The limited molecular classifications and disease signatures of osteoarthritis(OA)impede the development of prediagnosis and targeted therapeutics for OA *** classify and understand the subtypes of OA,we collected three types of tissue including cartilage,subchondral bone,and synovium from multiple clinical centers and constructed an extensive transcriptome atlas of OA *** applying unsupervised clustering analysis to the cartilage transcriptome,OA patients were classified into four subtypes with distinct molecular signatures:a glycosaminoglycan metabolic disorder subtype(C1),a collagen metabolic disorder subtype(C2),an activated sensory neuron subtype(C3),and an inflammation subtype(C4).Through ligand-receptor crosstalk analysis of the three knee tissue types,we linked molecular functions with the clinical symptoms of different OA *** example,the Gene Ontology functional term of vasculature development was enriched in the subchondral bone-cartilage crosstalk of C2 and the cartilage-subchondral bone crosstalk of C4,which might lead to severe osteophytes in C2 patients and apparent joint space narrowing in C4 *** on the marker genes of the four OA subtypes identified in this study,we modeled OA subtypes with two independent published RNA-seq datasets through random forest *** findings of this work contradicted traditional OA diagnosis by medical imaging and revealed distinct molecular subtypes in knee OA patients,which may allow for precise diagnosis and treatment of OA.

In recent years, tungsten disulfide(WS_(2)) and tungsten selenide(WSe_(2)) have emerged as favorable electrode materials because of their high theoretical capacity, large interlayer spacing, and high chemical activity;nevertheless, they have relatively low electronic conductivity and undergo large volume expansion during cycling, which greatly hinder them in practical applications. These drawbacks are addressed by combining a superior type of carbon material, graphene, with WS_(2) and WSe_(2) to form a WS_(2)/WSe_(2)@graphene *** materials have received considerable attention in electro-chemical energy storage applications such as lithium-ion batteries(LIBs), sodium-ion batteries(SIBs),and supercapacitors. Considering the rapidly growing research enthusiasm on this topic over the past several years, here the recent progress of WS_(2)/WSe_(2)@graphene nanocomposites in electrochemical energy storage applications is summarized. Furthermore, various methods for the synthesis of WS_(2)/WSe_(2)@graphene nanocomposites are reported and the relationships among these methods, nano/microstructures, and electrochemical performance are systematically summarized and discussed. In addition, the challenges and prospects for the future study and application of WS_(2)/WSe_(2)@graphene nanocomposites in electrochemical energy storage applications are proposed.

Terbium- and yttrium-group rare earths form β-complexes with CPApC in acidic medium. The sensitivities for determination of these rare earths by this reaction depend on their ionic radii. Cerium- or terbium-group rare earth in the presence of yttrium-group element produces the cocolouration effect which remarkably sensitizes the reaction. Yb-CPApC-Eu complex has a molar composition of 1:4:2 and gives a molar absorptivity of 2. 02 ×10~5 L·mol^(-1)·cm^(-1) at 746 nm for Eu. It is found that the closer the lengths of ionic radii of the rare earths are, the greater will be the cocolouration effect.

Alcoholic steatohepatitis (ASH) is a liver disease characterized by steatosis, inflammation, and necrosis of the liver tissue as a result of excessive alcohol consumption. Pregnane X receptor (PXR) is a xenobiotic nuclear receptor best known for its function in the transcriptional regulation of drug metabolism and disposition. Clinical reports suggested that the antibiotic rifampicin, a potent human PXR activator, is a contraindication in alcoholics, but the mechanism was unclear. In this study, we showed that the hepatic expression of fatty acid binding protein 4 (FABP4) was uniquely elevated in ASH patients and a mouse model of ASH. Pharmacological inhibiting FABP4 attenuated ASH in mice. Furthermore, treatment of mice with the mouse PXR agonist pregnenolon-16α-carbonitrile (PCN) induced the hepatic and circulating levels of FABP4 and exacerbated ASH in a PXR-dependent manner. Our mechanism study established FABP4 as a transcriptional target of PXR. Treatment with andrographolide, a natural compound and dual inhibitor of PXR and FABP4, alleviated mice from ASH. In summary, our results showed that the PXR-FABP4 gene regulatory axis plays an important role in the progression of ASH, which may have accounted for the contraindication of rifampicin in patients of alcoholic liver disease. Pharmacological inhibition of PXR and/or FABP4 may have its promise in the clinical management of ASH.

Plant carbon(C)concentration is a fundamental trait for estimating C storage and nutrient ***,the mechanisms of C concentration variations among different tree tissues and across species remains poorly *** this study,we explored the variations and determinants of C concentration of nine tissues from 216 individuals of 32 tree species,with particular attention on the effect of wood porosity(i.e.,non-porous wood,diffuse-porous wood,and ring-porous wood).The inter-tissue pattern of C concentration diverged across the three porosity types;metabolically active tissues(foliage and fine roots,except for the foliage of ring-porous species)generally had higher C levels compared with inactive *** poor inter-correlations between tissue C concentrations indicated a necessity of measuring tissue-and specific-C *** concentration for almost all tissues generally decreased from non-porous,to diffuse-porous and to *** C was often positively correlated with tissue(foliage and wood)density and tree size,while negatively correlated with growth rate,depending on wood *** results highlight the mediating effect of type of wood porosity on the variation in tissue C among temperate *** variations among tissues were more important than that among *** findings provided insights on tissue C concentration variability of temperate forest species.

Increasing evidence supports the hypothesis of autologous immune attack in severe aplastic anemia(SAA):the predominant role of activated cytotoxic T cells(CTL)expressing-interferon in inhibiting the growth of bone marrow(BM)cells,putative autoantigens,and oligoclonal expansion of CD8+T cells.1 For SAA patients,the definitive therapies are immunosuppressive therapy(IST)or hematopoietic stem transplantation(HSCT).

The purified terephthalic acid （PTA） petrochemical wastewater molecular toxicity detected by use of Mouse Genome 430A 2.0 GeneChip was conducted in this research. The toxic dose to male mice was 0.03 g/（kg, d） of PTA in the wastewater. The mice liver total RNA was isolated as the temple for synthesis of eDNA and then the cDNA as the temple for synthesis of cRNA. Hybridizing the cRNA with the target genes on the gene chip, there were 232 genes expression levels up-regulated and 74 genes down-regulated discovered obviously. The foremost 40 genes for both the highest and the lowest expression levels involved endogenetic steroid and hormone metabolism, immune system, the leukocyte activity and inflammation, detoxification in liver, reproduction and growth hormone, regulation immune factors of anti-tumor and anti-infection and cancer to the mice sampled. The data suggest the PTA wastewater contained over 5 aromatics and their toxicities integrated were much higher than the pure chemical PTA. And the pure chemical PTA toxicities data cannot be used to evaluate the toxicity of the PTA wastewater instead.

Fluoropyrimidine-based combination chemotherapy plus targeted therapy is the standard initial treatment for unresectable metastatic colorectal cancer(mCRC),but the prognosis remains *** phase 3 trial(***:NCT03950154)assessed the efficacy and adverse events(AEs)of the combination of PD-1 blockade-activated DC-CIK(PD1-T)cells with XELOX plus bevacizumab as a first-line therapy in patients with mCRC.A total of 202 participants were enrolled and randomly assigned in a 1:1 ratio to receive either first-line XELOX plus bevacizumab(the control group,n=102)or the same regimen plus autologous PD1-T cell immunotherapy(the immunotherapy group,n=100)every 21 days for up to 6 cycles,followed by maintenance treatment with capecitabine and *** main endpoint of the trial was progression-free survival(PFS).The median follow-up was 19.5 *** PFS was 14.8 months(95%CI,11.6-18.0)for the immunotherapy group compared with 9.9 months(8.0-11.8)for the control group(hazard ratio[HR],0.60[95%CI,0.40-0.88];p=0.009).Median overall survival(OS)was not reached for the immunotherapy group and 25.6 months(95%CI,18.3-32.8)for the control group(HR,0.57[95%CI,0.33-0.98];p=0.043).Grade 3 or higher AEs occurred in 20.0%of patients in the immunotherapy group and 23.5%in the control groups,with no toxicity-associated deaths *** addition of PD1-T cells to first-line XELOX plus bevacizumab demonstrates significant clinical improvement of PFS and OS with well tolerability in patients with previously untreated mCRC.

The stereospecific carbocupration of terminal alkynes via higher order cuprates to give 1,1'-disubstituted olefins with 99% configuration purity is described. Its synthetic utility as a general method for the preparation of substituted olefins is further illustrated by the direct synthesis of (±)-ipsenol and (E)-β-farnesene with highly geometric purity of the carbon-carbon double bond.
