Human hepatopoietin augmenter of liver regeneration──A hepatotrophic factor for liver regeneration, and its potential antihepatitis effect in vivo

The effect of human hapatopoietin i.e. augmenter of liver regeneration (hALr) was determined on hepatocyte DNA synthesis, and on CCl 4-induced hepatitis in animal in vitro and in vivo. It is found that ALr could directly stimulate DNA synthesis of hepatocytes in primary culture in a dose-dependent manner. In 3.% hepatectomied rats, significant DNA synthesis occurred in control rats ; even so, exogenous hALr gene encoding protein increased DNA synthesis of regeneration liver by 2.3.fold comp.r.d with control rats. A lower dose of CCl 4 was administrated in rats and the effect of ALr on DNA synthesis of regenerating liver 48 h after CCl 4 administration was analyzed. Few Budr-labeled hepatocytes were visible in control rats. However, exogenous hALr markedly increased the number of labeled cells in a dose-dependent manner. Statistical analysis showed that 10 or 40 μg·kg -1 ALr protein stimulated DNA synthesis by 1.7 and 4.8 fold respectively. In addition to enhancing cell growth, adiministration of hALr achieved a significant improvement in reversing the lethality of rat hepatic failure when comp.r.d with that of control group; the elevation of cytosolic enzymes was dramatically sup.r.ssed by exogenous hALr in CCl 4-treated mice in vivo and in vitro; histologically, hepatocytes around the central vein were necrotic, and the degree of hepatocyte necrosis in control mice was more prominent than that in the mice given 40 μg·kg-1 hALr 48 h after CCl 4 administration. We also noted that hALr had a strong antihepatitis effect in vitro which was determined with primary cultured rat hepatocytes. These findings suggest that hALr protects the integrity of hepatocytes against severe hepatitis, and indicate that ALr may be an imp.r.ant regulator of liver regeneration and play a major role in liver injury repair. It proves ALr adminstration to be a useful treatment to accelerate liver regeneration and to prevent the onset of hepatitis or intrahepatic cholestasis induced by toxin.

The diagnosis of prostate diseases by magnetic resonance imaging (MrI) in vivo is sometimes difficult for the lower signal to noise ratio (SNr). To increase the SNr of the prostate image, we designed a rF coil that can be inserted into rectum and was named endorectal coil. The prop.r.ies of rF coil were evaluated using a network analyzer. Moreover the images and spectroscopy of a special phantom were acquired and the results were comp.r.d to those of the commercial TOrSO coil (G.E. Medical Systems, USA). Our coil gave a significantly higher SNr at the region of interest (rOI). The achieved high local SNr and resulting high spatial resolution would add more anatomic and biochemical information to the diagnosis of prostate diseases.

The functional magnetic resonance imaging (fMrI) based on blood oxygen level dependent (BOLD) contrast has emerged as one of the most potent noninvasive tools for mapping brain function and has been widely used to explore physiological, pathological changes and mental activity in the brain. Exploring the nature and prop.r.y of BOLD signal has recently attracted more attentions. Despite that great progress has been made in investigation of the characteristics and neurophysiological basis, the exact nature of BOLD signal remains unclear. In this pap.r.we discuss the characteristics of BOLD signals, the nonlinear BOLD response to external stimuli and the relation between BOLD signals and neural electrophysiological recordings. Furthermore, we develop our new opinions regarding nonlinear BOLD response and make some p.r.pectives on future study.

Current serum neutralization assays based on the inhibition of the cytopathic effect（Nt-CpE） need to ma nipulate live viruses, which are time-consuming, labor-intensive, and have the potential exposure to infectious agents, so a safe and objective assay via pseudovirus for the fast and efficient detection of enterovirus 71（EV71） neutralizing antibodies was developed. First, we generated EV71 pseudovirus containing firefly luciferase gene in place of the capsid gene p1 in EV71 genome. Vero cells infected with 2.0 CCID50（50% cell culture infective dose） of EV71 pseudovirus for 2. h were found to have the best p.r.ormance. Seval sera were measured by EV71 pseudop.r.icle neutralization assay（Nt-ppN） and the conventional serological method Nt-CpE. Neutralizing antibody titers measured by Nt-ppN and those obtained by Nt-CpE demonstrate a high correlation between the two methods. Overall, the ppN assay represents a valid alternative to conventional serological methods for the evaluation of EV71 neutralizing anti bodies. This method can be used for detecting neutralizing antibodies of other picornaviruses, such as hepatitis A vi rus（HAV） and coxsackievirus 16（CVA16）, and make it possible to determine whether there is cross-reactivity be tween EV71 and CVA16.

Natural antioxidants, due to the different chemical structures and their unique biological protection effects on islet cell, have been received much attention in applied chemistry field for developing a new diabetic drug. However, the molecular targets of the islet cell protection have been remained obscure. For investigating molecular targets of combined natural antioxidant’s（CNA） roles in against diabetes, type 1 diabetic mouse model was made by multiple injection of low doses of streptozotocin into C57BL mice. Two combinations of natural antioxidants were orally supplemented to the mice. Blood glucose level, glutathione p.r.xidase（GSH-px） and sup.r.xide dismutase （SOD） activities, and malondialdehyde（MDA） contents were measured via biochemical assays. Tumor necrosis factor α（TNF-α）, interferon γ（IFN-γ） and Interleukin-12.IL-12. in CD4＋ p.r.pheral blood mononuclear cells（pBMC） were evaluated with flow cytometry（FCM）. In situ hybridization（ISH） and immunohistochemistry（IHC） were used to detect insulin gene expression. The results demonstrate both the combinations significantly： increased GSH-px and SOD, but decreased MDA levels in pancreas（p〈0.05―0.01）; up.r.gulated insulin expression at both mrNA and protein levels in islet cells（p〈0.05―0.01）; down-regulated TNF-α and IL-12.expression in pBMC（p〈0.05―0.01）. Our data suggest that the CAN may regulate multiple key molecular targets related to β cell function, including reduction of oxidative stress and pre-inflammatory cytokines, and upregulate insulin gene expression to reduce the development of diabetes.

p.r.utaneous coronary interventions have progressed through the era of plain balloon dilation, bare-metal stent insertion to drug-eluting stent treatment, which has significantly reduced the acute occlusion and restenosis rates of target vessels and improved patient prognosis, making drug-eluting stents the mainstream interventional treatment for coronary artery disease. In recent years, drug-coated balloons(DCBs) have become a new treatment strategy for coronary artery disease, and the drugs used in the coating and the coating technology have progressed in the past years. Without p.r.anent implant, a DCB delivers antiproliferative drugs rapidly and uniformly into the vessel wall via the excipient during a single balloon dilation. Many evidence suggests that DCB angioplasty is an effective measure for dealing with in-stent restenosis and de novo lesions in small coronary *** more clinical studies are published, new evidence is emerging for the use of DCB angioplasty in a wide range of coronary diseases, and the indications are expanding internationally. Based on the latest research from China and elsewhere, the Exp.r. Writing Committee of the Chinese Exp.r. Consensus on Clinical Applications of Drug-Coated Balloon has updated the previous DCB consensus after evidence-based discussions and meetings in terms of adequate prep.r.tion of in-stent restenosis lesions, expansion of the indications for coronary de novo lesions, and precise guidance of DCB treatment by intravascular imaging and functional evaluation.

Since the theory of generalized two-dimensional (2.D) correlation spectroscopy was proposed, it has been keenly concerned in scientific research and its analytical method has been widely applied in various analytical fields. The mathematical process to construct generalized 2.D correlation spectroscopy and the physical meaning of 2.D correlation spectral map are described, and three examples in the fields of chemical analysis and molecular biology are provided, such as the component analysis of organic solvent, the analysis of biological molecules in the solvent with different pH values and structural analysis of protein. The theory and analytical method of generalized 2.D correlation spectroscopy are also detailedly commented.

A 69-year old female patient was admitted because of 3.days of worsened chest *** angiography showed 60% stenosis of distal left main stem,chronic total occlusion of left anterior descending (LAD),70% stenosis at the ostium of a small left circumflex,70-90% stenosis at the p.r.xysmal and middle p.r. of a dominant right coronary artery (rCA),and a normal left internal mammary artery (LIMA) with normal origination and *** intervention was attempted but failed on the occluded lesion of *** patient received minimally invasive direct coronary artery bypass (MIDCAB) with left LIMA isolation by Davinci *** days later,the rCA lesion was treated by Sirolimus rapamicin eluting stents implantation *** the patient was discharged uneventfully after 3.days *** exp.r.ence suggests that two stop shops of hybrid technique be feasible and safe in the treatment of elderly patient with multiple coronary diseases.
