SFRP4 expression correlates with epithelial mesenchymal transitionlinked genes and poor overall survival in colon cancer patients

BACKGROUnD Colon cancer is among the most commonly diagnosed cancers in the United States with an estimated 97220 new cases expected by the end of *** affects 1.2 million people around the world and is responsible for about 0.6 million deaths every *** decline in overall incidence and mortality over the past 30 years,there continues to be an alarming rise in early-onset colon cancer cases(nts are often diagnosed at late stages of the disease and tend to have poor *** previously showed that the WnT“gatekeeper”gene,secreted frizzled-related protein 4(SFRP4),is over-expressed in early-onset colon ***4 is speculated to play an essential role in cancer by inhibiting the epithelial mesenchymal transition(EMT).AIM To investigate the correlation between SFRP4 expression and EMT-linked genes in colon cancer and how it affects patient *** SFRP4 expression relative to that of EMT-linked genes and survival analysis were performed using the University of California Santa Cruz Cancer Browser *** SFRP4 was found to be co-expressed with the EMT-linked markers CDH2,Fn1,VIM,TWIST1,TWIST2,SnAI1,SnAI2,ZEB1,ZEB2,POSTn,MMP2,MMP7,MMP9,and ***4 expression negatively correlated with the EMTlinked suppressors CLDn4,CLDn7,TJP3,MUC1,and *** expression of SFRP4 and the EMT-linked markers was higher in mesenchymal-like samples compared to epithelial-like samples which potentially implicates SFRP4-EMT mechanism in colon ***,patients overexpressing SFRP4 presented with poor overall survival(P=0.0293).COnCLUSIOn Considering the implication of SFRP4 in early-onset colon cancer,particularly in the context of EMT,tumor metastasis,and invasion,and the effect of increased expression on colon cancer patient survival,SFRP4 might be a potential biomarker for early-onset colon cancer that could be targeted for diagnosis and/or disease therapy.

The shiny cowbird Molothrus bonariensis parasitizes many species with different life-history traits and has a detrimental effect on the survival of the progeny of the *** response,hosts have developed numerous antiparasitic ***,we examined the effects of brood parasitism by shiny cowbird on the clutch and brood sizes(83 nests)in a small host,the black-backed water tyrant Fluvicola *** also studied whether the death of parasite nestlings was related to the care of the foster parents and whether the host had any antiparasitic defense against the shiny *** results indicate that brood parasitism significantly decreased the host hatching and fledging *** majority of nest failures(57%)were caused by brood *** cowbird parasitism occurred in 52%of nests and the intensity of parasitism was 1.23±0.53 eggs per parasitized *** the total host eggs,54%were *** the incubation stage,20 nests(47%)were abandoned because of egg punctures by shiny cowbirds *** two parasitic fledglings were recorded,while the remaining nestlings either died from starvation(n=12)or predation(n=3).Foster parents abandoned parasitic nestlings between 5 and 10 days *** findings demonstrate that the shiny cowbird has very low rates of fledging success when parasitizing black-backed water ***,parasitism had a high reproductive cost in the black-backed water tyrant because a very low proportion(7%)of the parasitized nests(n=43)were successful.

Minimally Invasive Surgery for necrotizing Pancreatitis: A Case Report

Introduction: necrotizing pancreatitis management is complex and varies significantly among clinicians. Minimally invasive approaches like transgastric necrosectomy via laparoscopy are emerging as effective treatment options. This case report underscores the technique’s efficacy, clinical outcomes, and role in reducing complications. Clinical Observation: A 59-year-old male with a history of smoking and alcoholism presented with severe abdominal pain, nausea, and vomiting. Over the following weeks, he developed symptoms including asthenia, weight loss, and melena. Diagnostic workup revealed severe anemia and Balthazar E necrotizing pancreatitis, with significant intra-abdominal fluid collections and signs of infection. After initial conservative management, the patient underwent transgastric necrosectomy via laparoscopy due to deteriorating clinical status. The procedure involved removing necrotic tissue and performing a cystogastroanastomosis and jejunostomy. Postoperative care included fasting, parenteral nutrition, broad-spectrum antibiotics, and enzymatic replacement. The patient recovered well, with reduced necrotic tissue on follow-up imaging, and was discharged twelve days post-surgery [1]. Conclusion: Transgastric necrosectomy by laparoscopy is a valuable first-line surgical option for patients with symptomatic necrotizing pancreatitis, particularly in cases without prior interventions. This minimally invasive technique helps reduce major complications and mortality, offering a less invasive alternative to traditional open necrosectomy. The multidisciplinary approach and careful postoperative management were crucial to the patient’s favorable outcome. The case highlights the potential of transgastric necrosectomy as an effective treatment strategy in managing complex pancreatitis cases, including those with associated duodenal perforation [2].

Ecological succession of benthic organisms on niche-type artificial reefs

As part of efforts to restore fishery resources and recover damaged coastal ecosystems,artificial reefs are often anchored on the seafloor in coastal zones,to provide new habitats for marine *** aim of the study was to describe the structure of a community of benthic invertebrates colonising a niche-type artificial reef(AATn in Spanish).nine structures were anchored at depths of 16±1.5 m for 99 weeks(22 months)in the Area of Benthic Resource Management and Exploitation(AMERB)in coastal waters of the Region of Bio Bio,*** results showed that,at 3 months from submersion of the nTAR,much of the artificial substrate remained bare and there were only low levels of specimens of Balanus ***,showing mean coverage of 11.26%,and even lower proportions of Rhodophyta,with mean coverage of 0.34%.At 6 months,the presence of hydrozoans was seen,decreasing the coverage of the barnacles,which was aided further at 8 months with the arrival of barnacle predatory *** 8 months,new benthic invertebrates appeared and competed for the *** included sponges and *** 99 weeks,the hydrozoans dominated the substrate,followed by barnacles and Rhodophyta,the first colonising organisms,leading to colonisation by motile macro-invertebrates,mainly consisting of crustaceans,echinoderms and *** AATn artificial system provides an ideal substrate for the development of early ecological succession and the use of this technology should be feasible in the recovery process of habitat damaged by anthropogenic actions and climate change.

Alcohol-induced alterations of the hepatocyte cytoskeleton

The hepatocyte cytoskeleton consists of three fi lamentous networks: microtubules, actin microfi laments and keratin intermediate filaments. Because of the abundance of the proteins that comprise each system and the central role each network plays in a variety of cellular processes, the three fi lament systems have been the focus of a host of studies aimed at understanding the progression of alcohol-induced liver injury. In this review, we will briefly discuss the hepatic organization of each cytoskeletal network and highlight some components of each system. We will also describe what is known about ethanol-induced changes in the dynamics and distributions of each cytoskeletal system and discuss what is known about changes in protein expression levels and post-translational modifi cations. Finally, we will describe the possible consequences of these cytoskeletal alterations on hepatocyte function and how they might contribute to the progression of liver disease.

Expression of genes that control core fucosylation in hepatocellular carcinoma: Systematic review

BACKGROUnD Changes in n-linked glycosylation have been observed in the circulation of individuals with hepatocellular carcinoma. In particular, an elevation in the level of core fucosylation has been observed. However, the mechanisms through which core fucose is increased are not well understood. We hypothesized that a review of the literature and related bioinformatic review regarding six genes known to be involved in the attachment of core fucosylation, the synthesis of the fucosylation substrate guanosine diphosphate(GDP)-fucose, or the transport of the substrate into the Golgi might offer mechanistic insight into the regulation of core fucose *** To survey the literature to capture the involvement of genes regulating core nlinked fucosylation in hepatocellular carcinoma METHODS The PubMed biomedical literature database was searched for the association of hepatocellular carcinoma and each of the core fucose-related genes and their protein products. We also queried The Cancer Genome Atlas Liver hepatocellular carcinoma(LIHC) dataset for genetic, epigenetic and gene expression changes for the set of six genes using the tools at *** A total of 27 citations involving one or more of the core fucosylation-related genes(FPGT, FUK, FUT8, GMDS, SLC35 C1, TSTA3) and hepatocellular carcinoma were identified. The same set of gene symbols was used to query the371 patients with liver cancer in the LIHC dataset to identify the frequency of m RnA over or under expression, as well as non-synonymous mutations, copy number variation and methylation level. Although all six genes trended to moresamples displaying over expression relative to under-expression, it was noted that a number of tumor samples had undergone amplification of the genes of the de novo synthesis pathway, GMDS(27 samples) and TSTA3(78 samples). Strong Suppression of Systemic Acquired Resistance in Arabidopsis by nRR is Dependent on its Ability to Interact with nPRland its Putative Repression Domain

Systemic Acquired Resistance （SAR） in plants confers lasting broad-spectrum resistance to pathogens and requires the phytohormone salicylic acid （SA）. Arabidopsis nPR1/nIM1 is a key regulator of the SAR response. Studies attempting to reveal the function of nPR1 and how it mediates SA signaling have led to isolation of two classes of proteins that interact with nPRI＂ the first class includes rice nRR, Arabidopsis nIMIn1, nIMIn2, and nIMIn3, and tobacco nIMIn2-1ike proteins; the second class belongs to TGA transcription factors. We have previously shown that overexpression of n RR in rice suppresses both basal and Xa21-mediated resistance. In order to test whether nRR affects SA-induced, nPRl-mediated SAR, we have transformed Arabidopsis with the rice nRR gene and tested its effects on the defense response. Expression of nRR in Arabidopsis results in suppression of PR gene induction by SAR inducer and resistance to pathogens. These phenotypes are even more severe than those of the nprl-1 mutant. The ability of nRR to suppress PR gene induction and disease resistance is correlated with its ability to bind to nPR1 because two point mutations in nRR, which reduce nPR1 binding, fail to suppress nPR1. In contrast, wild-type and a mutant nRR, which still binds to nPR1 strongly, retain the ability to suppress the SAR response. Replacing the C-terminal 79 amino acids of nRR with the VP16 activation domain turns the fusion protein into a transcriptional co-activator. These results indicate that nRR binds to nPR1 in vivo in a protein complex to inhibit transcriptional activation of PR genes and that nRR contains a transcription repression domain for active repression.

Immunotherapy with immune checkpoint inhibitors in colorectal cancer:what is the future beyond deficient mismatch-repair tumours?

Following initial success in melanoma and lung tumours,immune checkpoint inhibitors(ICIs)are now well recognized as a major immunotherapy treatment modality for multiple types of solid *** colorectal cancer(CRC),the small subset that is mismatch-repair-deficient and microsatellite-instability-high(dMMR/MSI-H)derive benefit from immunotherapy;however,the vast majority of patients with proficient MMR(pMMR)or with microsatellite stable(MSS)CRC do *** and the consensus molecular subtype classifications are promising biomarkers in predicting therapeutic efficacy in selected *** pMRR/MSS CRC,biomarkers are also needed to understand the molecular mechanisms governing immune reactivity and to predict their relationship to *** continuous development of such biomarkers would offer new perspectives and more personalized treatments by targeting oncological options,including ICIs,which modify the tumour-immune *** this review,we focus on CRC and discuss the current status of ICIs,the role of biomarkers to predict response to immunotherapy,and the approaches being explored to render pMMR/MSS CRC more immunogenic through the use of combined therapies.
