Bone marrow-derived mesenchymal stem cells ameliorate sodium nitrite-induced hypoxic brain injury in a rat model

Sodium nitrite（Na NO2） is an inorganic salt used broadly in chemical industry. Na NO2 is highly reactive with hemoglobin causing hypoxia. Mesenchymal stem cells（MSCs） are capable of differentiating into a variety of tissue specific cells and MSC therapy is a potential method for improving brain functions. This work aims to investigate the possible therapeutic role of bone marrow-derived MSCs against Na NO2 induced hypoxic brain injury. Rats were divided into control group（treated for 3 or 6 weeks）, hypoxic（HP） group（subcutaneous injection of 35 mg/kg Na NO2 for 3 weeks to induce hypoxic brain injury）, HP recovery groups N-2 w R and N-3 w R（treated with the same dose of Na NO2 for 2 and 3 weeks respectively, followed by 4-week or 3-week self-recovery respectively）, and MSCs treated groups N-2 w SC and N-3 w SC（treated with the same dose of Na NO2 for 2 and 3 weeks respectively, followed by one injection of 2 × 106 MSCs via the tail vein in combination with 4 week self-recovery or intravenous injection of Na NO2 for 1 week in combination with 3 week self-recovery）. The levels of neurotransmitters（norepinephrine, dopamine, serotonin）, energy substances（adenosine monophosphate, adenosine diphosphate, adenosine triphosphate）, and oxidative stress markers（malondialdehyde, nitric oxide, 8-hydroxy-2′-deoxyguanosine, glutathione reduced form, and oxidized glutathione） in the frontal cortex and midbrain were measured using high performance liquid chromatography. At the same time, hematoxylin-eosin staining was performed to observe the pathological change of the injured brain tissue. Compared with HP group, pathological change of brain tissue was milder, the levels of malondialdehyde, nitric oxide, oxidized glutathione, 8-hydroxy-2′-deoxyguanosine, norepinephrine, serotonin, glutathione reduced form, and adenosine triphosphate in the frontal cortex and midbrain were significantly decreased, and glutathione reduced form/oxidized glutathione and adenosine monophosphate/adenosine triphosphate ratio were significantly increased

Protective effects of Balanites aegyptiaca extract, melatonin and ursodeoxycholic acid against hepatotoxicity induced by methotrexate in male rats

Objective: To compare the degree of ameliorative effects of Melatonin(MEL), Ursodeoxycholic acid(UDCA) and Balanites aegyptiaca(BA) against hepatotoxicity induced by MTX for one month. Methods: Eighty adult male rats(Sprague Dawely) weighing(190±10g), were randomly divided into eight equal groups: Control, MTX, MEL, BA, UDCA, MTX+MEL, MTX+BA, MTX+UDCA. Liver function biomarker enzymes, liver tissue oxidative stress parameters, together with total antioxidant capacity and tumor necrosis factor(TNF-α) were determined. Histopathological and immunohistochemistry examinations for TNF-α were also done. Results: MTX showed significant increase in alanine transaminase(ALT), aspartate transaminase(AST), alkaline phosphatase(ALP), gamma glutamyl transferase(GGT), total and direct bilirubin, as well as TNF-α levels, oxidized glutathione(GSSG), malodialdehyde(MDA) and nitric oxide(NO). whereas, total protein, albumin, total antioxidant capacity, reduced glutathione(GSH), glutathione peroxidase(GPx), glutathione reductase(GR), glutathione S-transferase(GST), superoxide dismutase(SOD) and catalase(CAT) levels were significantly decreased in MTX treated group. These alterations were improved by MEL and BA treatment, whereas no improvement was noticed in UDCA treatment. Conclusions: BA may be as promising as MEL in the hepatoprotection against MTX toxicity through their antioxidant and radical scavenging activities. In addition, it is not recommended to co-administer UDCA with MTX as it enhanced inflammation and damage to the liver.

Tensile Strain Capacity Prediction of Engineered Cementitious Composites (ECC) Using Soft Computing Techniques

Plain concrete is strong in compression but brittle in tension,having a low tensile strain capacity that can significantly degrade the long-term performance of concrete structures,even when steel reinforcing is present.In order to address these challenges,short polymer fibers are randomly dispersed in a cement-based matrix to forma highly ductile engineered cementitious composite(ECC).Thismaterial exhibits high ductility under tensile forces,with its tensile strain being several hundred times greater than conventional concrete.Since concrete is inherently weak in tension,the tensile strain capacity(TSC)has become one of the most extensively researched properties.As a result,developing a model to predict the TSC of the ECC and to optimize the mixture proportions becomes challenging.Meanwhile,the effort required for laboratory trial batches to determine the TSC is reduced.To achieve the research objectives,five distinct models,artificial neural network(ANN),nonlinear model(NLR),linear relationship model(LR),multi-logistic model(MLR),and M5P-tree model(M5P),are investigated and employed to predict the TSCof ECCmixtures containing fly ash.Data from115 mixtures are gathered and analyzed to develop a new model.The input variables include mixture proportions,fiber length and diameter,and the time required for curing the various mixtures.The model’s effectiveness is evaluated and verified based on statistical parameters such as R2,mean absolute error(MAE),scatter index(SI),root mean squared error(RMSE),and objective function(OBJ)value.Consequently,the ANN model outperforms the others in predicting the TSC of the ECC,with RMSE,MAE,OBJ,SI,and R2 values of 0.42%,0.3%,0.33%,0.135%,and 0.98,respectively.

Effect of Exchange-Correlation Functional on the Structural, Mechanical, and Optoelectronic Properties of Orthorhombic RbSrBr3 Perovskite

In the present study, the effect of the exchange-correlation functional on the structural, mechanical, and optoelectronic properties of orthorhombic RbSrBr3 perovskite has been investigated using various functionals in Density Functional Theory (DFT) with the CASTEP code. The optimized lattice parameters are quite similar for all the functionals. The electronic properties have shown that RbSrBr3 perovskite is a wide direct band gap compound with a band gap energy ranging from 4.296 eV to 4.494 eV for all the functionals. The mechanical parameters like elastic constants, Young’s modulus, Shear modulus, Poisson’s ratio, Pugh’s ratio, and an anisotropic factor reveal that the RbSrBr3 perovskite has ductile behavior and an anisotropic nature which signifies the mechanical stability of the compound. The Debye temperature might withstand lattice vibration heat. High absorption coefficient (>104 cm−1), high optical conductivity, and very low reflectivity have been found in the RbSrBr3 perovskite for all functions. The computed findings on the RbSrBr3 perovskite suggested that the presented studied material is potentially applicable for photodetector and optoelectronic devices.

Impact of drivers'attributes on children injury severities in traffic crashes

Every year,a substantial number of children sustain injuries and fatalities in motor vehicle crashes in Wyoming.Understanding the factors contributing to child injury is crucial for the development of appropriate mitigation measures that aid in alleviating the severity of such injuries.In this study,a hierarchical Bayesian binary logit regression model was developed to investigate the factors that contribute to children’s injuries resulting from crashes while accounting for possible intra-class correlation effects(those of unobserved factors common to children involved in the same crash).A strong correlation among crashes justified the use of the hierarchical Bayesian logit model.As per the modeling results,the children’s ages,safety restraint types,vehicle types,drivers’ages,alcohol/drug involvement,drivers’seat belt use habits,drivers’actions,manners of collision and environmental conditions contributed to child injury risk.The child’s age was found to be inversely related to the risk of injury.Similarly,among safety restraint types,rear-facing car seats and forward-facing car seats were found to reduce injury likelihoods in crashes.When it comes to the drivers’characteristics,the probability of incurring injuries among the child population increased in the presence of young,unbuckled and impaired drivers.Furthermore,improper driving actions,such as running off the road,raised the risk of incurring injuries to children.The findings of this study may be beneficial to authorities regarding developing and implementing road safety programs aimed at ameliorating child injury concerns.

An excimer ‘ON–OFF’ switch based on telomeric G-quadruplex and rGO for trace thrombin detection

In the recent decade,GO has emerged as an amazing 2D nanomaterial for developing DNA-based biosensors due to its fluorescence quenching properties,whereas similar research based on rGO was reported rarely.Herein,a novel multi-pyrene functionalized G-rich DNA probe based on the screened rGO showed much higher fluorescence quenching efficiency and excimer emission than that of universal GO.Different from the universal thrombin detection of the G4-forming aptamer-TBA(GGTTGGTGTGGTTGG),the original telomeric sequence is used in this study.The excimer emission“ON-OFF”switch amplified the response of thrombin detection is as low as 50 units.Furthermore,for four pyrene moieties that are sited in a crowded steric circumstance,the melting temperature(T_(m))values and molecular dynamics simulations showed a positive effect on duplex G-quadruplex or _(G)DNA·_(c)DNA stability,without disturbing its helix structure.

Mapping Structurally Controlled Alterations Sparked by Hydrothermal Activity in the Fatira–Abu Zawal Area,Eastern Desert,Egypt

The Eastern Desert of Egypt suffered a protracted period of deformation triggered by cratonization of the new juvenile crust known as the Arabian Nubian Shield(ANS),which has been proposed for potential gold discoveries associated with the corresponding tectonic event.The Fatira area,on the border of Egypt’s Northern and Central Eastern Deserts,is covered with metavolcanic rocks twisted by a dextral relocation of the Fatira Shear Zone(FSZ)relative to the Barud magmatic body.The recent study evaluated many deformed post-orogenic granitic intrusions and felsite dikes associated with promising mineralization localities,notably orogenic gold deposits.The combination of various field observations and remote sensing data,followed by the analysis of aeromagnetic enhanced maps,allowed the differentiation of distinct lithologies,structural features,and hydrothermal alterations in the study area.Additionally,the integrated results obtained from the different interpretation techniques are utilized to identify and confirm the previously supposed mineralized localities in the Fatira and Abu Zawal areas and predict other matched localities.The final ASTER,Sentinel 2 hydrothermal alteration,and orientation entropy heat maps demonstrate the association between these mineralized regions and major structures related to the FSZ late stage of deformation rather than other structures studied throughout the area of interest.

Modeling severities of motorcycle crashes using random parameters

One of the critical areas of road safety is motorcycle safety. Motorcyclists are more vulnerable to injuries than occupants of other motor vehicles when involved in crashes.Researchers have studied the relationships between motorcycle crash severity and crash contributing factors. They are crash characteristics, roadway geometric design features,traffic characteristics, socio-demographics and environmental conditions. However, few researchers considered unobserved heterogeneity effects when modeling motorcycle crash injury severities, let alone interaction effects. In this research, motorcycle crashes in Wyoming that occurred from 2008 to 2017 were analyzed. Specifically, the injury severities of single motorcycle crashes and multiple vehicle crashes involving motorcycles were modeled. The response was whether the motorcycle crash incurred an incapacitating injury or fatality or not. The binary logistic regression and mixed binary logistic regression modeling structures were implemented. The mixed models revealed effects that otherwise would have been undisclosed in the binary logistic regression models’ results. According to the results of single motorcycle crashes, the majority of motorcycle-animal crashes and of motorcycle-barrier crashes were likely to be severe relative to other single motorcycle crashes. It was also found that horizontal curves increased the risk of severe injuries.Young riders were found to be less at risk of being gravely injured in single motorcycle crashes than older riders as well. Furthermore, riding under the influence and high posted speed limits increased the odds of severe crashes regardless of whether the crashes were single motorcycle crashes or multiple vehicle crashes involving motorcycles. Additionally,the mixed models uncovered interaction effects and unobserved effects pertaining to speed limits.

Examining the Risk Factors of Rear-End Crashes at Signalized Intersections

Rear-end crashes are among the most common crash types at signalized intersections. To examine the risk factors for the occurrence of this crash type, this study involved the analysis of nine years of intersection crash records in the state of Wyoming. With that, the contributing factors related to crash, driver, environmental, and roadway characteristics, including pavement surface friction, were investigated. A binomial logistic regression modeling approach was applied to achieve the study’s objective. The results showed that three factors related to crash and driver’s attributes (commercial vehicle involvement, speeding, and driver’s age) and four factors related to environmental and roadway characteristics (lighting, weather conditions, area type, whether urban or rural and pavement friction) are associated with the risk of rear-end crash occurrence at signalized intersections. This study provides insights into the mitigation measures to implement concerning rear-end crashes at signalized intersections.

Synergistic renoprotective effect of a compiled branched-chain amino acids and Cymbopogon schoenanthus extract against experimentally induced oxido-nitrosative renal insult

Objective: To better investigate the protective role of branched-chain amino acids(BCAAs)and Cymbopogon schoenanthus(CS) extract against the potassium dichromate(PDC)-induced oxido-nitrosative nephrotoxic insult in the experimental rat model. Methods: Thirty male rats were randomly divided into five equal groups: The 1 st group served as control; the 2^(nd)was injected with a single dose of PDC(15 mg/kg b.w i.p.);the 3^(rd), 4^(th), and 5^(th) groups were respectively treated with BCAAs, CS, and their combination for 15 d prior to induction of renal insult via PDC single dose(15 mg/kg b.w s.c.). The experimental period was terminated in all groups 2 d after induction of renal insult. The harvested kdney samples were divided for biochemical assays and histological examination. Results: The PDC-induced nephrotoxic effect caused a depletion of renal oxidative scavengers glutathione, superoxide dismutase with consequent lipo-oxidative cellular membrane deterioration manifested by a rise in malonaldehyde, oxidized glutathione, myeloperoxidase and the concomitant increase in inflammatory response elements tumor necrosis factor α, nitric oxide, and interleukin 1 β.Moreover, the comet assay and increased 8-hydroxy-2-deoxyguanosine proved an accelerated apoptotic DNA fragmentation. These local renal changes were met with global altered blood biochemistry. The BCAAs and CS or their compiled administration showed an ameliorative effect against PDC-induced nephrotoxic in a synergistic pattern. Conclusions: Both BCAAs and CS or their combined administration afford potential competitors against renal insult induced by polyvalent anion pollutants in experimentally studied animals model. As a route for novel drug discovery, further investigation should be attempted to optimize their augmenting reno-protecting potential.
