The difficulty of converting scientific research findings into novel pharmacological treatments for rare and life-threatening diseases is *** related to multiple biological processes involved in cell growth,proliferation,and disease occurrence have been identified in recent years with the development of immunology,molecular biology,and genomics *** are capable of reflecting normal physiological processes,pathological processes,and the response to therapeutic intervention;as such,they play vital roles in disease diagnosis,prevention,drug response,and other aspects of *** discovery of valuable biomarkers has become a focal point of current *** studies have identified molecular biomarkers based on the differential expression/concentration of molecules(e.g.,genes/proteins)for disease state diagnosis,characterization,and *** technological breakthroughs in molecular analysis platforms have enabled the identification of a large number of candidate biomarkers for rare diseases,only a small number of these candidates have been properly validated for use in patient *** traditional molecular biomarkers may lose vital information by ignoring molecular associations/interactions,and thus the concept of network biomarkers based on differential associations/correlations of molecule pairs has been *** approach promises to be more stable and reliable in diagnosing disease ***,the newly-emerged dynamic network biomarkers(DNBs)based on differential fluctuations/correlations of molecular groups are able to recognize pre-disease states or critical states instead of disease states,thereby achieving rare disease prediction or predictive/preventative medicine and providing deep insight into the dynamic characteristics of disease initiation and progression.

Primary biliary cholangitis(PBC) is an autoimmune disease involving dysregulation of a broad array of homeostatic and metabolic processes. Although considerable single-nucleotide polymorphisms have been unveiled, a large fraction of risk factors remains enigmatic. Candidate genes with rare mutations that tend to confer more deleterious effects need to be identified. To help pinpoint cellular and developmental mechanisms beyond common noncoding variants, we integrate whole exome sequencing with integrative network analysis to investigate genes harboring de novo mutations. Prominent convergence has been revealed on a network of disease-specific co-expression comprised of 55 genes associated with homeostasis and metabolism. The transcription factor gene MEF2 D and the DNA repair gene PARP2 are highlighted as hub genes and identified to be up-and down-regulated, respectively, in peripheral blood data set. Enrichment analysis demonstrates that altered expression of MEF2 D and PARP2 may trigger a series of molecular and cellular processes with pivotal roles in PBC pathophysiology. Our study identifies genes with de novo mutations in PBC and suggests that a subset of genes in homeostasis and metabolism tend to act in synergy through converging on co-expression network, providing novel insights into the etiology of PBC and expanding the pool of molecular candidates for discovering clinically actionable biomarkers.

In the centenary year of Einstein's General Theory of Relativity, this paper reviews the current status of gravitational wave astronomy across a spectrum which stretches from attohertz to kilohertz frequencies. Sect. 1 of this paper reviews the historical development of gravitational wave astronomy from Einstein's first prediction to our current understanding the spectrum. It is shown that detection of signals in the audio frequency spectrum can be expected very soon, and that a north-south pair of next generation detectors would provide large scientific benefits. Sect. 2 reviews the theory of gravitational waves and the principles of detection using laser interferometry. The state of the art Advanced LIGO detectors are then described. These detectors have a high chance of detecting the first events in the near future. Sect. 3 reviews the KAGRA detector currently under development in Japan,which will be the first laser interferometer detector to use cryogenic test masses. Sect. 4 of this paper reviews gravitational wave detection in the nanohertz frequency band using the technique of pulsar timing. Sect. 5 reviews the status of gravitational wave detection in the attohertz frequency band, detectable in the polarisation of the cosmic microwave background, and discusses the prospects for detection of primordial waves from the big bang. The techniques described in sects. 1–5 have already placed significant limits on the strength of gravitational wave sources. Sects. 6 and 7 review ambitious plans for future space based gravitational wave detectors in the millihertz frequency band. Sect. 6 presents a roadmap for development of space based gravitational wave detectors by china while sect. 7 discusses a key enabling technology for space interferometry known as time delay interferometry.

The 12C+12C fusion reaction is famous because of its complication of molecular resonances,and it plays an important role in both nuclear structural research and astrophysics. It is extremely difficult to measure the cross sections of 12C+12C fusions at energies of astrophysical relevance because of the very low reaction yields. To measure the complicated resonant structure that exists in this important reaction, an efficient thick target method has been developed and applied for the first time at energies Ec.m.<5.3 MeV. A scan of the cross sections over a relatively wide range of energies can be carried out using only a single beam energy. The result of measurement at Ec.m.=4.1 MeV is compared with results from previous *** method will be useful for searching for potentially existing resonances of 12C+12C in the energy range 1 MeV

正在建设中的强流重离子加速器装置HIAF配备了一条先进的放射性束流线HFRS,建成后将为中国高能放射性核束物理研究提供新的机遇。HFRS的特点是流强极高(初级束流强度1×10^(11) ppp),这对用于粒子鉴别的能损探测器提出了非常高的计数率要求。传统的能损探测器对信号处理一般依次采用电荷灵敏前放、主放大器、ADC的技术路线。该方案存在电子学响应速度较慢、灵活性较差、难以处理高计数率下信号堆积严重等问题。我们对此提出一种新的适用于高计数率的能损探测器方案:以耐辐照的多次取样电离室为能损探测器,优化结构和读出方式提高探测器响应速度,使用快电荷灵敏前放对能损探测器信号初步放大后、直接用波形数字化仪采集波形再进行后续数字算法处理。并利用放射源和束流对该方案进行了验证测试。利用3组分α源测试时,对采集的波形使用数字成形算法处理,能量分辨率(FWHM)可达1.31%。在放射性次级束流线RIBLL2提供300 MeV/u的~(56)Fe束流测试中,采用时间常数τ_(f)=2μs的快电荷灵敏前放在计数率接近1 MHz时仍未发生明显堆积。

HIV-1 Nef protein plays an important role in HIV-1 pathogenesis, including downregulation of the cell surface CD4 and class I major histocompatibility complex （MHC）, enhancement of virion infectivity and alteration of a variety of cellular signal transduction pathways.1.2 A recent report demonstrated that Nef down-regulates dendritic cell surface expression of MHC class I and up-regulates molecules known to be critical for apoptosis induction, such as TNF-α and FasL^3 It is also very interesting that Nef expression alone may be sufficient to cause an AIDS-like disease in transgenic mice.2 In many human cohort studies it has been reported that long-term nonprogressors （LTNPs）, who remain asymptomatic for more than one decade without antiretroviral therapy, harbored nef-deleted viruses.4-7 Intense studies on nef genes from LTNPs have been done in Europe and United States but none of such studies have been done in china.

Foxtail millet(Setaria italica),which was domesticatedfromthewild speciesgreenfoxtail(Setaria viridis),isa richsource of phytonutrientsfor *** evaluate how breeding changed themetabolome offoxtail millet grains,we generated and analyzed the datasets encompassing the genomes,transcriptomes,metabolomes,and anti-inflammatory indices from 398 foxtail millet *** identified hundreds of common variants that influence numerous secondary *** observed tremendous differences in natural variations of the metabolites and their underlying genetic architectures between distinct sub-groups of foxtail ***,we found that the selection of the gene alleles associated with yellow grains led to altered profiles of metabolites such as carotenoids and endogenous *** CRiSPR-mediated genome editing wevalidated the function of PHYTOENE SYNTHASE1(PSY1)gene in affecting milletgrain colorand ***,our in vitro cell inflammation assays showed that 83 metabolites in millet grains have anti-inflammatory *** together,ourmulti-omics study illustrates how the breeding history of foxtail millet has shaped its metabolite *** datasets we generated in this study also provide important resources for further understanding how millet grain quality is affected by different metabolites,laying the foundations for future millet genetic research and metabolome-assisted improvement.
