New insights into developmental biology of Eimeria tenella revealed by comparative analysis of mRNA N6-methyladenosine modification between unsporulated oocysts and sporulated oocysts

Evidence showed that N6-methyladenosine(m^(6)A)modification plays a pivotal role in influencing RNA fate and is strongly associated with cell growth and developmental processes in many ***,no information regarding m^(6)A modification in Eimeria tenella is currently *** the present study,we surveyed the transcriptome-wide prevalence of m^(6)A in sporulated oocysts and unsporulated oocysts of *** RNA immunoprecipitation sequencing(MeRIp-seq)analysis showed that m^(6)A modification was most abundant in the coding sequences,followed by stop *** were 3,903 hypermethylated and 3,178 hypomethylated mRNAs in sporulated oocysts compared with unsporulated *** joint analysis suggested that m^(6)A modification of the majority of genes was positively correlated with mRNA *** mRNA relative expression and m^(6)A level of the selected genes were confirmed by quantitative reverse transcription pCR(RT-qpCR)and *** and KEGG analysis indicated that differentially m^(6)A methylated genes(DMMGs)with significant differences in mRNA expression were closely related to processes such as regulation of gene expression,epigenetic,microtubule,autophagy-other and TOR ***,a total of 96 DMMGs without significant differences in mRNA expression showed significant differences at protein *** and pathway enrichment analysis of the 96 genes showed that RNA methylation may be involved in cell biosynthesis and metabolism of *** firstly present a map of RNA m^(6)A modification in ***,which provides significant insights into developmental biology of ***.

Autophagy plays a pro-apoptotic role in arsenic trioxide-induced cell death of liver cancer

Objective: The effects of arsenic trioxide(As2O3) on hepatocellular carcinoma have been documented widely. Autophagy plays dual roles in the survival and death of cancer cells. Therefore, we investigated the exact role of autophagy in As2O3-induced apoptosis in liver cancer ***: The viability of hepatoma cells was determined using the MTT assay with or without fetal bovine serum. The rate of apoptosis in liver cancer cells treated with As2O3was evaluated using flow cytometry, Hoechst 33258 staining, and TUNEL assays. The rate of autophagy among liver cancer cells treated with As2O3was detected using immunofluorescence, Western blot assay and transmission electron ***: Upon treatment with As2O3, the viability of HepG2 and SMMC-7721 cells was decreased in a time-and dose-dependent manner. The apoptosis rates of both liver cancer cell lines increased with the concentration of As2O3, as shown by flow cytometry. Apoptosis in liver cancer cells treated with As2O3was also shown by the activation of the caspase cascade and the regulation of Bcl-2/Bax ***, As2O3treatment induced autophagy in liver cancer cells;this finding was supported by Western blot, immunofluorescence of LC3-II and beclin 1, and transmission electron microscopy. In liver cancer cells, As2O3inhibited the phosphoinositide 3-kinase/protein kinase B/mammalian target of rapamycin(pI3K/AKT/mTOR) signal pathway that plays a vital role in both apoptosis and autophagy. The pI3K activator SC-79 partially reversed As2O3-induced autophagy and apoptosis. furthermore, inhibiting autophagy with 3-methyladenine partially reversed the negative effects of As2O3on cell *** starvation increased autophagy and amplified the effect of As2O3on cell ***: As2O3induces apoptosis and autophagy in liver cancer cells. The practice and Exploration of Applying EBM to Bilingual Teaching of Medical Genetics at OSBCM

In the process of teaching medical genetics of undergraduate clinical medicine, the practice and exploration of applying EBM to the bilingual teaching of OSBCM medical genetics are carried out. Using CBL and pBL as the carrier can make up for the shortcomings of a single teaching mode, synthesize the advantages of multiple teaching modes. It starts from integrating the basic theoretical knowledge of medicine and clinical practice knowledge, improving students’ bilingual level of medical genetics, cultivating students’ literature retrieval ability, and promoting early clinical, multi-clinical and repeated clinical consciousness for medical students. Therefore, it is more conducive to cultivate students’ ability to learn independently, accurately analyze and solve problems, improve medical students’ clinical thinking ability and scientific research awareness, improve medical students’ ability of international communication, and lay a solid foundation for improving medical students’ future post competence, innovative spirit and lifelong learning ability.

针对空间站暴露载荷操作任务,提出一种组合臂安装载荷的自主安全操控策略。该策略由机械臂末端视觉伺服进行安装操作的精细初定位,克服天地差异、舱体充压变形、组合臂大范围开环运动等误差影响,为机械臂安装载荷提供准确的初始条件;由力控制和自主交互流程进行载荷对接安装,适应载荷对接前残余位姿偏差,减少天地、机械臂与载荷交互时间、力控制持续时间,降低宏微机械臂间动力学耦合影响,自主柔顺和安全完成载荷插拔操作。该操控策略经过地面仿真和试验验证,并成功应用在组合臂操作舱外暴露载荷任务中,确保空间站载荷安装任务圆满成功。

Effect of electroacupuncture at Neiguan(pC6)at different time points on myocardial ischemia reperfusion arrhythmia in rats

OBJECTIVE:To observe the effects of electroacupuncture at Neiguan(pC6)at different time points on reperfusion arrhythmia(RA)after myocardial ischemia and reperfusion in rats,and to investigate the correlation of this protective effect with nerve growth factor(NGf),tyrosine kinase A(Trk A),tyrosine hydroxylase(TH),and norepinephrine(NE).METHODS:A total of 72 Sprague-Dawley male rats were randomly divided into six groups(n=12 rats/group):normal group(Norm),sham operation group(Sham),ischemia reperfusion group(I/R),pre-ischemic electroacupuncture group(EAI),pre-reperfusion electroacupuncture group(EAII),post-reperfusion electroacupuncture group(EAIII).The myocardial ischemia-reperfusion injury(MIRI)model was induced by occlusion of left anterior descending coronary artery for 20 min followed by reperfusion for 40 min in *** no intervention in the Norm group and only threading without ligation in the Sham *** pretreatment at 20 min/d for 7 d before ligation in the EAⅠgroup,20 min of electroacupuncture before reperfusion in the EAII group and 20 min of electroacupuncture after reperfusion in the EAIII *** electrocardiogram(ECG)of each group was recorded throughout the whole process,and the success of the MIRI model was determined based on the changs of J-point and T-wave in the *** arrhythmia score was used to record premature ventricular contractions,ventricular tachycardia and ventricular fibrillation during the reperfusion period to assess the reperfusion induced *** expression levels of NGf,Trk A,TH protein were measured by Western ***,the expression levels of plasma and myocardial NE levels were detected by enzyme linked immunosorbent ***:The differences between Norm group and Sham group were not statistically significant in all *** score,myocardial NGf,Trk A,TH,and NE expression were significantly higher in the I/R group compared with the Sham *** score,myocardial NGf,T

fruit wart is an important appearance trait influencing consumer preferences of bitter gourd(Momordica charantia L.).The molecular genetic mechanisms underlying fruit wart formation in bitter gourd are largely *** this study,genetic analysis based on four generations showed that fruit wart formation in bitter gourd was controlled by a single dominant locus named as *** fwa locus was initially mapped into a 4.82 Mb region on pseudochromosome 4 by BSA-seq analysis and subsequently narrowed down to a 286.30 kb region by linkage analysis.A large f2population consisting of 2360 individuals was used to screen recombinants,and the fwa locus was finally fine mapped into a 22.70 kb region harboring four protein-coding genes through recombination ***04g1399,encoding an epidermal patterning factor 2-like protein,was proposed as the best candidate gene for fwa via sequence variation and expression *** addition,a 1-bp insertion and deletion(InDel)variation within MC04g1399 was converted to a cleaved amplified polymorphic sequence(CApS)marker that could precisely distinguish between the warty and non-warty types with an accuracy rate of 100%among a wide panel of 126 bitter gourd germplasm *** results not only provide a scientific basis for deciphering the molecular mechanisms underlying fruit wart formation but also provide a powerful tool for efficient genetic improvement of fruit wart via marker-assisted selection.

本文提出了一种基于模型定义的光学遥感卫星星上处理系统设计方法,构建了“硬件资源-算子模块-通用处理核-典型应用”的星上处理任务行为模型范式。在硬件层面,采用星上异构嵌入式计算平台,将多处理器一体化设计,通过标准化高速数据互连,完成高速信号传输和数据处理;通过网络拓扑实现良好的扩展性,支持设备规模及数据处理复杂性的变化;在软件层面,针对星上智能处理任务中频繁数据读写操作、大量重复计算操作、卷积神经网络的通用计算与加速等需求,设计了星上平台指令集、星上算法通用算子和星上智能网络组件组成的可配置算子模块,并基于该模块可快速实现特定算法的硬件Ip核。经仿真试验验证,本文方法可根据卫星平台和星上处理任务需求,按需适配最佳软硬件解决方案,并有效提高了计算资源利用率;提出的结合云检测的实时流水压缩编码方案,显著提升了压缩性能;设计的轻量化目标检测识别方法,计算资源效率达到91.5%;以高分一号、高分七号原始数据率为例进行分析,相比常规方案整体计算资源利用率分别提高了16.51%、17.77%。实现了星上处理系统研制过程模型化、工作模式可定义、逻辑资源共享化,是适应卫星小型化、快速部署的一种合理优化选择。
