Validated Gradient Stability Indicating UpLC Method for the Determination of Related Substances of posaconazole in Bulk Drug

A stability-indicating UpLC method has been developed and validated for the determination of related substances of posaconazole with its four related substances (Hydroxytriazole, Tosylated compound, Deshydroxy posaconazole and Benzylated posaconazole) in the drug substance. Forthwith simple UpLC chromatographic separations were achieved on a Waters Acquity BEH shield C18 (100 mm length, 2.1 mm internal diameter and 1.7 μm particle size) with a mobile phase containing 0.1% Orthophosphoric acid (i.e. 1 mL in 1000 mL water) in gradient combination with acetonitrile (ACN) at a flow rate of 0.5 mL/min and the eluent were monitored at 210 nm. As a result, the resolution of posaconazole from any of impurities was found to be greater than 2.0. The test solution and spiked solutions were found to be stable in the diluent for 48 h. For the purpose method to be stability indicating, forced degradation studies were conducted and the method resolved the drug from its known impurities, stated above, and from additional impurities generated when pOS subjected to forced degradation;the mass balance was found close to 100%. Regression analyses indicate correlation coefficient value greater than 0.999 for posaconazole and its known impurities. The LOD for posaconazole and the known impurities was at a level below 0.05%. The method has shown good, consistent recoveries for known impurities (89% - 106%). To summarise, the method was found to be accurate, precise, linear, specific, sensitive, rugged, robust, and stability-indicating.

Enhanced Energy Efficient with a Trust Aware in MANET for Real-Time Applications

Mobile ad hoc networks (MANETs) are subjected to attack detectionfor transmitting and creating new messages or existing message *** attacker on another node evaluates the forging activity in themessage directly or indirectly. Every node sends short packets in a MANETenvironment with its identifier, location on the map, and time through *** attackers on the network broadcast the warning message usingfaked coordinates, providing the appearance of a network collision. Similarly,MANET degrades the channel utilization performance. performancehighly affects network performance through security algorithms. This paperdeveloped a trust management technique called Enhanced Beacon TrustManagement with Hybrid Optimization (EBTM-Hyopt) for efficient clusterhead selection and malicious node detection. It tries to build trust amongconnected nodes and may improve security by requiring every participatingnode to develop and distribute genuine, accurate, and trustworthy materialacross the network. Specifically, optimized cluster head election is done periodicallyto reduce and balance the energy consumption to improve the lifetimenetwork. The cluster head election optimization is based on hybridizingparticle Swarm Optimization (pSO) and Gravitational Search OptimizationAlgorithm (GSOA) concepts to enable and ensure reliable routing. Simulationresults show that the proposed EBTM-HYOpT outperforms the state-of-thearttrust model in terms of 297.99 kbps of throughput, 46.34% of pDR, 13%of energy consumption, 165.6 kbps of packet loss, 67.49% of end-to-end delay,and 16.34% of packet length.

Floral extract of Tecoma stans:A potent inhibitor of gentamicin-induced nephrotoxicity in vivo

Objective:To highlight the nephroprotective activity of ethyl acetate extract of dried flowers of Tecomu stans for its protective effects on genlamicin-induced nephrotoxicity in albino rats. Methods:For studying acute toxicity study,single oral dose of 5（KM） mg ethyl acetate floral extract/kg hodv weight was administered to albino rats（five females,five males）.Nephrotoxicity was induced in albino rats by intraperitoneal administration of gentamicin 80 mg/kg/day for eight *** of concurrent administration of ethyl acetate floral extract of Tecoma stans at a dose of 100.200 and 300 mg/kg/day given by oral mute was determined using serum creatinine,serum uric acid,blood urea nitrogen and serum urea as indicators of kidney *** study groups contained six rats in each *** nephrotoxicity of gentamicin is known to involve induction of oxidative stress,in vitro antioxidant aclivity and free radical-scavenging activity of this extract was also ***:For acute toxicity testing both female and male rats administered with the extract at a dose of 5 000 mg/*** results showed no toxicity in terms of general behavior change,mortality,or change in gross appearance of internal organs(LD50>5 000 mg/kg). Rice sheath blight disease （ShB）, caused by Rhizoctonia solani, gives rise to significant grain yield losses. The present study evaluated the efficacy of Integral, the commercial liquid formulation of Bacillus subtilis strain MBI 600, against rice ShB and for plant growth promotion. In greenhouse studies, four log concentrations of Integral （from 2.2×10^6 to 2.2×10^9 cfu/mL） were used as seed treatment （ST）. After 25 d, seedlings were dipped （SD） into Integral prior to transplanting. At 30 d after transplanting （DAT）, leaf sheaths were inoculated with immature sclerotia of the pathogen. At 45 DAT, a foliar spray （FS） with Integral was applied to some treatments. The fungicide control was 50% carbendazim at 1.0 g/L, and a nontreated control was also included. Overall, there were 10 treatments, each with five replications. ShB severity was rated at 52 DAT, and seedling height and number of tillers per plant were rated at 60 DAT. In 2009, two field trials evaluated Integral at 2.2×10^8 and 2.2×10^9 cfu/mL. Integral was applied as ST, and seedlings were produced in a nursery bed. After 32 d, seedlings were treated with Integral as SD and transplanted into 10 m^2 blocks. Foliar sprays were given at 45 and 60 DAT. There were seven treatments, each with eight replications arranged as a factorial randomized complete block design. At 20 DAT, the plots were broadcast inoculated with R. solani produced on rice grains. Seedling height before transplanting, ShB severity at 90 DAT, and grain yield at harvest were recorded. Integral at 2.2×10^9 cfu/mL provided significant increase of seedling heights over other treatments under greenhouse conditions. The Integral treatments of ST ＋ SD ＋ FS at 2.2×10^9 cfu/mL significantly suppressed ShB over other treatments. In field studies, Integral provided significant increase of seedling height in nursery, and number of tillers per plant, compared with the control.
