in biology, we continue to appreciate the fact that the DNA sequence alone falls short when attempting to explain the intricate inheritance patterns for complex traits. This is particularly true for human disorders that appear to have simple genetic causes. The study of epigenetics, and the increased access to the epigenetic profiles of different tissues has begun to shed light on the genetic complexity of many basic biological processes, both physiological and pathological. Epigenetics refers to heritable changes in gene expression that are not due to alterations in the DNA sequence. Various mechanisms of epigenetic regulation exist, including DNA methylation and histone modification. The identification, and increased understanding of key players and mechanisms of epigenetic regulation have begun to provide significant insight into the underlying origins of various human genetic disorders. One such disorder is CHARGE syndrome （OMiM #214800）, which is a leading cause of deaf-blindness worldwide. A majority of CHARGE syndrome cases are caused by haploinsufficiency for the CHD7 gene, which encodes an ATP-dependent chromatin remodeling protein involved in the epigenetic regulation of gene expression. The CHD7 protein has been highly conserved throughout evolution, and research into the function of CHD7 homologs in multiple model systems has increased our understanding of this family of proteins, and epigenetic mechanisms in general. Here we provide a review of CHARGE syndrome, and discuss the epigenetic functions of CHD7 in humans and CHD7 homologs in model organisms.

Analysis of monitored ground support and rock mass response in a longwall tailgate entry

A comprehensive monitoring program was conducted to measure the rock mass displacements, support response, and stress changes at a longwall tailgate entry in West *** was initiated a few days after development of the gateroad entries and continued during passage of the longwall panels on both sides of the *** included overcore stress measurements of the initial stress within the rock mass, changes in cable bolt loading, standing support pressure, roof deformation, rib deformation,stress changes in the coal pillar, and changes in the full three-dimensional stress tensor within the rock mass at six locations around the monitoring *** the passage of the first longwall, stress measurements in the rock and coal detected minor changes in loading while minor changes were detected in roof *** a result of the relatively favorable stress and geological conditions, the support systems did not experience severe loading or rock deformation until the second panel approached within 10–15 m of the instrumented *** reaching the peak loading at about 50–75 mm of roof sag, the cable bolts started to unload, and load was transferred to the standing *** standing support system was able to maintain an adequate opening inby the shields to provide ventilation to the first crosscut inby the face, as *** results were used to calibrate modeled cable bolt response to field data, and to validate numerical modeling procedures that have been developed to evaluate entry support *** is concluded that the support system was more than adequate to control the roof of the tailgate up to the longwall face *** monitoring results have provided valuable data for the development and validation of support design strategies for longwall tailgate entries.

Assessing support alternatives for longwall gateroads subject to changing stress

Longwall gateroad entries are subject to changing horizontal and vertical stress induced by redistribution of loads around the extracted *** stress changes can result in significant deformation of the entries that may include roof sag,rib dilation,and floor *** operators install different types of supports to control the ground response and maintain safe access and ventilation of the longwall *** paper describes recent research aimed at quantifying the effect of longwall-induced stress changes on ground stability and using the information to assess support *** research included monitoring of ground and support interaction at several operating longwall mines in the U.S.,analysis and calibration of numerical models that adequately represent the bedded rock mass,and observation of the support systems and their response to changes in *** models were then used to investigate the impact of geology and stress conditions on ground deformation and support response for various depths of cover and geologic *** research results were summarized in two regression equations that can be used to estimate the likely roof deformation and height of roof yield due to longwall-induced stress *** information is then used to assess the ability of support systems to maintain the stability of the *** application of the method is demonstrated with a retrospective analysis of the support performance at an operating longwall mine that experienced a headgate roof *** method is shown to produce realistic estimates of gateroad entry stability and support performance,allowing alternative support systems to be assessed during the design and planning stage of longwall operations.

Linear logistic regression with weight thresholding for flow regime classification of a stratified wake

A stratified wake has multiple flow regimes,and exhibits different behaviors in these regimes due to the competing physical effects of momentum and *** work aims at automated classification of the weakly and the strongly stratified turbulence regimes based on information available in a full Reynolds stress ***,we generate a direct numerical simulation database with Reynolds numbers from 10,000 to 50,000 and froude numbers from 2 to ***(100)independent realizations of temporally evolving wakes are computed to get converged ***,we train a linear logistic regression classifier with weight thresholding for automated flow regime *** classifier is designed to identify the physics critical to *** against data at one flow condition,the classifier is found to generalize well to other Reynolds and froude *** results show that the physics governing wake evolution is universal,and that the classifier captures that physics.

Twinning in intermetallic Compounds Are Long Shear Vectors and/or Shuffles Really Necessary?

in this paper the geometric description and general theory of mechanical twinning are reviewed, the twins in general lattices and superlattices are summarized, and the kinetic process by which mechanical twins form is revisited. A case study of mechanical twinning of HfV2+Nb, (cubic) Laves phase, is presented and the synchroshear of selected atomic layers is proposed to explain the physical process of twin formation. if the twins form in this way, then long shear vectors and / or atomicshuffles are not really necessary.

This work demonstrated the viability of friction stir welding for the welding of medium-Mn steels when used as cryogenic vessel materials for liquefied gas *** used an intercritically annealed fe-7 Mn-0.2 C-3 Al(wt.%)steel with a dual-phase(α'martensite andγ_(R) retained austenite)nanolaminate structure as a base material and systematically compared its microstructure and impact toughness after friction stir and tungsten inert gas *** friction stir welded specimen exhibited a large amount ofγ_(R) phase owing to a relatively low temperature during welding,whereas the tungsten inert gas welded specimen comprised only theα'***,the friction stir welded steel exhibited a tuned morphology of nanoscale globular microstructure at the weld zone and did not exhibit any prior austenite grain boundary due to active recrystallization caused by deformation during *** preserved fraction ofγ_(R) and morphological tuning in the weldment improved the impact toughness of the friction stir welded steel at low *** the steel processed by tungsten inert gas welding,the notch crack propagated rapidly along the prior austenite grain boundaries-weakened by Mn and P segregations-resulting in poor impact ***,the friction stir welded steel exhibited a higher resistance against notch crack propagation due to the slow crack propagation along the ultrafine ferrite/ferrite(α/α)interfaces,damage tolerance by the active transformation-induced plasticity from the large amount ofγR,and enhanced boundary cohesion by suppressed Mn and P segregations.

Xylariaceous endophytes are commonly encountered in plants,and it has recently been found out that insect vectors play a major role in their life cycle,as well as in the apparent host-specificity of the fungal *** this study,a new insect-associated,endophytic anamorph species of Daldinia is recognised from a comparison of (a)morphological and ecological data;(b)molecular phylogenetic data based on rDNA,alpha-actin and beta-tubulin genes;and (c)secondary metabolite profiles based on high performance liquid chromatography/diode array/mass spectrometry(HPLC-DAD/MS)and gas chromatography/mass spectrometry(GC-MS)in comparison with authentic strains of Daldinia and other *** new species,Daldinia hawksworthii,is a symbiont of the willow woodwasp,Xiphydria prolongata,which is in turn preferentially associated with Salix.A comparison of HPLCDAD/MS profiles from standardised cultures revealed the presence of a specific compound,which was not encountered in any of the ca.250 Daldinia strains studied for *** compound was isolated after fermentation of the fungus in 10 l scale by preparative *** elucidation by NMR spectroscopy revealed a novel pyrone,for which the name dalsymbiopyrone is *** compound showed weak antimicrobial and cytotoxic effects and might be a signal molecule involved in the trilateral ecological interaction between the host plant,the insect,and its fungal symbiont.

We report on the microstructure and the strengthening mechanisms of additively manufactured parts fabricated by directed energy deposition of Ti-6Al-4V(Ti64)powders blended with yttria-stabilized zirconia(YSZ)*** specimens showed refined microstructures as compared to bare as-deposited Ti64,where theαand columnar priorβgrain sizes decreased with increasing YSZ *** YSZ nanoparticles decomposed during the deposition process and led to the formation of yttrium oxide and some excess oxygen in the Ti64 *** decrease in the sizes of the priorβgrains could be attributed to the increasing amount of dissolved oxygen and yttrium,which promoted constitutional ***,the reduction in the size of theαgrains could be ascribed to a shift of the onset of theβ→α+βtransformation to a higher temperature and shorter time with increasing concentration of dissolved ***,the contributions of the underlying strengthening mechanisms for the as-deposited specimens were quantitatively determined.
