S100B protein in tissue development,repair and regeneration

The Ca 2+-binding protein of the EF-hand type,S100B,exerts both intracellular and extracellular regulatory *** an intracellular regulator,S100B is involved in the regulation of energy metabolism,transcription,protein phosphorylation,cell proliferation,survival,differentiation and motility,and Ca 2+ homeostasis,by interacting with a wide array of proteins(i.e.,enzymes,enzyme substrates,cytoskeletal subunits,scaffold/adaptor proteins,transcription factors,ubiquitin E3 ligases,ion channels) in a restricted number of cell *** an extracellular signal,S100B engages the pattern recognition receptor,receptor for advanced glycation end-products(RAGE),on immune cells as well as on neuronal,astrocytic and microglial cells,vascular smooth muscle cells,skeletal myoblasts and ***,RAGE may not be the sole receptor activated by S100B,the protein being able to enhance bFGF-FGFR1 signaling by interacting with FGFR1-bound bFGF in particular cell ***,extracellular effects of S100B vary depending on its local *** evidence suggests that at the concentration found in extracellular fluids in normal physiological conditions and locally upon acute tissue injury,which is up to a few nM levels,S100B exerts trophic effects in the central and peripheral nervous system and in skeletal muscle tissue thus participating in tissue *** present commentary summarizes results implicating intracellular and extracellular S100B in tissue development,repair and regeneration.

根据中意国际合作项目'四川无病毒果苗繁育中心'的要求,为了弄清四川葡萄产区内病毒病的种类、危害程度及其流行规律,中意双方成立了联合工作组,并于2002年9月在成都龙泉及攀枝花两地开展了葡萄病毒病的田间普查和采样.考察期间共收集葡萄枝样50份,随后送往意大利巴厘大学,经双抗体和三抗体夹心酶联免疫吸附法测试,共检出了6种危险性病毒种类及株系.现对它们在田间的症状表现及其流行趋势作如下阐述.

Cultivation of Microalgae <i>Monoraphidium</i>sp., in the plant pilot the Grand Valle Bio Energy, for Biodiesel production

At present, Brazil imports approximately 11 billion liters/year of diesel. With the interruption of the works in the new petrobras refineries, the projection is that by 2025 this volume will increase to 24.2 billion liters of diesel/year. In this sense, the biodiesel factory Grand Valle Bio Energy Ltda., located in the state of Rio de Janeiro, in conjunction with the FApERJ makes some investments in technology development for the cultivation and use of microalgae as an alternative raw material in the production of biodiesel. Based on arguments previously said, this work presents the results of the microalgae cultivation Monoraphidium sp. in photobioreactors the pilot plant of the company. The installation with an area of 120 m2 is included with 2 open photobioreactors of type falling film (20 m × 1 m), with a cascade of 18mm and capacity of 4000 L. The lineage cultivated is selected from previous ecophysiological studies that are identified as promising for biodiesel production by having a high potential for the production of lipids. This lineage is maintained at collection of the stock of cultures Laboratory of Green Technologies of the School of Chemistry/ UFRJ. The cultivation was performed in means ASM-1 (Gorham et al., 1964), initial pH 8.0, with aeration and circulation average of 8 hours a day during 19 days. The culture was started with an inoculum of 1 × 107 cel/ml. The lipid production was determined in two phases of growth: on day 4 (exponential phase) and 15 day (stationary phase). For the determination and quantification of lipid content, two different methods were assessed for a sample of biomass, submitted to the same processes the separation and drying. The results showed the methodology of Bligh & Dyer with modifications as the most efficient in extracting lipids. The total lipid content of the biomass Monoraphidium sp. was 30.58%. The growth rate varied between 0.74 ± 0.01 and 0.68 ± 0.02.

Yield, Water Use Efficiency and physiological Characteristic of “Tommy Atkins” Mango under partial Rootzone Drying Irrigation System

This study was conducted to evaluate the yield, water use efficiency and physiologic characteristics of “Tommy Atkins” mango under partial rootzone drying. The experiment was carried out in an orchard of mango in the irrigated perimeter of Ceraíma, BA. Five treatments and six repetitions were used with one tree per experimental plot: 01—Full Irrigation, 100% of ETc, 02—100% of ETc with frequency of alternance (FA) of 15 days, 03—80% ETc with FA of 15 days, 04—60% of ETc with FA of 15 days, 05—40% ETc with FA of 15 days. The treatments were conductedfrom the beginning of flowering to the harvest of mango in an orchard irrigated by drip. There was no significant reduction in productivity of “Tommy Atkins” mango under pRD up to 40% of ETc and pRD with 40% ETc resulted in greater water use efficiency. There was no significant difference among the averages of photosynthetically active radiation, Qleaf;leaf temperature, Tl;internal CO2 concentration, Ci;transpiration, E;stomatal conductance, gs;photosynthesis, A;carboxylation efficiency, A/Ci;quantum efficiency of photosynthesis, A/Qleaf and instantaneous water use efficiency, A/E between full irrigation and pRD with different water depth. The increase in leaf temperature causes reduction in A/Qleaf and A/E, however, it increases the transpiration. Increased gs results in higher rates of E, A and A/Ci. The use of the pRD with 40% of ETc causes no interference in gas exchange, maintains the yield and increases water use efficiency in “Tommy Atkins” mango tree.

Induction of IgA and sustained deficiency of cell proliferative response in chronic hepatitis C

AIM:In the present study,antibody and peripheral blood mononuclear cells (pBMC) proliferative responses against hepatitis C virus (HCV) antigens were evaluated in HCV chronically infected patients. METHODS:paired serum and pBMC samples were taken six months apart from 34 individuals,either treated or not,and tested by enzyme-linked immunosorbent assay (ELISA) and carboxyfluorescein succinimidyl ester staining. RESULTS:Over 70% of the patients showed specifi c IgG and IgM against capsid,E1 and NS3,while HVR-1 was recognized by half of the patients. An increase in the levels of the anti-capsid IgM (p = 0.027) and IgG (p = 0.0006) was observed in six-month samples,compared to baseline. Similarly,a signifi cantly higher percent of patients had detectable IgA reactivity to capsid (p = 0.017) and NS3 (p = 0.005) after six months,compared to baseline. particularly,IgA against structural antigens positively correlated with hepatic damage (p = 0.036). IgG subclasses evaluation against capsid and NS3 revealed a positive recognition mediated by IgG1 in more than 80% of the individuals. On the contrary,less than 30% of the patients showed a positive proliferative response either of CD4+ or CD8+ T cells,being the capsid poorly recognized. CONCLUSION:These results confi rm that while the cellular immune response is narrow and weak,a broad and vigorous humoral response occurs in HCV chronic infection. The observed correlation between IgA and hepatic damage may have diagnostic significance,although it warrants further confi rmation.

肿瘤性心包炎发现在癌瘤扩散患者的尸检中10～20%肿瘤已累及心脏及心包,而心包是转移性病变的最好发部位,占心脏受累的85%。在心包转移瘤病例中,有渗出液者仅12～25%。转移癌累及心脏的发病率已有增加,可能是使用化疗及放疗增加生存期的结果。心包恶性肿瘤大多起源于心外,男性主要来自肺,女性主要来自乳房,但任何原发肿瘤均可能转移至心包。如黑色素瘤、多发性骨髓瘤及结肠腺癌等。妇科肿瘤同所有发生在膈下的原发恶性病变一样,不是恶性心包病常见的原因。转移至心包的妇癌大多数来源于子宫(宫体+宫颈)或卵巢,组织学诊断有鳞癌、腺癌和肉瘤。

Antifungal activity of total and fractionated phenolic extracts from two wild edible herbs

The chemical characterization of phenolic extracts of salad burnet (Sanguisorba minor Scop. s.l.) and broomrape (Orobanche crenata Forsk.) was performed. Active fractions were separated and characterized and, in the case of O. crenata, two phenolic compounds, verbascoside and is overbascoside, were quantitatively extracted, purified and characterized. These compounds proved to be effective in reducing the growth of some important fungi responsible for the decay of fresh fruit and vegetables in postharvest. Verbascoside showed significant activity against penicillium italicum, fairly inhibited Monilinia laxa, p. expansum, and Aspergillus carbonarius, and showed little activity against Botrytis cinerea, M. fructicola and p. digitatum. Isoverbascoside completely inhibited B. cinerea, p. digitatum, p. italicum, and p. expansum;it fairly inhibited M. laxa and A. carbonarius;while it showed to be less effective against M. fructicola. The obtained results support the possible use of some wild edible plants as a source of phenolic substances for the postharvest biological control of fresh fruit and vegetable rots.

目的评价5种地西泮类衍生物B3、B7、B8、B26、B30抗血吸虫效应并探讨其作用机制。方法取雄性日本血吸虫和曼氏血吸虫成虫(10条/皿)培养于DMEM培养液中,分别加入5种地西泮衍生物B3、B7、B8、B26、B30,工作浓度均为50 mol/mL,培养16h,在体视显微镜下观察药物洗涤后24h～72h的虫体存活状态,计算其存活率及活力降低率。以细胞肌松素D和钙通道阻滞剂预处理虫体1h后再加B3和B30共同培养16h,观察拮抗效应。结果在B3实验组中,日本血吸虫存活率和活力降低率为0%和100%,曼氏血吸虫为20%和93.3%,B30实验组中,日本血吸虫和曼氏血吸虫的存活率分别为0%和13%;两者的活力降低率则分别为100%和94.3%。B7、B8、B26实验组抗日本血吸虫效应不明显,但对曼氏血吸虫的作用略好于日本血吸虫。细胞肌松素D可显著拮抗B3、B30的抗日本血吸虫效应,拮抗后B3实验组虫体存活率和活力降低率分别为80%和59%～63%、B30实验组为70%和46%～55%,钙通道阻滞剂尼非地平、尼群地平拮抗B3、B30后虫体存活率为10%～40%,活力降低率为85%～96%。结论地西泮衍生物B3、B30在体外具有显著抗血吸虫效应(p<0.01),且日本血吸虫对该2种衍生物的敏感性好于曼氏血吸虫;而其余3种衍生物的抗曼氏血吸虫作用则更明显。细胞肌松素D对B3、B30具有显著的拮抗作用;钙通道阻滞剂尼非地平、尼群地平也有一定的拮抗效应,提示地西泮衍生物的抗血吸虫效应也可能与钙通道有关。

Leaf nutrient content and transcriptomic analyses of endive (Cichorium endivia) stressed by downpour-induced waterlog reveal a gene network regulating kestose and inulin contents

Endive(Cichorium endivia L.),a vegetable consumed as fresh or packaged salads,is mostly cultivated outdoors and known to be sensitive to waterlogging in terms of yield and ***,metabolic and transcriptomic analyses were used to study variations in curly-(‘Domari’,‘Myrna’)and smooth-leafed(‘Flester’,‘Confiance’)cultivars grown in short-term waterlog due to rainfall excess before *** recording loss of head weights in all cultivars(6-35%),which was minimal in‘Flester’,NMR untargeted profiling revealed variations as influenced by genotype,environment and interactions,and included drop of total carbohydrates(6–50%)and polyols(3–37%),gain of organic acids(2–30%)and phenylpropanoids(98–560%),and cultivar-specific fluctuations of amino acids(−37 to+15%).The analysis of differentially expressed genes showed GO term enrichment consistent with waterlog stress and included the carbohydrate metabolic *** loss of sucrose,kestose and inulin recurred in all cultivars and the sucrose-inulin route was investigated by covering over 50 genes of sucrose branch and key inulin synthesis(fructosyltransferases)and catabolism(fructan exohydrolases)*** lowered expression of a sucrose gene subset together with that of SUCROSE:SUCROSE-1-FRUCTOSYLTRANSFERASE(1-SST)may have accounted for sucrose and kestose contents drop in the leaves of waterlogged *** anti-correlated modules harbouring candidate hub-genes,including 1-SST,were identified by weighted gene correlation network analysis,and proposed to control positively and negatively kestose *** silico analysis further pointed at transcription factors of GATA,DOF,WRKY types as putative regulators of 1-SST.

Biodiesel production Based in Microalgae: A Biorefinery Approach

It is of great knowledge nowadays that the use of fossil fuels is responsible for the emission of gases that intensify the greenhouse effect, which threatens the survival of the humankind. The gravity of this fact could be mitigated through the indirect use of solar energy for fuels derived from vegetable that can be planted and cultivated by the world of renewable and non-polisher. Microalgae play an important role in this regard, as they have promising characteristics as potential raw material for the production of biofuels, able to absorb large amounts of CO2. Chlorophyll organisms convert these simple substances in the atmosphere, absorbing sunlight into chemical energy stored, that is, compounds with high energy, biomass can also be used to obtain biocompounds human nutritional supplement and food animal, however, have been found an important number of difficulties to economically viable production like high cost of production of dry biomass and oil extraction. Here, we review the main approaches of biorefinery concept appearing as an alternative to achieve economic viability of the production of bio-diesel based on microalgae. The major points are the following: 1) use of re-residual water, 2) marketing of Carbon Credits, and 3) development of co-products resulting from high value added.
