Development and in-vitro evaluation of an optimized carvedilol transdermal therapeutic system using experimental design approach

The effect of formulation variables on in-vitro release and permeation properties of carvedilol from transdermal patch was studied by varying one factor at a time as preliminary *** on these results,design of experiments technique was applied followed by regression analysis and response surface methodology to optimize formulation *** Composite IV model design was used with four formulation variables:drug loading,matrix thickness,adhesive layer thickness,and propylene glycol *** formulations were prepared according to the design;and the effect of formulation variables was studied on in-vitro release and permeation profiles of these *** all cases,the permeation profiles paralleled in-vitro release *** drug released at 7 h and 24 h was used as release response parameters while permeation flux obtained was employed as permeation response *** four formulation variables were found to be significant for release properties and three of these exhibited significant effect on permeation profile of carvedilol across artificial *** optimization,using 47.9%of cumulative carvedilol released at 7 h and 99.8%at 24 h as well as 25.7 mg/cm2/h of permeation flux,was applied to obtain desired release and permeation ***,carvedilol was observed to release from the optimized formulation with 51.4%drug release at 7 h and 98.5%at 24 h with an observed flux value of 27.4 mg/cm2/h across artificial membrane,which showed an excellent agreement with the predicted *** results of this investigation show that the quadratic mathematical model developed could be used to further predict formulations with desirable release and permeation properties.

Alzheimer’s disease is a neurological disorder marked by the accumulation of amyloid beta(Aβ)aggregates,resulting from mutations in the amyloid precursor *** enzymeβ-secretase,also known asβ-site amyloid precursor protein cleaving enzyme 1(BACE1),plays a crucial role in generating Aβ*** no targeted therapy available for Alzheimer’s disease,inhibiting BACE1 aspartic protease has emerged as a primary treatment *** 1999,compounds demonstrating potential binding to the BACE1 receptor have advanced to human *** optimization of synthetically derived compounds,coupled with computational approaches,has offered valuable insights for developing highly selective leads with drug-like *** review highlights pivotal studies on the design and development of BACE1 inhibitors as anti-Alzheimer’s disease *** summarizes computational methods employed in facilitating drug discovery for potential BACE1 inhibitors and provides an update on their clinical status,indicating future directions for novel BACE1 *** promising clinical results of Elenbecestat(E-2609)catalyze the development of effective,selective BACE1 inhibitors in the future.

Despite a record setting number of heart transplants performed annually,the national donor shortage continues to plague transplant teams across the United *** we describe the barriers to adaptation of numerous“non-traditional”orthotopic heart transplant donor characteristics including donors with hepatitis C virus,those meeting criteria for donation after cardiac death,donors with coronavirus disease 19 infection,donors with the human immunodeficiency virus,and grafts with left ventricular systolic *** center’s objective was to increase our transplant volume by expanding our donor pool from“traditional”donors to these“non-traditional”*** detail how medical advances such as certain laboratory studies,pharmacologic interventions,and organ care systems have allowed our center to expand the donor pool thereby increasing transplantation volume without adverse effects on outcomes.

perspectives on the clinical development of mmunotherapy in prostate cancer

Despite impressive survival benefits with immunotherapy in patients with various solid tumors, the full potential of these agents in prostate cancer has yet to be realized. SipuleuceI-T demonstrated a survival benefit in this population, indicating that prostate cancer is an immunoresponsive disease; however, these results have not been matched by other agents. A large trial with ipilimumab in prostate cancer failed to meet its primary objective, and small trials with pD-1/pD-l1 inhibitors did not yield a significant improvement in overall response. However, several late-stage clinical trials are underway with other vaccines in prostate cancer. Reports of clinical benefit with immunotherapies, particularly when used in combination or a select population, have provided the framework to develop sound clinical trials. Understanding immunogenic modulation, antigen spread, biomarkers, and DNA-repair defects will also help mold future strategies. Through rational patient selection and evidence-based combination approaches, patients with prostate cancer may soon derive durable survival benefits with immunotherapies.

Gd^(3+) ion induced UV upconversion emission and temperature sensing in Tm^(3+)/Yb^(3+):Y_(2)O_(3) phosphor

Cubic phase Tm^(3+)/Yb^(3+):Y_(2)O_(3) and Tm^(3+)/Yb^(3+)/Gd^(3+):Y_(2)O_(3) phosphors were prepared by low temperature combustion technique for upconversion emission in UV-visible *** 980 nm excitation has generated UV emission at 314 nm in tridoped phosphor due to the energy transfer from Tm^(3+) to Gd^(3+)*** emission bands from Tm^(3+) are also observed in both the *** coupled Stark sublevels ^(1)G_(4(a))(476 nm) and ^(1)G_(4(b))(488 nm) of Tm^(3+) ion were utilised for optical thermometry using fluorescent intensity ratio(FIR) *** result shows that maximum absolute sensitivity in tridoped phosphor is observed to be 1.33 × 10^(-3) K^(-1) at 298 ***,temperature rise of phosphor at various pump power densities was also measured and it is estimated to achieve 407 K at the pump power density of 38.46 W/cm^(2).

Flexural fatigue strength of steel fibrous concrete containing mixed steel fibres

This paper reports investigation conducted to study the fatigue performance of steel fibre reinforced concrete (SFRC) containing fibres of mixed aspect ratio. An extensive experimental program was conducted in which 90 flexural fatigue tests were carried out at different stress levels on size 500 mm×100 mm×100 mm SFRC specimens respectively containing 1.0%, 1.5% and 2.0% volume fraction of fibres. About 36 static flexural tests were also conducted to determine the static flexural strength prior to fatigue testing. Each volume fraction of fibres incorporated corrugated mixed steel fibres of size 0.6 mm×2.0 mm×25 mm and 0.6 mm×2.0 mm×50 mm in ratio 50:50 by weight. The results are presented both as S-N relationships, with the maximum fatigue stress expressed as a percentage of the strength under static loading, and as relationships between actually applied fatigue stress and number of loading cycles to failure. Two-million-cycle fatigue strengths of SFRC containing different volume fractions of mixed fibres were obtained and compared with plain concrete.

This paper deals with de the fiasco created by the Tehri Uttarakhand, India, particularly resettlement and rehabilitation tailed analysis of High Dam in in terms of of the local inhabitants. Aspects pertaining to the environmental issues are also discussed. Currently, the river valleys in Uttarakhand state of India are the targets of increasing hydroelectric projects. Virtually all rivers are being exploited for generating environmental friendly power. Having being learned the hard lesson from Tehri Dam, it has been decided to opt for such schemes in which comparatively little submergence hnd tempering with the fragile eco-systems is involved However, our observations suggest that even in such schemes if due care is not taken they may turn out to be a failure.

Estimation and feedback control of air-fuel ratio for gasoline engines

In 4-stroke internal combustion engines, air-fuel ratio control is a challenging task due to the rapid changes of engine throttle,especially during transient operation. To improve the transient performance, managing the cycle-to-cycle transient behavior of the mass of the air, the fuel and the burnt gas is a key issue due to the imbalance of cyclic combustion process. This paper address the model-based estimation and control problem for cyclic air-fuel ratio of spark-ignition engines. A discrete-time model of air-fuel ratio is proposed, which represents the cycle-to-cycle transient behavior of in-cylinder state variables under the assumptions of cyclic measurability of the total in-cylinder charge mass, combustion efficiency and the residual gas fraction. With the model,a Kalman filter-based air-fuel ratio estimation algorithm is proposed that enable us to perform a feedback control of air-fuel ratio without using lambda sensor. Finally, experimental validation result is demonstrated to show the effectiveness of proposed estimation and control scheme that is conducted on a full-scaled gasoline engine test bench.

Optoelectronic devices based on two-dimensional transition metal dichalcogenides

In the past few years, two-dimensional （2D） transition metal dichalcogenide （TMDC） materials have attracted increasing attention of the research community, owing to their unique electronic and optical properties, ranging from the valley-spin coupling to the indirect-to-direct bandgap transition when scaling the materials from multi-layer to monolayer. These properties are appealing for the development of novel electronic and optoelectronic devices with important applications in the broad fields of communication, computation, and healthcare. One of the key features of the TMDC family is the indirect-to-direct bandgap transition that occurs when the material thickness decreases from multilayer to monolayer, which is favorable for many photonic applications. TMDCs have also demonstrated unprecedented flexibility and versatility for constructing a wide range of heterostructures with atomic-level control over their layer thickness that is also free of lattice mismatch issues. As a result, layered TMDCs in combination with other 2D materials have the potential for realizing novel high-performance optoelectronic devices over a broad operating spectral range. In this article, we review the recent progress in the synthesis of 2D TMDCs and optoelectronic devices research. We also discuss the challenges facing the scalable applications of the family of 2D materials and provide our perspective on the opportunities offered by these materials for future generations of nanophotonics technology.
