Impact of source tissue and ex vivo expansion on the characterization of goat mesenchymal stem cells

Background： There is considerable interest in using goats as models for genetically engineering dairy animals and also for using stem cells as therapeutics for bone and cartilage repair. Mesenchymal stem cells （MSCs） have been isolated and characterized from various species, but are poorly characterized in goats. Results： Goat MSCs isolated from bone marrow （BM-MSCs） and adipose tissue （ASCs） have the ability to undergo osteogenic, adipogenic and chondrogenic differentiation. Cytochemical staining and gene expression analysis show that ASCs have a greater capacity for adipogenic differentiation compared to BM-MSCs and fibroblasts. Different methods of inducing adipogenesis also affect the extent and profile of adipogenic differentiation in MSCs. Goat fibroblasts were not capable of osteogenesis, hence distinguishing them from the MSCs. Goat MSCs and fibroblasts express CD90, CD105, CD73 but not CD45, and exhibit cytoplasmic localization of OCT4 protein. Goat MSCs can be stably transfected by Nucleofection, but, as evidenced by colony-forming efficiency （CFE）, yield significantly different levels of progenitor cells that are robust enough to proliferate into colonies of integrants following G418 selection. BM-MSCs expanded over increasing passages in vitro maintained karyotypic stability up to 20 passages in culture, exhibited an increase in adipogenic differentiation and CFE, but showed altered morphology and amenability to genetic modification by selection. Conclusions： Our findings provide characterization information on goat MSCs, and show that there can be significant differences between MSCs isolated from different tissues and from within the same tissue. Fibroblasts do not exhibit trilineage differentiation potential at the same capacity as MSCs, making it a more reliable method for distinguishing MSCs from fibroblasts, compared to cell surface marker expression.

The role of the most common land iguana(Conolophus subcristatus)in the Galápagos Islands as an effective seed disperser is explored in this study.A total of 5705 seeds of 32 plant species were identified from 160 scats,4545 of which(80%)appeared visually *** trials of 849 seeds from 29 species revealed that at least 10 species remained viable after passing through the iguana’s gut,although only a small proportion of those seeds(4%)*** any case,we argue that *** exerts an important role on the 7 Galapagos islands where it occurs because of its abundance and capacity to ingest and disperse seeds at long *** results strongly suggest that the Galápagos *** plays an important role as a seed disperser of not only of native species but also some introduced plants in the Galápagos Islands.

Frugivory and seed dispersal in the Galápagos:what is the state of the art?

The Galápagos are considered a model oceanic archipelago,with unique flora and fauna currently threatened by alien invasive *** dispersal is an important ecosystem function with consequences for plant population dynamics and vegetation ***,understanding the seed dispersal abilities of the assemblages of frugivores will inform scientists and managers of the dynamics of plant invasions and improve management *** we provide the first comprehensive review of published information on frugivory and animal seed dispersal in the Galá*** collected data from a variety of sources,including notes of the first naturalist expeditions,gray literature available only in Galápagos collections,and peer-reviewed journal ***–animal frugivorous interactions were retrieved from 43 studies and compiled into an interaction matrix describing 366 unique *** studies focused on fruit consumption as a driving force for natural selection,but seed fate was seldom *** most(71%)of the interactions involved native plants,more than one-quarter(28%)involved introduced *** involving birds are considerably more common than those of reptiles and mammals,probably reflecting a research bias towards *** the historical importance of the archipelago as the laboratory for evolutionary and ecological research,understanding of its seed dispersal systems is *** end the review by suggesting 3 priority areas of research on frugivory and seed dispersal in the Galápagos:(i)target research to close knowledge gaps;(ii)the use of a network approach to frame seed dispersal at the community level;and(iii)evaluation of the effect of seed dispersal as a selective pressure acting upon plants and ***,the output of this research has to be properly delivered to the Galápagos National Park Services to help increase management effectiveness.

Phytochrome Signaling in Green Arabidopsis Seedlings： Impact Assessment of a Mutually Negative phyB-PIF Feedback Loop

The reversibly red （R）/far-red （FR）-Iight-responsive phytochrome （phy） photosensory system initiates both the deetiolation process in dark-germinated seedlings upon first exposure to light, and the shade-avoidance process in fully deetiolated seedlings upon exposure to vegetational shade. The intracellular signaling pathway from the light-activated photoreceptor conformer （Pfr） to the transcriptional network that drives these responses involves direct, physical inter- action of Pfr with a small subfamily of bHLH transcription factors, termed Phy-lnteracting Factors （PIFs）, which induces rapid PIF proteolytic degradation. In addition, there is evidence of further complexity in light-grown seedlings, whereby phyB-PIF interaction reciprocally induces phyB degradation, in a mutually-negative, feedback-loop configuration. Here, to assess the relative contributions of these antagonistic activities to the net phenotypic readout in light-grown seedlings, we have examined the magnitude of the light- and simulated-shade-induced responses of a pentuple phyBpiflpif3pif4pif5 （phyBpifq） mutant and various multiple pif-mutant combinations. The data （1） reaffirm that phyB is the predominant, if not exclusive, photoreceptor imposing the inhibition of hypocotyl elongation in deetiolating seedlings in response to pro- longed continuous R irradiation and （2） show that the PIF quartet （PIF1, PIF3, PIF4, and PIF5） retain and exert a dual capacity to modulate hypocotyl elongation under these conditions, by concomitantly promoting cell elongation through intrinsic transcriptional-regulatory activity, and reducing phyB-inhibitory capacity through feedback-loop-induced phyB degrada- tion. In shade-exposed seedlings, immunoblot analysis shows that the shade-imposed reduction in Pfr levels induces increases in the abundance of PIF3, and mutant analysis indicates that PIF3 acts, in conjunction with PIF4 and PIF5, to promote the known shade-induced acceleration of hypocotyl elongation. Factors affecting the dominance hierarchy dynamics in a hummingbird assemblage

Intra and in terspecific competiti on for n ectar play an imports nt role in hummingbird communities. Larger sized species usually exclude smaller species from the rich floral resources. However, it has been recently postulated that the competitive advantages of a large body size decline as the evolutionary distance between the contending species in creases. In this study, we analyzed dominance hierarchy dynamics in a hummingbird assemblage in central Mexico. By monitoring hummingbird territories established in three plant species through 1 year, we assessed the effects of energy within territories and the territory owners identity in the frequency of inter and intraspecific encounters. We also evaluated if these factors affect the dominance of larger species when they compete against smaller distantly related contenders. Our results show that their frequency of intraspecific encounters was related with the identity of the territory's owner. On the contrary, the frequency of interspecific encounters was related with both the territory and the identity of the territory's owner. We did not find a significant difference between the number of encounters dominated by larger and smaller species and their conte nders. However, the in crease in genetic dista nee between contenders was positively associated with a higher frequency of encounters dominated by small *** results showed that the ecological factors and evolutionary relationships among contenders play important roles in the dominance hierarchy dynamics.
