The physiopathological Crossroads of Aging

Stress, inflammation and plasminogen activator inhibitor 1 (pAI-1) are key mechanisms throughout the development of aging, constituting a crossroad in the most frequent pathologies that accompany it. Among metabolic processes, obesity, metabolic syndrome and type 2 diabetes mellitus are included and Alzheimer’s disease among the neurodegenerative processes. Stress is a mechanism of defense of the organism against exogenous and endogenous actions called stressors. In the case of low intensity stimuli, the organism responds with actions aimed at a physiological adaptation (Homeostasis). On the other hand, when a high intensity (experimental level) or chronic stimulus (oxidative stress) is repeated, structural and functional changes are observed in different organs with activation of the hypothalamus-pituitary-adrenal axis, the renin angiotensin system and the sympathetic nervous system, stimulating the production of hormones that release cytokines with proin-flammatory/antiinflammatory properties that play an important role in the previously mentioned pathologies, as well as a marked increase in pAI-1, a gene regulated by stress and by cytokines, with manifest action at the origin of thromboembolic disease, so frequent in aging. The objective of this review is to highlight the importance of the binomial stress and pAI-1 in aging and in the pathologies that accompany it. Because pAI-1 is part of the pathology and complications in aging, some authors suggest the study of pAI-1 inhibitors to achieve its physiological levels, as part of the treatment of these diseases.

Although amyloid-β peptide is considered neurotoxic, it may mediate several physiological processes during embryonic development and in the adult brain. The pathological function of amyloid-β peptide has been extensively studied due to its implication in Alzheimer’s disease, but its physiological function remains poorly understood. Amyloid-β peptide can be detected in non-aggregated (monomeric) and aggregated (oligomeric and fibrillary) forms. Each form has different cytotoxic and/or physiological properties, so amyloid-β peptide and its role in Alzheimer’s disease need to be studied further. Neural stem cells and neural precursor cells are good tools for the study on neurodegenerative diseases and can provide future therapeutic applications in diseases such as Alzheimer’s disease. In this review, we provide an outline of the effects of amyloid-β peptide, in monomeric and aggregated forms, on the biology of neural stem cells/neural precursor cells, and discuss the controversies. We also describe the possible molecular targets that could be implicated in these effects, especially GSK3β. A better understanding of amyloid-β peptide (both physiological and pathological), and the signaling pathways involved are essential to advance the field of Alzheimer’s disease.

Effect of particle phase Volume, Shape and Liquid phase Concentrations on Rheological properties of Large particulate-Liquid Model Food Systems by Using Ball Measuring System

Rheological properties of large particulate-liquid model food systems were studied by using the BMS （ball measuring system）. The model food systems were composed of alginate gel particles （-10mm） and a gelatinised starch solution with 1% w/w sodium chloride as a liquid phase. The effects of particle phase volume （Ф, 0-0.60）, particle shapes （cube, sphere, rod and disc） and starch concentrations （3% and 5% w/w starch） were investigated. The power law model was successfully applied to characterize the flow properties of each system and the consistency K and power law index n were obtained. The K increased and n decreased with increasing # for samples at all particle shapes at 3% w/w starch in the liquid phase. The particle effect on the viscosity is further analysed by means of the Krieger-Dougherty model and the maximum packing fraction #,, and the intrinsic viscosity [η] were obtained in each system. The Фm, depended on the particle shape, as expected. The [7] value depended on particle shape and was largely in the order of 4.04 （cube）, 3.28 （disc）, 2.56 （sphere） and 2.32 （rod） at 3% w/w starch. The [η] also depended on starch concentration and was 1.1 at 5%,6 w/w starch in the liquid phase with spherical particles. The present results show successful application of BMS to study the rheological properties of large particulate liquid food systems at relatively small scale experiment （-0.5 L） and also that existing models for suspension rheology are applicable for such food systems to a great extend.

The immune responses of humans and animals to insults(i.e., infections, traumas, tumoral transformation and radiation) are based on an intricate network of cells and chemical messengers. Abnormally high inflammation immediately after insult or abnormally prolonged proinflammatory stimuli bringing about chronic inflammation can lead to life-threatening or severely debilitating diseases. Mesenchymal stem cell(MSC) transplant has proved to be an effective therapy in preclinical studies which evaluated a vast diversity of inflammatory conditions. MSCs lead to resolution of inflammation, preparation for regeneration and actual regeneration, and then ultimate return to normal baseline or homeostasis. However, in clinical trials of transplanted MSCs, the expectations of great medical benefit have not yet been fulfilled. As a practical alternative to MSC transplant, a synthetic drug with the capacity to boost endogenous MSC expansion and/or activation may also be effective. Regarding this, IMT504, the prototype of a major class of immunomodulatory oligonucleotides, induces in vivo expansion of MSCs, resulting in a marked improvement in preclinical models of neuropathic pain, osteoporosis, diabetes and sepsis. IMT504 is easily manufactured and has an excellent preclinical safety record. In the small number of patients studied thus far, IMT504 has been well-tolerated, even at very high dosage. Further clinical investigation is necessary to demonstrate the utility of IMT504 for resolution of inflammation and regeneration in a broad array of human diseases that would likely benefit from an immunoprotective/immunoregenerative therapy.

Effect of a protein-Energy Supplement of Low Consumption with Coconut pulp Expeller (<i>Acrocomia aculeata</i>) of Beef Cattle at pasture

The purpose of this study is to evaluate the effect of a low consumption protein-energy supplement that incorporates 15% of coconut pulp expeller in bovine animals. These pasture fattening cattle were checked on daily weight gain, supplement consumption and efficiency of pasture use. This study was carried out in a livestock establishment in the Department of Concepción, paraguay. A total of 108 animals were selected;castrated male bovines, 342 ± 30.2 of body weight (BW), age average 28 months, Nelore breed distributed in 2 treatments (T1 and T2): T1 = 42 animals;control group (pasture + conventional mineral salt) and T2 = 66 animals;treatment group with a protein-energy supplement (pasture + protein-energy supplement with minerals). In both treatments, the animals grazed on pasture of Brachiaria brizantha cv. Marandú and panincum maximaun cv. Tanzania used a system of pasture rotation of 1 - 3 days of occupation and 35 - 40 days of rest. The average daily weight gain (DG) was 0.82 and 0.83 kg/day (p > 0.05), the supplement consumption was 0.1 and 0.28 kg/day per animal and the % disappearance of the biomass of 18 and 23% for T1 and T2, respectively.
