J. curcas and J. macrocarpa are useful for restoring degraded areas and their seeds contain oils for b.odiesel production. The aim of the work was to determine the reserve sub.tances in the endosperm and the emb.yo of J. curcas and J. macrocarpa which is important in understanding the germination process, the estab.ishment of these species and its industrial employment. Seeds were imb.b.d in distilled water for 24 h, to facilitate removal of seed coat with the aim to separate the emb.yo and nutritive tissues. In b.th species, the endosperm contained aleurone grains consisting of a crystalloid and glob.id, lipids of red color and the starch was not ob.erved. Four major fatty acids were determined in J. curcas seed: oleic, palmitic, stearic, palmitoleic and oleic fatty acid represents ab.ut 70% oil content. Oleic acid was the most ab.ndant in J. macrocarpa seeds, while, there was not palmitoleic acid. Seed with predominantly unsaturated fatty acids is ideal for b.odiesel industry. The means of the sugar content were: 14.3 μg/mg in endosperm and 104.76 μg/mg in emb.yo of J. curcas and 6.48 μg/mg in endosperm and 59.20 μg/mg in emb.yo of J. macrocarpa. The means of the protein content were: 4.2 μg/mg in endosperm and 45.02 μg/mg in emb.yo of J. curcas and 3.26 μg/mg in endosperm and 31.08 μg/mg in emb.yo of J. macrocarpa. Sugar and protein contents of Jatropha seeds were significantly higher in emb.yo in b.th species (p 0.1), which suggests early mob.lization towards the emb.yo during imb.b.tion period.

蛋壳颜色作为家禽的重要经济性状,在家禽养殖业中受到广泛的关注。目前,鸡绿色蛋壳的遗传基础已经发现。然而,对于鸭而言,绿色蛋壳颜色的遗传机制并未发现。在这项研究中,我们利用高通量测序技术对莆田白鸭白色蛋壳,莆田黑鸭白色蛋壳和莆田黑鸭绿色蛋壳的蛋壳腺进行测序,寻找绿壳蛋颜色形成的miRNA。我们获得了 108471471个高质量的读取,并将它们映射到鸭基因组。总共获得了 152520个已知的miRNAs和1526个新的miRNAs。为进一步分析差异表达miRNAs的功能,对miRNA靶基因进行KEGG通路分析。经由白色蛋壳和绿色蛋壳的比较,我们发现,miRNA-186-x和miRNA-497-x 均差异显著(Pb.和 SCP2 分别在VEGF信号通路,卟啉和叶绿素通路、钙信号通路和过氧化物酶体上调。推测这几个基因可能是调节绿蛋壳的形成。本研究为鸭蛋壳腺组织中mRNA和miRNA调控提供了有价值的信息,所鉴定的差异表达基因将为鸭蛋壳颜色的进一步遗传分析提供有价值的资源。

Implication of Electrostatic Forces on the Adsorption Capacity of a Modified b.ick for the Removal of Divalent Cations from Water

Adsorption properties of b.ick for the removal of divalent cations increased significantly after this material were pre-activated b. HCl and sub.equently impregnated with ferrihydrite. Scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDS) analysis demonstrated that ferrihydrite was preferentially attached to clays (mainly to metakaolinite) and possessed Na atoms at levels higher than those ob.erved in iron-poor aggregates. Sodium is b.und to hydroxyl groups which have a function as reactive sites and give rise to surface charge. Zeta potential measurements were conducted to determine the isoelectric point (IEP) and salt-addition method was used to assess the point of zero charge (PZC) of this b.ick. Modified b.ick has a positive charge in water up to pH ≈ 3.2 and negative charge ab.ve this pH. Moreover, pH was found to b. the most important factor affecting the adsorption process, suggesting the possib.e implication of electrostatic forces at the b.ick-water interface. The complexation model proposed b. James and Healy was applied to our system: theoretical data on free-energy changes due to effects associated b.th with electrostatic attraction and solvation, were found to b. in agreement with those determined from kinetic experiments. Column experiments permitted further to show that adsorption reactions were strongly inhib.ted b. addition of an inert electrolyte (like NaNO3). Under this condition, ionic strength increased and most surface sites of the b.ick would b. occupied b. Na+ ions, leading to a charge neutralization and thereb. a depletion of electrostatic forces.

Stem and immune cells in colorectal primary tumour:Numb.r and function of sub.ets may diagnose metastasis

An important percentage of colorectal cancer （CRC） patients will develop metastasis, mainly in the liver, even after a successful curative resection. This leads to a very high mortality rate if metastasis is not detected early on. Disseminated cancer cells develop from metastatic stem cells （MetSCs）. Recent knowledge has accumulated ab.ut these cells particularly in CRC, so they may now b. tracked from the removed primary tumour. This approach could b. especially important in prognosis of metastasis b.cause it is b.coming clear that metastasis does not particularly rely on testab.e driver mutations. Among the many traits supporting an epigenetic amplifcation of cell survival and self-renewal mechanisms of MetSCs, the role of many immune cell populations present in tumour tissues is b.coming clear. The amount of tumour-infiltrating lymphocytes （T, b.and natural killer cells）, dendritic cells and some regulatory populations have already shown prognostic value or to b. correlated with disease-free survival time, mainly in immunohistochemistry studies of unique cell populations. Parallel analyses of these immune cell populations together with MetSCs in the primary tumour of patients, with later follow-up data of the patients, will define the usefulness of specific comb.nations of b.th immune and MetSCs cell populations. It is expected that these comb.nations, together to different b.omarkers in the form of an immune score, may predict future tumour recurrences, metastases and/or mortality in CRC. It will also support the future design of improved immunotherapeutic approaches against metastasis.

Immunomodulation b. mesenchymal stem cells:Interplay b.tween mesenchymal stem cells and regulatory lymphocytes

Mesenchymal stem cells(MSCs) possess immunomodulatory properties, which confer enormous potential for clinical application. Considerab.e evidence revealed their efficacy on various animal models of autoimmune diseases, such as multiple sclerosis, systemic lupus erythematosus and uveitis. MSCs elicit their immunomodulatory effects b. inhib.ting lymphocyte activation and proliferation, forb.dding the secretion of proinflammatory cytokines, limiting the function of antigen presenting cells, and inducing regulatory T(Treg) and b.b.eg) cells. The induction of Treg and b.eg cells is of particular interest since Treg and b.eg cells have significant roles in maintaining immune tolerance. Several mechanisms have b.en proposed regarding to the MSCs-mediated induction of Treg and b.eg cells. Accordingly, MSCs induce regulatory lymphocytes through secretion of multiple pleiotropic cytokines, cell-to-cell contact with target cells and modulation of antigen-presenting cells. Here, we summarized how MSCs induce Treg and b.eg cells to provoke immunosuppression.

Activity optimization method in SPECT:A comparison with ROC analysis

A discriminant method for optimizing activity in nuclear medicine studies is validated b. comparison with ROC （received operating characteristic）-curves. The method is tested in 21 single photon emission computerized tomography （SPECT）, performed with a cardiac phantom. Three different lesions （Lb.L2 and L3） were placed in the myocardium-wall b. pairs fb. each SPECT. Three activities （84, 37 or 18.5 Mb.） of 99mTc were used as b.ckground. Linear discriminant analysis was used to select the parameters that characterize image quality among the measured variab.es in the images [（b.ckground-to-Lesion （b.Li） and Signal-to-Noise （S/N） ratios）]. Two clusters with different image quality （P=0.021 ） were ob.ained. The ratios b.Lj, b.L2 and b.L3 are the parameters used to construct the function with 100% of cases correctly classified into the clusters. The value of 37 Mb. was the lowest tested activity for which good results for the b.Li ratios were ob.ained. The result coincides with the applied ROC-analysis （r=0.89）.
