layer under SL was 0.0097, being significantly higher than that under sf(0.0071) by 37.23%. Salt storage under SL, SK and sf in the 40–45 cm layer accounted for 4.50, 16.92 and 7.43% of total storage in the 1-m column profile. Cumulative evaporation under SL and sf decreased significantly by 41.20 and 49.00%, with both treatments having the most significant inhibition of salt accumulation(resalinization rate being 36.06 and 47.15% lower than CK) in the 0–40 cm soil layer. In conclusion, the different forms of straw layers have desalting effects under high irrigation level(446 mm). In particular, SL and sf performed better than SK in promoting deep salt leaching and inhibiting salt accumulation on the soil surface. However,

The Guanine nucleotide exchange factor Vav2（Vav2） is a member of the Vav family that serves as an important regulators for the Rho family of Ras-related GTPases. In the current study, an ortholog（Lj-Vav2） of Vav2 was identified in the lamprey（Lampetra japonica）. To elucidate the phylogenetic relationship of Vav2, the metazoan genome databases were analyzed to mine the ortholog of Vav. It was found that Vav2 genes were only existed in vertebrates and Lj-Vav2 was the original one found in agnathans. The evolutionary dynamics of conserved motifs of Vav2 were explored using combined amino acid sequence as markers, and it is revealed that the Calponin homology（CH） domain, Dbl-homologous（DH） domain, Pleckstrin homology（PH） domain, Cysteine-rich（C1）domains, Src homology 3（SH3） domains and Src homology 2（SH2） domain were conserved throughout the Vav2 gene family in vertebrates during gene evolution. Relative quantitative real-time PCR analysis showed that the LjVav2 was distributed in the heart, kidney, supraneural myeloid body, liver, gill and lymphocyte-like cells. The LjVav2 was found to be expressed in these tissues, and the level of which was upregulated in lymphocyte-like cells after the animal was stimulated with LPS. These results indicated that the Lj-Vav2 might be involved in the immune response of lymphocyte-like cells in lamprey. Meanwhile, our findings provided a foundation for further investigation of the function of Lj-Vav2 in the primary vertebrate.

Argatroban is a synthetic thrombin inhibitor approved by *** and Drug Administration for the treatment of ***,whether it plays a role in the repair of spinal cord injury is *** this study,we established a rat model of T10 moderate spinal cord injury using an NYU Impactor ModerⅢand performed intraperitoneal injection of argatroban for 3 consecutive *** results showed that argatroban effectively promoted neurological function recovery after spinal cord injury and decreased thrombin expression and activity in the local injured spinal *** sequencing transcriptomic analysis revealed that the differentially expressed genes in the argatroban-treated group were enriched in the JAK2/STAT3 pathway,which is involved in astrogliosis and glial scar *** blotting and immunofluorescence results showed that argatroban downregulated the expression of the thrombin receptor PAR1 in the injured spinal cord and the JAK2/STAT3 signal *** also inhibited the activation and proliferation of astrocytes and reduced glial scar formation in the spinal *** together,these findings suggest that argatroban may inhibit astrogliosis by inhibiting the thrombin-mediated PAR1/JAK2/STAT3 signal pathway,thereby promoting the recovery of neurological function after spinal cord injury.

Identification of SPP1 as an extracellular matrix signature for metastatic castration-resistant prostate cancer

OBJECTIVE Resistance to androgen deprivation therapy(ADT) is the main challenge for advanced fatal prostate cancer(PCa), which can gradually develop into metastatic castration-resistant prostate cancer(m CRPC). However, the pathologic mechanisms of m CRPC are still far from clear. Given the high incidence and mortality related to m CRPC, understanding the causes and pathogenesis of this condition as well as identifying potential biomarkers are of great *** In the research reported here, we integrated several gene expression profiles from hormone sensitive prostate cancer(HSPC) and m CRPC datasets to identify differentially expressed genes(DEGs), key biological pathways, and cellular components. K-nearest neighbor,logistic regression, naive Bayes, and random forest classifiers models were established to identify key regulated genes. Furthermore, organoid models were employed to validate the results in vitro. RESULTS We found that extracellular matrix(ECM) genes were significantly enriched, and further filtered them using Pearson correlation analysis and stepwise regression to find ECM signatures to differentiate between the HSPC and m CRPC phenotypes. Six ECM signatures were input into K-nearest neighbor, logistic regression, naive Bayes, and random forest classifiers models. Random forest algorithm with the six-gene prognostic signature showed best performance, which had high sensitivity and specificity for HSPC and m CRPC classification. Among the six ECM genes, SPP1 was identified as the key hub signature for PCa metastasis and drug resistance development; we found that both protein and m RNA expression levels of SPP1 were remarkably up-regulated in m CRPC compared with HSPC in organoid models and could regulate the androgen receptor signaling pathway. CONCLUSION Therefore, SPP1 is a potential novel biomarker and therapeutic target for m CRPC. Yield improvement has long been an important task for soybean breeding in the world in order to meet the increasing demand for food and animal *** R396 genes have been shown to negatively regulate grain size in rice,but whether mi R396 family members may function in a similar manner in soybean is ***,we generated eight soybean mutants harboring different combinations of homozygous mutations in the six soybean mi R396genes through genome editing with clustered regularly interspaced palindromic repeats(CRISPR)/CRISPR-associated nuclease(Cas)12sf01 in the elite soybean cultivar Zhonghuang 302(ZH302).Four triple mutants(mir396aci,mir396acd,mir396adf,and mir396cdf),two quadruple mutants(mir396-abcd and mir396acfi),and two quintuple mutants(mir396abcdf and mir396bcdfi)were *** found that plants of all the mir396 mutants produced larger seeds compared to ZH302 *** tests showed that mir396adf and mir396cdf plants have significantly increased yield in growth zones with relatively high latitude which are suited for ZH302 and moderately increased yield in lower *** contrast,mir396abcdf and mir396bcdfiplants have increased plant height and decreased yield in growth zones with relatively high latitude due to lodging issues,but they are suited for low latitude growth zones with increased yield without lodging *** together,our study demonstrated that loss-of-function of mi R396 genes leads to significantly enlarged seed size and increased yield in soybean,providing valuable germplasms for breeding high-yield soybean.
