cD4^(+)T cells can"help"or"license" conventional type 1 dendritic cells(cDc1s)to induce cD8^(+)cytotoxic T lymphocyte(cTL)anticancer responses,as proven in mouse *** recently identified cDc1s with a transcriptomic imprint of cD4^(+)T-cell help,sp.c.fically in T-cell-infiltrated human cancers,and these cells were associated with a good prognosis and response to pD-1-targeting ***,we delineate the mechanism of cDc1 licensing by cD4^(+)T cells in *** cD4^(+)T cells produce IFNβvia the STING pathway,which promotes MHc-I antigen(cross-)presentation by cDc1s and thereby improves their ability to induce cTL anticancer *** cooperation with cD40 ligand(L),IFNβalso optimizes the costimulatory and other functions of cDc1s required for cTL response ***-I-producing cD4^(+)T cells are present in diverse T-cell-infiltrated cancers and likely deliver“help”signals to cTLs locally,according to their transcriptomic profile and colocalization with“helped/licensed”cDcs and tumor-reactive cD8^(+)T *** agreement with this scenario,the presence of IFN-I-producing cD4^(+)T cells in the TME is associated with overall survival and the response to pD-1 checkpoint blockade in cancer patients.

The precise synthesis of subporphyrinoid hybrids withπ-expanded topologies and unique material properties plays a promising role in the design of functional ***,selective,and controllable routes to boron subphthalocyanine−subnaphthalocyanine hybrids,Bsub(pc_(3‑p)-Nc_(p))s,are desirable for this purpose yet synthetically challenging due to random mixtures of c_(s-),c_(3v-),and,in some cases,c_(1)-symmetric compounds that form during traditional statistical mixed ***,we addressed this issue by developing a sterically driven mixed cyclotrimerization with enhanced selectivity for the targeted c_(s)-symmetric hybrid and complete suppression of sterically crowded macrocyclic *** process,coupled with a rationally designed precursor bearing bulky phenyl substituents,enabled the synthesis and characterization of bay-position phenylated ph_(2)-(R_(p))_(8)Bsub(pc_(2)-Nc_(1))hybrids with halogens(Rp.c. or F)in their peripheral isoindole *** selectivity ranged between 59 and 72%with remarkable yields,significantly higher than that of conventional mixed *** findings were augmented by theoretical calculations on precursor Lewis basicity as guiding principles into hybrid macrocycle ***,the incorporation of unfused phenyl groups and halogen atoms into the hybrid framework resulted in fine-tuned optical,structural,electronic,and electrochemical *** straightforward approach achieved improved selectivity and controlled narrowing of the product distribution,affording the efficient synthesis of structurally sophisticated Bsub(pc_(2)-Nc_(1))*** then expands the library of 3-dimensionalπ-extended macrocycles for use in a range of applications,such as in optoelectronic devices with precisely tailored optical properties.

Aims:portal hypertension(pH)is a complication of cirrhosis that leads to hepatic *** the portal pressure gradient(ppG)provides valuable information for disease assessment,staging,and *** this study,we aimed to report the utilization of endoscop.c.ultrasound(EUS)-guided ppG(EUS-ppG)measurements in a real-world ***:This retrosp.c.ive analysis included patients at a tertiary care center who underwent EUS-ppG between February 2021 and May *** most common indication was to establish or exclude the diagnosis of cirrhosis,followed by an assessment of surgical risk in the setting of susp.c.ed *** demographic and clinical data were collected,and statistical analysis was performed using the Student's t ***:Eighteen patients underwent *** success was achieved in 17 of the 18 *** complications were *** mean scores of child-pugh,Model for End-Stage Liver Disease-Sodium,and Fibrosis-4 scores were 5.1±0.5,9.0±3.0,and 3.7±4.5,*** the 18 patients,two presented with esophageal varices and six with portal hypertensive *** mean ppG was 5.8±4.5 mmHg,and five patients exhibited clinically significant pH(cSpH).Of the 18 patients,15 underwent concurrent EUS-liver biopsy,which was 100%successful and provided diagnostic histology for all *** ppG data led to changes in the clinical management of 17 ***:EUS-ppG is safe and technically *** child-pugh score did not correlate well with the presence of cSpH;however,ppG measurements resulted in changes in the management of almost all *** studies correlating EUS-ppG with other assessments of liver diseases are required.
