Systematic Characterization of Long Non-Coding RNAs and Their Responses to Drought Stress in Dongxiang Wild Rice

Long non-coding RNAs(lncRNAs) play important roles in response to various biotic and abiotic stresses. So far, systematic identification and characterization of lncRNAs have been reported in a few model plant species and major crops, but their roles in abiotic stress response have not yet been reported in common wild rice(Oryza rufipogon). Dongxiang wild rice(DXWR) possesses a high degree of drought resistance and has been well recognized as a precious genetic resource for drought resistant rice breeding. We presented the reference catalog of 1 655 novel lncRNA transcripts in DXWR using strand-specific RNA sequencing and bioinformatics approaches. Meanwhile, a total of 1092 lncRNAs were determined as differentially expressed lncRNAs under drought stress. Quantitative real-time PCR results exhibited a high concordance with RNA sequencing data, which confirmed that the expression patterns of lncRNAs based on RNA sequencing were highly reliable. Furthermore, 8 711 transcripts were predicted as target genes of the differentially expressed lncRNAs. Functional annotation analysis based on Gene Ontology and Kyoto Encyclopedia of Genes and Genomes databases showed that the target genes were significantly enriched in cellular and metabolic processes, cell part, binding and plant hormone signal transduction, as well as many other terms related to abiotic stress resistance. These results expanded our understanding of lncRNA biology and provided candidate regulators for genetic improvement of drought resistance in rice cultivars.

Species-level monitoring of rare and invasive fishes using eDNA metabarcoding in the middle and upper Yarlung Zangbo River, Tibet

Fish diversity of the Yarlung Zangbo River is very sensitive and vulnerable to biological invasion,anthropogenic activities and climate change,especially in the upper and middle reaches where several endemic fishes have become endangered and nearly ten invasive fishes have been ***,we used environmental DNA(eDNA)metabarcoding to monitor rare and invasive fishes,and to assess diversity in 25 sites from two wetlands(Lalu and Chabalang)and the main channel(YT),within the upper and middle *** obtain a species-level resolution,we evaluated species discrimination potentials of three mitochondrial markers and found Cytb had the highest average genetic distance at each taxonomic level followed by COI and *** 12S was unqualified for species assignment,as two species shared identical *** newly designed Cytb primers used for metabarcoding showed an average mismatch of 0.28 and amplified well across *** total,8942 operational taxonomic units(OTUs)were obtained based on a 100%identity threshold,among which 98.1%were assigned to 24 fishes based on our custom-made database and the remaining were assigned to six fishes based on the NCBI nt *** all captured fishes were detected by the eDNA method except for two ***,12 fishes detected by the eDNA method were not listed in catch data for several sites,including one endangered species(Oxygymnocypris stewartii),four previously recorded non-native species and two unrecorded non-native species(Monopterus albus and Siniperca chuatsi).The alpha diversities estimated by eDNA and capture-based methods were correlated for sites at *** methods revealed significant differences in community composition between YT and the *** results provide both basic information for conservation and management of rare and invasive fishes in the Yarlung Zangbo River and a framework of fish eDNA metabarcoding with a species-level resolution.

Experimental research on acoustic wave velocity of frozen soils during the uniaxial loading process

Ultrasonic P-wave tests of frozen silt and frozen sand were conducted during uniaxial loading by using an RSM^-SY5（T） nonmetal ultrasonic test meter to study the velocity characteristics of P-waves. The experimental results indicate that the P-wave velocity is affected by soil materials, temperature, and external loads, so the P-wave velocity is different in frozen silt and frozen sand, but all decrease with an increase of temperature and increase at first and then decrease with strain during the loading process. There is an exponential relationship between uniaxial compressive strength and P-wave ve- locity, and the correlation between them is very good. The characteristic parameters of acoustic waves can, to some extent, reflect the development of internal cracks in frozen soils during loading.

Expression and role of autophagy related protein p62 and LC3 in the retina in a rat model of acute ocular hypertension

AIM: To investigate the expression and possible role of the autophagy related protein p62 and LC3 in the retina based on a rat model of acute ocular ***: Fifty rats were randomized into five groups: control group A, B, C, and D. Groups A to D all received normal saline perfusion into the anterior chamber with pressure of 80 mm Hg for one hour, and retina tissue was obtained at 6, 12, 24 and 48 h after perfusion respectively, to investigate the activation of autophagy following ischemiareperfusion. The distribution and semi-quantification of autophagy related protein p62 and LC3 in the retina were detected using immunohistochemistry technique. The expression level of these two proteins was evaluated using Western ***: The number of retinal ganglion cells(RGCs) decreased with increasing reperfusion time, and significant reduction in the retinal thickness was observed 48 h after perfusion. In normal adult rats, LC3 protein was mainly expressed in the ganglion cell layer(GCL), and p62 protein was expressed in the nerve fiber layer(NFL), GCL, inner plexiform layer(IPL), inner nuclear layer(INL) and outer plexiform layer(OPL). In comparison to the control group, the expression level of LC3-II was higher in all the experimental groups(P<0.05), with the peak expression at 12 h after reperfusion. Additionally, the expression level of p62 was higher in all the experimental groups than the control(P<0.05, except for group A), with the peak level occurred 24 h after reperfusion. CONCLUSION: Both p62 and LC3 show low level and uneven expression in the retina of normal adult rats. Acute ocular hypertension can lead to upregulation of LC3-II and p62 expression in the retina. Autophagy flux is damaged 12 h after reperfusion, potentially resulting in further loss of RGCs.

Microstructure and finite element analysis of hot continuous rolling of doped tungsten rod

The microstructures of doped tungsten deformed by multi-pass hot continuous rolling were investigated, and the stress and strain fields were simulated by finite element(FE) method. After the continuous rolling, the grains of the tungsten rod were refined, and the microhardness was improved; however, a ring region of abnormal grain growth was present at a distance of about 3/5 R(R is the radius of the rod) from the center of the cross section. FE modeling results showed that the equivalent residual strains were minimum around the region of abnormal grain growth; this was due to the release of strain energy by severe plastic deformation, leading a situation where the migration force of grain boundaries was higher than the pinning force of potassium bubbles. By decreasing the initial rolling temperature and rolling speeds, the inhomogeneity of the equivalent residual stain decreased, improving the microstructure uniformity of the doped tungsten.

Implementing an“Impracticable”Copolymerization to Fabricate a Desired Polymer Precursor for N-doped Porous Carbons

It is common that a proof-of-concept of a desired reaction,which might generate materials with new functions or application potential,is eventually proved impracticable or commercially *** efforts have been made but wasted in searching for unknown reaction conditions in solvent environments because it was believed that the activity of reactants can be enhanced to facilitate reactions by dissolving them in ***,an abnormal case was discovered in this study.A desired copolymerization reaction between 1,3,5-tris(chloromethyl)-2,4,6-trimethylbenzene and melamine was confirmed to be impracticable under various solvent conditions;however,it was successfully implemented using a solvent-free *** first-principle calculations and molecular dynamics simulations,two decisive factors that the reaction in solvents cannot possess,namely the reaction equilibrium being pushed by the timely release of by-products and the confined thermal motions of the activated monomer molecules in the solid phase,were demonstrated to make the copolymerization successful in the solvent-free *** to the high aromaticity and azacyclo-content,the as-synthetic copolymer exhibited good application potential as a precursor to fabricate N-doped porous carbons with satisfactory carbon yields,ideal N contents,desired textural properties,and competitive CO_(2)capture abilities compared to other representative counterparts reported recently.
