Optical tracking of picosecond coherent phonon pulse focusing inside a sub-micron object

By means of an ultrafast optical technique,we track focused gigahertz coherent phonon pulses in objects down to sub-micron in *** light pulses illuminating the surface of a single metal-coated silica fibre generate longitudinal-phonon wave *** of visible probe light pulses from the fibre surface allows the vibrational modes of the fibre to be detected,and Brillouin optical scattering of partially transmitted light pulses allows the acoustic wavefronts inside the transparent fibre to be continuously *** thereby probe acoustic focusing in the time domain resulting from generation at the curved fibre *** analytical model,supported by three-dimensional simulations,suggests that we have followed the focusing of the acoustic beam down to a~150-nm diameter waist inside the *** work significantly narrows the lateral resolution for focusing of picosecond acoustic pulses,normally limited by the diffraction limit of focused optical pulses to~1μm,and thereby opens up a new range of possibilities including nanoscale acoustic microscopy and nanoscale computed tomography.

Fulminant hepatic failure in a case of autoimmune hepatitis in hepatitis C during peg-interferon-alpha 2b plus ribavirin treatment

A 27-year-old Caucasian female with hepatitis C virus (HCV) infection treated with interferon (IFN) who developed severe autoimmune hepatitis (AIH) is described. The infecting viral strain was of genotype Ib and the pre-treatment HCV viral load was at a high level. The patient was treated with pegylated IFN-alpha 2b and ribavirin,and her HCV-RNA became negative at wk 12,but after that she developed fulminant hepatic failure. The patient recovered after steroid pulse therapy consisting of methylprednisolone 1000 mg/d for three days which was administered twice. A needle liver biopsy revealed the typical pathological findings of AIH.

In-silico clearing approach for deep refractive index tomography by partial reconstruction and wave-backpropagation

Refractive index(RI)is considered to be a fundamental physical and biophysical parameter in biological imaging,as it governs light-matter interactions and light propagation while reflecting cellular *** tomography enables volumetric visualization of RI distribution,allowing biologically relevant analysis of a ***,multiple scattering(MS)and sample-induced aberration(SIA)caused by the inhomogeneity in RI distribution of a thick sample make its visualization *** paper proposes a deep RI tomographic approach to overcome MS and SIA and allow the enhanced reconstruction of thick samples compared to that enabled by conventional linear-model-based RI *** proposed approach consists of partial RI reconstruction using multiple holograms acquired with angular diversity and their backpropagation using the reconstructed partial RI map,which unambiguously reconstructs the next partial *** this operation efficiently reconstructs the entire RI tomogram while suppressing MS and *** visualized a multicellular spheroid of diameter 140μm within minutes of reconstruction,thereby demonstrating the enhanced deep visualization capability and computational efficiency of the proposed method compared to those of conventional RI ***,we quantified the high-RI structures and morphological changes inside multicellular spheroids,indicating that the proposed method can retrieve biologically relevant information from the RI *** from the excellent biological interpretability of RI distributions,the label-free deep visualization capability of the proposed method facilitates a noninvasive understanding of the architecture and time-course morphological changes of thick multicellular specimens.

Efficacy of MK615 for the treatment of patients with liver disorders

AIM： To investigate the hepatoprotective effect of MK615, a Japanese apricot extract, in an animal model, and its clinical therapeutic effect. METHODS： Wistar rats were administered physiologi- cal saline （4 mL/kg） or MK615 solution （4 mL/kg） for 7 d. On the sixth d, acute hepatic injury was induced by administering a single intraperitoneal injection （ip） of D-galactosamine hydrochloride （D-GaIN）（600 mg/kg）. Plasma levels of alanine aminotransferase （ALT） and aspartate aminotransferase （AST） were determined, and liver tissues were used for histopathological analy- sis. Fifty-eight patients with liver disorders [hepatitis C （n = 40）, non-alcoholic fatty liver disease （n = 15）, and autoimmune liver disease （n = 3）] were orally admin- istered commercially available Misatol ME-containing MK615 （13 g/d） daily for 12 wk. Blood and urine were sampled immediately before and 6 wk, 12 wk, and 16 wk after the start of intake to measure various bio- chemical parameters. The percentage change in ALT and AST levels after 12 wk from the pre-intake baseline served as a primary endpoint. RESULTS： D-GaIN effectively induced acute hepatic injury in the rats. At 48 h after the ip injection of D-GaIN, the plasma levels of ALT （475.6 ：t： 191.5 IU/L vs 225.3 ＋ 194.2 IU/L, P 〈 0.05） and AST （1253.9：1：223.4 IU/L vs 621.9 ＋ 478.2 IU/L, P 〈 0.05） in the MK615 group were significantly lower than the control group. Scattered single cell necrosis, loss of hepatocytes, and extensive inflammatory cell infiltration were observed in hepatic tissue samples collected from the control group. However, these findings were less pronounced in the group receiving MK615. At the end of the clinical study, serum ALT and AST levels were significantly de- creased compared with pre-intake baseline levels from 103.5 ：l： 58.8 IU/L to 71.8 ＋ 39.3 IU/L （P 〈 0.05） and from 93.5 ：E 55.6 IU/L to 65.5 ＋ 34.8 IU/L （P 〈 0.05）, respectively. Sustained viral response of a case of acute hepatitis C virus infection via needle-stick injury

A 29-year-old nurse with a hepatitis C virus （HCV） infection caused by needle-stick injury was treated with interferon-beta starting about one year after the onset of acute hepatitis. The patient developed acute hepatitis C with symptoms of general fatigues, jaundice, and ascites 4 wk after the needle-stick injury. When these symptoms were presented, the patient was pregnant by artificial insemination. She hoped to continue her pregnancy. After delivery, biochemical liver enzyme returned to normal levels. Nevertheless, HCV RNA was positive and the pathological finding indicated a progression to chronicity. The genotype was lb with low viral load. Daily intravenous injection of interferon-beta at the dosage of six million units was started and continued for eight weeks. HCV was eradicated without severe adverse effects. In acute hepatitis C, delaying therapy is considered to reduce the efficacy but interferon-beta therapy is one of the useful treatments for hepatitis C infection in chronic phase.

Effects of estradiol-17β and bisphenol A administered chronically to mice throughout pregnancy and lactation on the male pups＇ reproductive system

Aim： To assess the effect of estradiol-17β （E2） and bisphenol A （BPA） administered chronically by implanting a silicone tube throughout pregnancy and lactation on male pups＇ reproductive system in ICR mice. Methods： Female mice were implanted with a tube filled with 10 ng, 500 ng, 1 μg, or 10 μg of E2, or 100 μg or 5 mg of BPA, before mating. The tube was kept in the mice throughout pregnancy and lactation, until the pups had weaned at 4 weeks of age. During the period, E2 was released from the tube at 120 pg or 6, 12 or 120 ng/day, and BPA at 1.2 or 60 μg/day. Results： Most of the mice given 1 μg and 10 lag of E2 did not maintain their pregnancy. However, the other groups showed high rates of birth, more than 70%. At age of 4 weeks, the male pups were killed. Body weight and reproductive organ weights （testes, epididymides and accessory reproductive glands） in the treated groups did not differ from the control values, whereas the percentage of seminiferous tubules in the testis with mature spermatids was significantly lower in the groups given 10 ng and 500 ng of E2 and 5 mg of BPA than that in the control. Conclusion： Chronic exposure to E2 and BPA might disrupt spermatogenesis in male pups. （Asian JAndrol 2008 Mar; 10： 271-276）

Expulsion of geopressured hydrothermal system along active faults and its relation to the occurrence of earthquakes in the Shinanogawa seismic belt, Japan

The 1995 Northern Niigata Earthquake (M 6.0) occurred at a shallow depth in the Niigata seismic gap. The anomaly areas in temperature, electrical conductivity and Cl- concentration of groundwater trend northeast as linear distribution in the epicentral area and are approximately coincident with the area of the seismic intensity 6 (JMA scale). The distributions of seismic intensity 6 and groundwater anomalies convincingly imaged the presence of a buried active fault beneath the epicentral area. The occurrence of this earthquake and the anomalies of groundwater were related to the expulsion of geopressured hydrothermal system (GHS). All epicenters of the destructive earthquakes along the Shinanogawa seismic belt are actually located in the buried active fault zones characterized by the areas of temperature and geochemical anomalies of groundwater. These earthquakes might have been triggered by the activity of GHS. The expulsion of GHS along an active fault in combination with the thermal softening of fault zone attributed to high rock temperature may reduce fracture strength of the rock, and trigger earthquake occurrence. The dimension of the anomaly area of groundwater temperature at the epicentral area reflected the scale of the earthquake fault. The linear anomaly areas of groundwater resulting from the expulsion of GHS and having no historical destructive earthquake are of the predicated areas of future destructive earthquakes. Monitoring of groundwater conditions in these areas may provide useful information regarding the future occurrence of earthquakes.
