Flavin monooxygenases （FMOs） play critical roles in plant growth and development by synthesizing auxin and other signaling molecules. However, the structure and function relationship within plant FMOs is not understood. Here we defined the important residues and domains of the Arabidopsis YUC1 FMO, a key enzyme in auxin biosynthesis. We previously showed that simultaneous inactivation of YUC1 and its homologue YUC4 caused severe defects in vascular and floral development. We mutagenized the yuc4 mutant and screened for mutants with phenotypes similar to those of yucl yuc4 double mutants. Among the isolated mutants, five of them contained mutations in the YUC1 gene. Interestingly, the mutations identified in the new yucl alleles were concentrated in the two GXGXXG motifs that are highly conserved among the plant FMOs. One such motif presumably binds to flavin adenine dinucleotide （FAD） cofactor and the other binds to nicotinamide adenine dinucleotide phosphate （NADPH）. We also identified the Ser139 to Phe conversion in yucl, a mutation that is located between the two nucleotide-binding sites. By analyzing a series of yucl mutants, we identified key residues and motifs essential for the functions of YUC1 FMO.

Sabellaria alveolata(Linnaeus 1767)is a polychaete able to build bioconstructions of different thickness,size and patchiness,in intertidal and subtidal *** biological features have been the object of numerous studies *** worm reefs are formed by millions of tubes built by sand and shells(whole or in fragments)bonded together with a strong glue produced by the worm ***,Sabellaria alveolata represents a sedimentological asset for the coastal protection,since it contributes to create natural barriers against storm waves and erosion,and supplies the beach with new sandy *** work shows a multidisciplinary approach to studying a bioconstruction of Sabellaria alveolata along the latium coast(Ostia,Tyrrhenian Sea,central Italy),proposing image analysis as a novel technique to investigate worm reefs,along with classical sedimentological/ecological *** Sabellaria bioconstructions have been analysed at different scales of observation,suggesting the more appropriate strategies to develop a reliable model illustrating the different growth steps of these bioconstructions.

In this paper we analysed the historical and instrumental seismicity of the seismic district “Penisola Salentina” (Salento peninsula) in the southern part of the Apuliaregion, making use of the most recent seismological database. Relocation of available dataset points out that the events are spatially distributed over a belt of deformation that approximately corresponds to Soglia Messapica (Taranto-Brindisi depression). Besides, computed source characteristics indicate dextral strike-slip solutions with an approximately E-W orientation that seismologically confirm previous geodynamic studies indicating a NE-SW extension in the Taranto-Brindisi depression. In particular, the tensional stress associated to the present seismic activity could be the consequence of the relaxation of the buckling process following the extensional re-arrangement of the Apenninic belt masses. Moreover attenuation QPvalues obtained in this study are much greater than those inferred in other parts of Italian peninsula;this result agrees with previous macroseismic investigations and indicates a greater efficiency of the studied area in the transmission of body waves.

The incidence of obesity and its related conditions, including non-alcoholic fatty liver disease(NAFLD), has dramatically increased in all age groups worldwide. Given the health consequences of these conditions, and the subsequent economic burden on healthcare systems, their prevention and treatment have become major priorities. Because standard dietary and lifestyle changes and pathogenically-oriented therapies(e.g., antioxidants, oral hypoglycemic agents, and lipid-lowering agents) often fail due to poor compliance and/or lack of efficacy, novel approaches directed toward other pathomechanisms are needed. Here we present several lines of evidence indicating that, by increasing energy extraction in some dysbiosis conditions or small intestinal bacterial overgrowth,specific gut microbiota and/or a"low bacterial richness"may play a role in obesity,metabolic syndrome,and fatty *** conditions involving a damaged intestinal barrier("leaky gut"),the gut-liver axis may enhance the natural interactions between intestinal bacteria/bacterial products and hepatic receptors(e.g.,toll-like receptors),thus promoting the following cascade of events:oxidative stress,insulinresistance,hepatic inflammation,and *** also discuss the possible modulation of gut microbiota by probiotics,as attempted in NAFLD animal model studies and in several pilot pediatric and adult human ***,this approach appears to be a promising and innovative add-on therapeutic tool for NAFLD in the context of multi-target therapy.

Seedlessness represents a highly appreciated trait in table *** on an interesting case of seedless fruit production described in the crop species Annona squamosa,we focused on the Vitis vinifera INNER NO OUTER(INO)gene as a *** gene encodes a transcription factor belonging to the YABBY family involved in the determination of abaxial identity in several *** Arabidopsis thaliana,this gene was shown to be essential for the formation and asymmetric growth of the ovule outer integument and its mutation leads to a phenotypic defect of ovules and failure in seed *** this study,we identified in silico the *** orthologue and investigated its phylogenetic relationship to INO genes from other species and its expression in different organs in seeded and seedless *** cross-species complementation,we have tested its functionality in the Arabidopsis ino-1 *** show that the *** INO successfully rescues the ovule outer integument growth and seeds set and also partially complements the outer integument asymmetric growth in the Arabidopsis mutant,differently from orthologues from other *** data demonstrate that VviINO retains similar activity and protein targets in grapevine as in *** implications for grapevine breeding are discussed.

AIM: To check the safety and efficacy of boceprevir/telaprevir with peginterferon/ribavirin for hepatitis C virus(HCV) genotype 1 in the real-world settings. METHODS: This study was a non-randomized, observational, prospective, multicenter. This study involved 47 centers in Italy. A database was prepared for the homogenous collection of the data, was used by all of the centers for data collection, and was updated continuously. All of the patients enrolled in this study were older than 18 years of age and were diagnosed with chronic infection due to HCV genotype 1. The HCV RNA testing was performed using COBAS-Taq Man2.0(Roche, LLQ 25 IU/m L). RESULTS: All consecutively treated patients were included. Forty-seven centers enrolled 834 patients as follows: Male 64%; median age 57(range 18-78), of whom 18.3% were over 65; mean body mass index 25.6(range 16-39); genotype 1b(79.4%); diagnosis of cirrhosis(38.2%); and fibrosis F3/4(71.2%). The following drugs were used: Telaprevir(66.2%) and PEG-IFN-alpha2a(67.6%). patients were na?ve(24.4%), relapsers(30.5%), partial responders(14.8%) and null responders(30.3%). Overall, adverse events(AEs) occurred in 617 patients(73.9%) during the treatment. Anemia was the most frequent AE(52.9% of cases), especially in cirrhotic. The therapy was stopped for 14.6% of the patients because of adverse events or virological failure(15%). Sustained virological response was achieved in 62.7% of the cases, but was 43.8% in cirrhotic patients over 65 years of age. CONCLUSION: In everyday practice, triple therapy is safe but has moderate efficacy, especially for patients over 65 years of age, with advanced fibrosis, nonresponders to peginterferon + ribavirin.
