Hepatocellular carcinoma xenograft supports HCV replication:A mouse model for evaluating antivirals

AIM: To develop a hepatocellular carcinoma (HCC) xenograft model for studying hepatitis C virus (HCV) replication in a mice, and antiviral ***: We developed a stable S3-green fluorescence protein (GFP) cell line that replicated the GFP-tagged HCV sub-genomic RNA derived from a highly efficient JFH1 virus. S3-GFP replicon cell line was injected subcutaneously into γ-irradiated SCID mice. We showed that the S3-GFP replicon cell line formed human HCC xenografts in SCID mice. Cells were isolated from subcutaneous tumors and then serially passaged multiple times in SCID mice by culturing in growth medium supplemented with G-418. The mouse-adapted S3-GFP replicon cells were implanted subcutaneously and also into the liver of SCID mice via intrasplenic infusion to study the replication of HCV in the HCC xenografts. The tumor model was validated for antiviral testing after intraperitoneal injection of interferon-α (IFN-α). RESULTS: A highly tumorigenic S3-GFP replicon cell line was developed that formed subcutaneous tumors within 2 wk and diffuse liver metastasis within 4 wk in SCID mice. Replication of HCV in the subcutaneous and liver tumors was confirmed by cell colony assay, detection of the viral RNA by ribonuclease protection assay and real-time quantitative reverse transcription polymerase chain reaction. High-level replication of HCV sub-genomic RNA in the tumor could be visualized by GFP expression using fluorescence microscopy. IFN-α cleared HCV RNA replication in the subcutaneous tumors within 2 wk and 4 wk in the liver tumor model. CONCLUSION: A non-infectious mouse model allows us to study replication of HCV in subcutaneous and metastatic liver tumors. Clearance of HCV by IFN-α supports use of this model to test other anti-HCV drugs.

Growth Rate and Chemical Muscle Composition of Oreochromis niloticus Fish Cultured in Drainage Water and Fed Antioxidant Supplemented Diet

Irrigation of fish farms with agricultural drainage water may affect cultured fish species. So, the present study utilizes antioxidants supplemented diet to overcome deterioration of drainage water and its negative effect on fish. The studied groups are fish cultured in dechlorinated tap water as control group or drainage water and fed commercial basal diet. While, the other studied groups represented by Oreochromis niloticus fish cultured in drainage water and fed either commercial diet supplemented with vitamin C (5 g/kg diet) or fennel (5 g/kg diet) for 12 weeks. Results of the water physico-chemical parameters of all studied treatments revealed deterioration of the drainage water with a decrease in dissolved oxygen and an increase in pH, total hardness, total alkalinity, salinity, ammonia, nitrite and heavy metals (Cu, Pb and Cd) with significant differences (P ≤ 0.01) in comparison to that of the control dechlorinated tap water group. Data clarified also that Oreochromis niloticus cultured in drainage water showed a decrease in growth rate accompanied by deterioration of fish meat quality. However, fish reared in the same drainage water for the same exposure period and fed vitamin C or fennel supplemented basal diet (5 g/kg diet) recorded values of the studied parameters more or less similar to that of control group fish. Data of the present study, empowered aquaculturist to supplement fish rations with fennel or vitamin C as antioxidants to improve fish growth rate, meat quality as well as protect fish against heavy metals toxicity that could threat Human Being.

Background: In India, undergraduate medical education has remained more or less static and for last so many decades no effective development has been done. Therefore this study is done for restructuring it and to meet the challenges faced thereby. Material and methods: Depending on the problems faced during undergraduate pathology teaching, questions were prepared and sent to pathology teachers of Odisha. In few aspects, suggestions were also asked for modification of the present system of medical curriculum including the problem of factual or information overload on students. Results: 18 questions were prepared and asked to 96 pathology teachers. The opinions mostly were to introduce small group teaching or problem based learning (PBL) and multidisciplinary integrated teaching. The opinion of students of 3rd and 4th MBBS were also taken into consideration for taking the decisions of modification. Conclusion: Efforts should be taken to bring the desired changes in undergraduate pathology education to produce need based human resources for health care in society.

Role of hydrazine and hydrogen peroxide in aluminium hydroxide precipitation from sodium aluminate solution

Aluminium hydroxide precipitation from synthetic sodium aluminate solution was studied in the presence of hydrazine or hydrogen peroxide. The addition of low concentration of hydrazine is found to be effective, while higher amount of hydrogen peroxide is required to generate similar effect. XRD data confirm the product phase to be gibbsitic by nature. The scanning electron micrographs （SEM） show that agglomerated products form in the presence of hydrazine while fine discrete particles are produced with hydrogen peroxide. The probable mechanism of precipitation in the presence of hydrazine and hydrogen peroxide is also discussed.

Morbidity Profile and Causes of Mortality in Type 2 Diabetes Patients: Data from a Tertiary Teaching Hospital from Eastern India

Background: Data regarding the prevalence of morbidity and mortality in patients of Type 2 Diabetes Mellitus (T2DM) is scanty in India. Objectives: To determine the prevalence of micro and macro vascular complications, acute metabolic complications, infections, Non Alcoholic Fatty Liver Disease (NAFLD) and cause of mortality in T2DM patients admitted to a tertiary care teaching hospital in Eastern India. Material and Methods: This was a hospital-based prospective study evaluating 150 T2DM patients admitted to a tertiary care institution in Eastern India. Diagnosis of micro and macro vascular complications, infections and NAFLD was made using standard protocols. In case of death, the most probable cause was noted. Results: Out of 150 patients, 14.7% of patients were newly diagnosed T2DM and out of them 41% of patients had vascular complications and 54.5% had infections. Of the total patients, 56% had nephropathy, 20% neuropathy, 17.3% retinopathy, 31.3% CVD, 11.3% CAD, 4.6% acute metabolic complications, 44% infections and 16.6% had NAFLD respectively. Macrovascular events occured earlier than microvascular complications. Multiple logistic regression analysis showed strong association of age, duration of diabetes, serum cholesterol, triglyceride, LDL-C with retinopathy (Regression coefficient β: -0.1086807, 0.4127152, -0.0513393, 0.0146429, 0.0587475;p < 0.05, < 0.001, < 0.05, < 0.05, < 0.05 respectively), while only duration of diabetes was strongly associated with nephropathy and neuropathy (Regression coefficient β: 0.2538751, 0.2261636;p < 0.001 for each). Increasing age was associated with CAD (Regression coefficient β: 0.055392;p β: 0.0055014;p 18.6% patients died due to diabetes related complications. Cardiovascular (CV)-related deaths (CVD+CAD) were most common cause (51.5%: CVD 36.4%, CAD 15.1%) to be followed by infections (27.3%) and then chronic kidney disease (12.1%). Objective:To investigation of cytotoxic,antimicrobial and analgesic activities of different fractions of Rhaphidophora glauca(Wall.)***:Two partially purified aqueous methanolic fractions from ethyl acetate extract(AMF-1)and chloroform extract(AMF-2)obtained from the partitioning were used in *** cytotoxic effect was determined by brine shrimp lethality *** activity was investigated by disc diffusion and minimal inhibitory concentration *** plate method and acetic acid test was used for determining analgesic ***:The LC_(50)values of AMF-1 and AMF-2 were found to be 287.73 and 428.54μg/mL respectively,where colchicines showed LC_(50)of 11.16μg/*** zone of inhibition of the fractions AMF-1 and AMF-2 was found to be in the range of 8-26 mm in 2000μg/disc,as compared to reference antibiotics kanamycin(11-28 mm at 30μg/disc)and ciprofloxacin(20-25 mm at 30μg/disc)indicating the antibacterial *** hot plate test,the highest pain inhibitory activity was found at a dose of 250 mg/kg for AMF-1 which was statistically significant(P<0.05)compared to both positive and negative control at 30 min *** acid induced model,both AMF-1 and AMF-2 at a dose of 500 mg/kg showed significant activity compared to positive and negative ***:This study found that Rhaphidophora glauca possesses potential cytotoxic,antibacterial and analgesic *** study may be needed to isolate the bioactive compounds responsible for different activities with subsequent mechanistic study.
