新疆古尔班通古特沙漠链霉菌:从可培养资源多样性到抗菌天然产物的生物合成潜力探索

全球抗生素耐药性和新型抗菌药物短缺问题日益严峻,因此迫切需要发现新的或更有效的抗生素.沙漠生境蕴藏着丰富的链霉菌资源,是新型活性天然产物的重要潜在来源,但其在很大程度上仍未得到充分探索.本研究旨在深入挖掘新疆古尔班通古特沙漠中的可培养链霉菌资源,并探究其抗菌天然产物生物合成潜力.本文利用8种培养基对4份沙漠样品中的链霉菌进行分离、纯化和鉴定,随后对代表性链霉菌进行培养特征观察、次级代谢产物生物合成基因簇(biosynthetic gene cluster, BGC)预测和抗菌活性评价.研究结果显示,共分离得到469株链霉菌,占放线菌总菌株数的44.0%.从64株代表性链霉菌基因组中共预测得到1937个BGCs,分属于1068个基因簇家族(gene cluster family, GCF),其中97.5%的GCFs表现出较高的新颖性和次级代谢功能谱系多样性.此外,通过抗菌活性筛选,发现41株代表性链霉菌对至少一种临床病原菌具有拮抗作用,且其中2株具有广谱抗菌活性.这些发现表明,新疆古尔班通古特沙漠中蕴藏着极为丰富多样的可培养链霉菌资源,且绝大多数具有巨大的次级代谢产物生物合成潜力和较好的抗菌活性,为新型抗生素药物先导化合物的开发提供了重要依据和优良菌种来源.

【背景】植物内生菌作为微生物群落中一类非常重要的组成部分,在多个领域都有广泛的应用价值,如促进植物生长、抵抗病虫害、生物固氮、降解有毒有害化合物等。【目的】进一步丰富新疆野果林苹果腐烂病害的生防资源以及分析牛至内生细菌多样性特征。【方法】通过纯培养方法,对健康植物牛至组织进行内生细菌的分离纯化,并进行16S rRNA基因序列分析;再利用平板对峙法筛选具有抑制苹果腐烂病病原菌生长的内生细菌。【结果】共分离到168株内生细菌,最终确定为4门5纲8目12科17属,其中优势属为芽胞杆菌属(Bacillus)。由分离实验结果可知,M1号TSA培养基,M5号组氨酸-棉子糖培养基和M6号NA培养基是分离牛至内生细菌较为理想的培养基;采自新源县野果林的牛至内生细菌多样性较为丰富,在牛至根部内生细菌种类更多;通过拮抗实验共筛选出59株牛至内生细菌具有较好的抑菌效果。【结论】新疆药用植物牛至内生细菌的物种多样性较为丰富,而且富含一批有效抑制野苹果腐烂病病原菌生长的内生细菌菌株。因此,本研究在新疆野果林苹果腐烂病的生物防治和药用植物内生细菌种质资源的填补等方面具有重要意义。

Gold and Base Metals Associated with Uranium Mineralization in Some Lower Carboniferous Rock Units in the Wadi Abu El Mogheirat Area, Southwestern Sinai, Egypt

The Lower Carboniferous sediments at Wadi Abu El Mogheirat consist of four stratigraphic formations, from base upwards: Um Bogma, El Hashash, Magharet El Maiah and Abu Zarab formations. The most-concentrated radioactive anomalies at Wadi Abu El Mogheirat are in the middle member of the Um Bogma Formation. The gibbsitic marl in this formation shows the highest uranium contents(around 710 ppm). Gibbsitic marl and black gibbsite of the middle Um Bogma Formation show higher enrichment in Co, Cu, Mn, Ni, Pb, Zn, U, V, Ce and Th. Black gibbsite contains the highest total rare earth elements concentration, especially in terms of LREEs, while gibbsitic marl contains the highest content of HREEs. Anomalous contents of gold at Wadi Abu El Mogheirat are recorded for the first time in the gibbsitic marl(content: 10.4 ppm), black gibbsite(2.8 ppm) of the middle member of the Um Bogma Formation and in the siltstones of the El Hashash Formation(0.6 ppm). Gibbsitic marl shows the presence of uranothorite, celestite, zircon, atacamite, barite, xenotime and rutile. These characteristic demonstrate that the middle member of the Um Bogma Formation constitutes a potential source for gold. This conclusion is reinforced by the potential to exploit other metals such as Co, Cu, Mn, Ni, Pb, Zn, U, V, Th and Ce as by-products.

Studies on the Immunomodulatory Effects of Lactoferrin in Rats Infected with E. coli

Eighty male albino rats of Westar strain (350 ± 10 g), 10 to 12 weeks old were divided into four groups. The groups treated are as following. The first control group (Gp. I) was given intraperitoneally in normal saline (1 mL). The second group (Gp. II) was orally infected with 3 × 1012 CFU of E. coli /Kg. BW. The third group (Gp. III) was infected with E. coli and treated with (0.5%) lactoferrin (LF) 72 hours before E. coli infection in filtered tap water for the duration of the experiment (21 days). The fourth group (Gp. IV) was administrated with LF only (0.5%) in drinking water. Two separate blood samples were collected from heart puncture at the end of 1st, and 3rd week post-treatment for immunological studies. The Leukogram in E. coli treated group was insignificant compared with the control group while lymphocytosis was clear compared with the infected group. The Role of Kappa and Lambda in Subclassification of B Cell Lymphoblastic Leukemia in Sudanese Patients Using Flow Cytometry

Background: B-cell Acute lymphoblastic leukemia (B-ALL) is a neoplasm of lymphoblasts which are of B-cell lineage typically composed of small to medium sized blast cells, moderately condensed to dispersed chromatin with scanty cytoplasm and inconspicuous nucleoli, involving the bone marrow and/or blood. Methods and materials: This is a prospective cross-sectional study in which 50 blood and/or bone marrow samples of newly diagnosed patients (B-ALL) were tested for immunophenotyping. All samples were prepared for surface and cytoplasmic markers including kappa and lambda human antibody for 10 minutes in dark place and then run by the Flow Cytometer. Results: 64% of the study populations were males and 36% were females. Cases were classified according to immunophenotype and the age into different subtypes and showed the following frequencies: Pro B-ALL (8%), early pre B-ALL (56%), common B-ALL (16%), Pre-B-ALL (14%) and Mature B-ALL (only 6%). Surface immunoglobulin was positive in 10% and negative in 90% of all patients, showing 100% positivity in mature B-ALL and totally negative in other subtypes. While cytoplasmic immunoglobulin was positive in 16% and negative in 84% of all patients and was positive in 100% of Pre-B-ALL and in 50% of mature B-ALL. Surface kappa was more expressed in mature B-ALL than lambda giving a ratio of 2:1, while cytoplasmic kappa:lambda was 6:1 in Pre-B-ALL. Conclusion: Kappa and lambda have important role in B-ALL classification which necessitates their presence in immunophenotyping of B-ALL.

Effects of Zero Tillage and Residue Retention on Soil Quality in the Mediterranean Region of Northern Syria

Zero-tillage (ZT) and crop residue retention on the soil surface—two components of Conservation Agriculture (CA)—have been identified as promising management practices for sustainable agri- cultural intensification for some time. However, CA technology uptake by farmers in the dry areas of West Asia and North Africa (WANA) has yet to happen large-scale, even though the positive im- pact on yield has been demonstrated repeatedly. To explain the observed consistent increases in crop yield under ZT, a range of soil quality indicators were regularly monitored during 2008 to 2012 in the long-term ZT/CA trials at the headquarters of the International Center for Agricultural Research in the Dry Areas (ICARDA) in northern Syria. Results showed that CA had a positive im- pact on soil fertility. This was measurable by higher soil organic matter (SOM) and microbial bio- mass contents, increased levels of extractable phosphate, sometimes (but not always) higher amounts of larger water-stable soil aggregates, increased soil infiltration capacity and soil water retention. The buildup of SOM and associated carbon (C) sequestration was in the range of 0.29 Mg C/ha/yr, i.e. rather modest. High amounts of surface residues delayed the desiccation of the topsoil during the fallow period, but could not diminish the overall longer-term drying of the topsoil. The observed positive changes in soil quality were little, but nevertheless, in combination with the economic savings that ZT offers, this type of agricultural intensification provides an attractive op- tion for farmers in WANA, from the standpoints of economy and ecological efficiency.
