Bacteria capable of denitrification play a significant role in the nitrogen cycle of freshwater ecosystems. By metabolizing nitrogen compounds they e.g. counteract the eutrophication of natural waters. To get detailed insights into the in situ turnover rates of nitrogen a reliable tool of quantification for active microorganisms is essential. In the present investigation, quantification capabilities of a molecular tool (polymerase Chain Reaction—pCR) and a cultivation based tool (Most probable number—MpN) were investigated and compared. The total bacterial concentration yielded by the molecular pCR approach was up to 6-fold higher compared to the results of the MpN approach. However, the portion of culturable denitrifying bacteria compared to the number of specific gene copies (nirS) was much lower. Depending on the environmental conditions, the difference between the pCR and the MpN approach was up to three orders of magnitude. From lab scale experiments with a pure p. aeroginosa strain it can be concludes, that these differences are not the result of inappropriate culture conditions but rather reflect the portion of so called viable but not culturable bacteria (VBNC). Low nitrate concentrations as found in many fresh water ecosystems induced a significant increase in the portion of non culturable denitrifying bacteria. Referred to the investigation of dynamic populations, the number of metabolic active bacteria is represented by the MpN rather than by the pCR approach.

Atherosclerosis is the most prevalent disease in middle and later years of human life. Heart attacks and strokes are among the most frequent causes of death. New approaches in therapy are the use of NSAID’s (nonsteroidal antiphlogistical drugs) of the new generation (selective inhibitors of COX-2) and DHEA (Dehydroepiandrostenon). The key enzyme of prostaglandin synthesis, the COX-2 isoenzyme, is predominantly found in inflammatory tissue. Out of this results a possible importance of COX-2-inhibitors in prophylaxis of atherosclerosis. Our study intended to examine the significance of COX-2 and the COX-2-formed prostanoids, as well as the significance of COX-2-independent iso-prostanes on atherosclerosis. For that purpose we tested the effect of the selective COX-2-inhibitor Celecoxib on rabbits fed with cholesterol and compared this with the effect of the steroid hormone DHEA in 4 groups: healthy control, cholesterol-fed control, DHEA-group and Celecoxib group. In the test prostanoids, nitrate and nitrite were measured by gas chromatograpy/tandem-mass-spectrometry (GC-MS/MS) in 24-hours-collected urine. Additionally we measured cholesterol and triglycerides in plasma. The aortas of the examinated animals were measured optically using a planimetrical method. The measurement of prostanoids, isoprostanes, nitrate and nitrite showed considerable variations and particulary significant differences (p parison to the cholesterol group and the DHEA-group. Consequently the test demonstrated a significant role of COX-2 in the development of atherogenesis.

The ascending application of enzymes in organic synthesis creates a growing demand for novel biocatalysts. The applied methods for their identification range from microbial enrichment cultures over metagenome screenings to solely computational methods. In this communication, we demonstrate a straightforward screening approach for the detection of novel biocatalysts in fungi belonging to the phylum Basidiomycota. It basically relies on mincing of the whole fruit bodies of freshly collected mushrooms with subsequent direct screening. Suitability was demonstrated with eight different mushrooms which were investigated for carbonyl reductase activity on sterically demanding carbonyl compounds. The results indicate the presence of potentially useful carbonyl reductases (KREDs) in all tested fungi. Closer characterization of the preparation from pigskin poison puffball (Scleroderma citrinum) showed the presence of KRED exhibiting a broad substrate range. Thus, applicability of this low-tech screening approach could be verified in this study.

当空心微结构光纤纤芯的尺寸和波长相近时,光在纤芯中的传输大大增强,并会在纤芯周围产生很强的倏逝场。报道了一种新型的纤芯直径仅为2μm的空气悬浮芯微结构光纤,该光纤通过薄片堆积法拉制而成,具有大倏逝场和微米级孔径的单元结构,在532nm波长处的损耗为0.16dB/cm,非常适合用于生化物质的传感探测。以该光纤作为传感探针,结合激光技术搭建了一套简易的荧光光谱探测系统,使用此系统对纳升量级的生物荧光标记材料CdTe/CdS/ZnS量子点进行荧光探测分析。利用该系统可探测荧光量子点的极限约为1nmol/L,相当于3.78×107个量子点,实现了高灵敏度、快速探测。基于空气悬浮芯微结构光纤的荧光检测系统为量子点标记的生物材料的灵敏检测提供了新的方法和思路。

标准物质定值分析的溯源性要求比常规分析高很多。在常规分析中,有证标准物质(CRM)可用于校准以及建立可溯源性。尽管与标准物质生产和认定相关的ISO导则并没有十分明确地提到校准,但众多标准物质生产者在定值分析时却不接受基准标准物质用于校准。这些要求制约了标准物质定值方法的使用,因为这些方法可以采用已知高纯度的标准物质、化学计量法及已知高纯度标准物质的混合物进行校准。有效的固体取样技术,例如通常使用的块状标准物质进行校准的辉光放电质谱法(GD-MS)或火花源发射光谱法(spark-OES),似乎不太适合定值分析。尤其是固体样品中痕量元素分析测定的有力工具辉光放电质谱法,除金属杂质外,使用特定气体混合物,这种方法也可以分析相关的非金属杂质如硫和磷。我们开发了一种可以使用已知高纯度的标准物质混合物和化学计量法的校准方法,这种校准方法同样可用于辉光放电质谱法。与溶液中基体匹配技术类似,此校准方法基于掺杂的粉末压片。本文还结合铜和钢标准物质的定值分析结果进行了陈述。

Alternative mRNA splicing is a fundamental process to increase the versatility of the *** humans,cardiac mRNA splicing is involved in the pathophysiology of heart *** in the splicing factor RNA binding motif protein 20(RBM20) cause severe forms of *** identify novel cardiomyopathy-associated splicing factors,RNA-seq and tissue-enrichment analyses were performed,which identified up-regulated expression of Sam68-Like mammalian protein 2(SLM2) in the left ventricle of dilated cardiomyopathy(DCM) *** the human heart,SLM2 binds to important transcripts of sarcomere constituents,such as those encoding myosin light chain2(MYL2),troponin 13(TNNI3),troponin T2(TNNT2),tropomyosin 1/2(TpM1/2),and titin(TTN).Mechanistically,SLM2 mediates intron retention,prevents exon exclusion,and thereby mediates alternative splicing of the mRNA regions encoding the variable proline-,glutamate-,valine-,and lysine-rich(pEVK) domain and another part of the I-band region of *** summary,SLM2 is a novel cardiac splicing regulator with essential functions for maintaining cardiomyocyte integrity by binding to and processing the mRNAs of essential cardiac constituents such as titin.
