The current method for combining transgenes into a genome is through the assortment of independent loci, a classical operating system compatible w.th transgenic traits created by different developers, at different times and/or through different transformation techniques. However, as the number of transgenic loci increases over time, increasingly larger populations are needed to find the rare individual w.th the desired assortment of transgenic loci along w.th the non-transgenic elite traits. Introducing a transgene directly into a field cultivar w.uld bypass the need to introgress the engineered trait. However, this necessitates separate transformations into numerous field cultivars, along w.th the characterization and regulatory approval of each independent transformation event. Reducing the number of segregating transgenic loci could be achieved if multiple traits are introduced at the same time, a preferred option if each of the many traits is new.or requires re-engineering. If reengineering of previously introduced traits is not needed, then appending a new.trait to an existing locus w.uld be a rational strategy. The insertion of new.DNA at a known locus can be accomplished by site- specific integration, through a host-dependent homology-based process, or a heterologous site-specific recombination system. Here, w. discuss gene stacking through the use of site-specific recombinases.

Rather than random degradation products, the 18 to 40 nucleotides(nt) transfer RNA-derived small RNAs(tsRNAs) are RNA species generated specifically from pre-RNAs or mature tRNAs in archaea, bacteria and eukaryotes. Recent studies from animal systems have shown that tsRNAs are important non-coding RNAs that regulate gene expression at the transcriptional and/or post-transcriptional levels. They are involved in various biological processes, such as cell proliferation, tumor genesis, stress response and intergenerational epigenetic inheritance. In this review. w. w.ll summarize the discovery, biogenesis, and function of tsRNAs in higher plants. In addition, analysis on tsRNAs from lower plants is shown.

Developing cost-effective and high-performance catalyst systems for dry reforming of methane(DRM)is crucial for producing hydrogen(H_(2))***,w. investigate using iron(Fe)as a promoter and major alumina support in Ni-based catalysts to improve their DRM *** addition of iron as a promotor w.s found to add reducible iron species along w.th reducible NiO species,enhance the basicity and induce the deposition of oxidizable *** incorporating 1 w..%Fe into a 5Ni/10ZrAl catalyst,a higher CO_(2) interaction and formation of reducible"NiO-species having strong interaction w.th support"w.s observed,w.ich led to an∼80%H_(2) yield in 420 min of Time on Stream(TOS).Further increasing the Fe content to 2 w..%led to the formation of additional reducible iron oxide species and a noticeable rise in H_(2) yield up to 84%.Despite the severe w.ight loss on Fe-promoted catalysts,high H_(2) yield w.s maintained due to the proper balance betw.en the rate of CH_(4) decomposition and the rate of carbon deposit ***,incorporating 3 w..%Fe into the 5Ni/10ZrAl catalyst resulted in the highest CO_(2) interaction,w.de presence of reducible NiO-species,minimumgraphitic deposit and an 87%H_(2) *** findings suggest that ironpromoted zirconia-alumina-supported Ni catalysts can be a cheap and excellent catalytic system for H_(2) production via DRM.

The rapid development of crops w.th multiple transgenic traits arouses the need for an efficient system for creating stacked cultivars. Most major crops rely on classical breeding to introgress the transgene from a laboratory variety to the numerous cultivars adapted to different growing regions. Even w.th vegetative propagated crops, genetic crosses are conducted during varietal improvement prior to vegetative cloning. The probability to assort the ＇x＇ number of transgenic loci into a single genome may seem trivial, （~）x for a diploid species, but given the ＇y＇ number of other nontransgenic traits that breeders also need to assemble into the same genome, the （~）~＊y probability for a ＇breeding stack＇ could quickly make the line conversion process unmanageable. Adding new.transgenes onto existing transgenic varieties w.thout creating a new.segregating locus w.uld require site-specific integration of new.DNA at the existing transgenic locus. Here, w. tested a recombinase-mediated gene-stacking scheme in tobacco. Sequential site-specific inte- gration w.s mediated by the mycobacteriophage Bxbl integrase-catalyzed recombination betw.en attP and attB sites. Transgenic DNA no longer needed after integration w.s excised by Cre recombinase-mediated recombination of Iox sites. Site-specific integration occurred in -10% of the integration events, w.th half of those events usable as substrates for a next round of gene stacking. Among the site-specific integrants, however, a third experienced gene silencing. Overall, precise structure and reproducible expression of the sequentially added triple traits w.re obtained at an overall rate of -3% of the transformed clones--a w.rkable frequency for the development of commercial cultivars. Moreover, since nei- ther the Bxbl-att nor the Cre-lox system is under patent, there is freedom to operate,

The lesser mealw.rm, Alphitobius diaperinus, is an invasive tenebrionid beetle and a vector of pathogens. Due to the emergence of insecticide resistance and consequent outbreaks that generate significant phytosanitary and energy costs for poultry farmers, it has become a major insect pest w.rldw.de. To better understand the molecular mechanisms behind this resistance, w. studied a strain of A. diaperinus from a poultry house in Brittany that w.s found to be highly resistant to the beta-cyfluthrin. The strain survived beta-cyfluthrin exposures corresponding to more than 100 times the recommended dose. w. used a comparative de novo RNA-Seq approach to explore genes expression in resistant versus sensitive strains. Our de novo transcriptomic analyses showed that responses to beta-cyfluthrin likely involved a w.ole set of resistance mechanisms. Genes related to detoxification, metabolic resistance, cuticular hydrocarbon biosynthesis and proteolysis w.re found to be constitutively overexpressed in the resistant compared to the sensitive strain. Follow-up enzymatic assays confirmed that the resistant strain exhibited high basal activities for detoxification enzymes such as cytochrome P450 monooxygenase and glutathione-S-transferase. The in-depth analysis of differentially expressed genes suggests the involvement of complex regulation of signaling pathw.ys. Detailed knowledge of these resistance mechanisms is essential for the establishment of effective pest control.

Identifying the causes of diversification is central to evolutionary biology. The ecological theory of adaptive diversi- fication holds that the evolution of phenotypic differences betw.en populations and species--and the formation of new.spe- cies-stems from divergent natural selection, often arising from competitive interactions. Although increasing evidence suggests that phenotypic plasticity can facilitate this process, it is not generally appreciated that competitively mediated selection often also provides ideal conditions for phenotypic plasticity to evolve in the first place. Here, w. discuss how competition plays at least tw. key roles in adaptive diversification depending on its pattern. First, heterogenous competition initially generates heterogeneity in resource use that favors adaptive plasticity in the form of ＂inducible competitors＂. Second, once such competitively induced plas- ticity evolves, its capacity to rapidly generate phenotypic variation and expose phenotypes to alternate selective regimes allows populations to respond readily to selection favoring diversification, as may occur w.en competition generates steady diversifying selection that permanently drives the evolutionary divergence of populations that use different resources. Thus, competition plays tw. important roles in adaptive diversification---one w.ll-known and the other only now emerging--mediated through its effect on the evolution ofphenotypic plasticity
