Isolation, characterization and modulatory potentials of β-stigmasterol, ergosterol and xylopic acid from Anchomanes difformis on mitochondrial permeability transition pore in vitro

Objective: Secondary metabolites and polyphenolic compounds from medicinal plants have been demonstrated to have multiple biological functions with promising research and development prospects. This study examined the effect of β-stigmasterol(with ergosterol) and xylopic acid isolated from Anchomanes difformis on liver mitochondrial permeability transition pore(mPTP).Methods: The compounds were isolated by vacuum liquid chromatography. Mitochondrial swelling was assessed as changes in absorbance under succinate-energized ***:1H and13C NMR spectroscopic elucidation of the isolates affirmed the presence of β-stigmasterol with ergosterol(1:0.3) and xylopic acid. The isolates reversed the increase in lipid peroxidation and inhibited the opening of mitochondrial permeability transition pores caused by calcium and *** inhibition of m PTP offers a promising therapeutic target for the treatment of mitochondrial-associated ***: Reduction in the activity of calcium ATPase and the expression of Caspase-3 and-9 were observed, suggesting that they could play a role in protecting physicochemical properties of membrane bilayers from free radical-induced severe cellular damage and be useful in the management of diseases where much apoptosis occurs.

Chronic Exposure to No-Effect Concentration of Diazinon Induced Histological Lesions in Organs of <i>Clarias</i><i>gariepinus</i>

In all parts of the world pesticides have been found in the aquatic ecosystem and scientific evidence has also shown that they can enter the food chain. Diazinon is an organophosphate pesticide, widely used in agriculture to control a wide variety of suckling and leaf eating insects and recently in fish culture to suppress some parasitic diseases;nevertheless, there is little study on its adverse effect on fish. In this study, seventy-two (72) apparently healthy catfish comprising adult and juvenile of both sexes were used to set up triplicate experimental groups of those exposed to culture water alone (control group), fish exposed to pre-determined no-effect concentration (0.405 ppm) of Diazinon (test group). The fish were exposed for 28 days and fish were sacrificed and organs harvested on days 21 and 28 to determine the effect of long-term exposure to diazinon on organ histology. Histological changes observed in diazinon-exposed catfish were hyperplasia and fusion of the gill epithelium, hyperplasia of mucoid producing cells and aggregation of melanin pigment in the skin. Histological lesions were also seen observed in other organs, including severe diffuse cellular swelling and fatty degeneration of the liver, interstitial congestion of the kidney, carbon deposit on the wall of the heart and multifocal haemorrhage. The water quality of the control was not significantly different from that of the test group throughout the experiment. The lesions detected in cells, tissue, or organs represent an integration of cumulative effects of physiological and biochemical stressors. The histological alterations observed in vital organ of fish show that exposure to “no-effect” concentration of diazinon induced structural damage in fish organs and are likely to affect the functionality of the organs. For example, the adverse effect on the gill might disrupt its feeding and oxygen uptake.

目的:研究扁桃斑鸠菊及非洲印楝叶的提取物对链脲佐菌素致糖尿病大鼠肝脏形态学、肝脏氧化性应激标志物及部分肝脏酶类的影响。方法:大鼠腹腔注射链脲佐菌素致糖尿病。不同治疗组大鼠分别口服扁桃斑鸠菊及非洲印楝叶的提取物(500mg/kg)或二甲双胍(150mg/kg),疗程8周,每周测量大鼠的血糖水平及体质量变化。8周后麻醉处死大鼠。取肝组织制成切片,希夫染色法染色,并测量肝匀浆中丙二醛和谷胱甘肽过氧化物酶的含量;下腔静脉采血,分离血浆,检测血浆中丙氨酸氨基转移酶和天冬氨酸氨基转移酶的活性。结果:二甲双胍及植物提取物均能显著改善糖尿病大鼠的血糖水平,且与糖尿病模型组相比,治疗组的体质量明显增加(P0.05),而丙二醛含量明显下降(P<0.05)。结论:扁桃斑鸠菊及非洲印楝叶的提取物对糖尿病大鼠有明显的降糖作用,而肝脏形态学及肝毒性标志物没有明显变化。

Cultivation of Candida utilis on Cassava Peel Hydrolysates for Single-cell Protein Production

The growth ofCandida utilis NRRL Y-1084 in acid and enzymatic hydrolysates of cassava peel and on glucose in a mineral salts medium was investigated in aerobic submerged cultivation. Kinetic and stoichiometric parameters for growth were determined. The cardinal temperatures of this yeast strain were 14 ℃, 33 ℃ and 41 ℃. C. utilis exhibited no absolute requirement for growth factors, although its maximum specific growth rate （μmax） was higher in the mineral salts medium with yeast extract than without, but its biomass yield coefficient （Yx/s） did not differ much in these two media. In the enzymatic hydrolysate, its Yx/s value on sugar was 0.44 with a μmax of 0.35 h^-1, whereas the corresponding values were 0.52 and 0.48 h^-1 in the acid hydrolysate and 0.50 and 0.37 h^-1 in the mineral salts medium without yeast extract. The crude protein content of biomass grown in the glucose medium and the acid and enzymatic hydrolysates were 47.5%, 49.1% and 56.7%, respectively. The amino acid profile of the yeast biomass compared favourably with the FAO standard. Cassava peel hydrolysate has potential as a cheap carbohydrate feedstock for the production of yeast single cell protein by using C. utilis.

Hydro-Ethanol Extract of Persea americana Fruit Pulp Ameliorates Dyslipidaemia and Cardiotoxicity Exerted by Alloxan-Induced Diabetes Mellitus

Diabetes-induced dyslipidaemia has been associated with an increased risk of atherosclerosis and coronary heart diseases. Persea americana fruit has been reported to possess anti-diabetic properties. Therefore, this study assessed the lipid profile and likely cardio-protective effects of hydroethanolic extracts of P. americana fruits in alloxan-induced diabetic Wistar rats. Thirty-five male rats (150 ± 30 g) were divided into 5 groups (n = 7) and treated orally as follows;groups I-II were normal animals treated with distilled water (0.3 ml/day) and P. americana (300 mg/kg) only respectively. Animals in groups III-V were made diabetic using alloxan monohydrate (100 mg/kg i.p.) and treated orally with distilled water (0.3 ml/day), P. americana (300 mg/kg) and glibenclamide (5 mg/kg) respectively for 21 days. Fasting blood glucose level was monitored prior to, after induction of diabetes mellitus, and on day 21 post-treatment, respectively. Thereafter, retro-orbital blood samples were collected after anaesthesia and analysed for insulin, total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL), high density lipoprotein (HDL) levels, apolipoproteins A1 and B, superoxide dismutase (SOD) and catalase activities, reduced glutathione (GSH), Vitamin C and malondialdehyde levels, respectively. VLDL, atherogenic index (AI) and ApoB/A1 ratio were estimated mathematically. Pancreatic and cardiac structures were also investigated using Haematoxylin and Eosin stains. Treatment with P. ameriacana extracts reduced (p P. americana treated diabetic group. The hydro-ethanol fruit extract of Persea americana attenuates diabetes induced dyslipidaemia and reduces the susceptibility to cardiac impairment in experimental diabetes mellitus.

Amino acid quality indices of the leaves of Clerodendrum volubile

Objective:To evaluate the amino acid profile and quality indices of Clerodendrum volubile(***)***:Dried leaves of *** were blended,defatted and subjected to amino acid analysis using the technicon sequential multi-sample amino acid *** amino acid quality indices which covers for chemical score,essential amino acid index,nutritional index,true digestibility,protein digestibility corrected amino acid score,and digestible indispensable amino acid score were evaluated using standard ***:Amino acid analysis revealed glutamic acid to have the highest concentration,with cysteine having the *** acid had the highest chemical score,this was followed by glycine,histidine and arginine,*** least scores were observed in serine and *** acid had the highest value for true digestibility and protein digestibility corrected amino acid score,with the least observed in *** indispensable amino acid score evaluation showed histidine to have the highest value for infants(birth to 6 months),threonine for children(6 months to 3 years),while isoleucine was observed to have the highest value for older children,adolescents and *** essential amino acid index value was less than 4,while nutritional index value was less than ***:These results indicated the leaves of *** as a potential source of amino acids in the human diet as portrayed by its amino acids profile and qualities.

Genistein Mitigates the Gastro-Toxic Effects of Bisphenol A in Male Wistar Rats

Exposure to Bisphenol A (BPA) worldwide is on the increase. Its toxicities in various tissues expressing estrogen receptors have been reported. However, limited information exists on its effects on the gastric tissue. This study therefore investigated the gastric effects of BPA exposure and the likely ameliorative potential of genistein, a phytoestrogen antioxidant, known to interact with estrogen receptors. Eighty-four male Wistar rats were randomly divided into 6 groups (n = 14) and orally treated for 28 days. Group I-IV received distilled water (0.2 ml), corn oil (3 ml/kg), BPA (50 mg/kg) and genistein (50 mg/kg) respectively. Group V was pre-treated daily with BPA one hour prior to genistein administration while Group VI was pre-treated daily with genistein one hour prior to BPA treatment. Thereafter, blood was collected into heparinized bottles for hematological analysis. Gastric juice was collected for pH and electrolytes determination. Gastric tissue was excised and analyzed for superoxide dismutase (SOD), reduced glutathione (GSH), malondialdehyde, nitric oxide (NO), mucin, parietal and mucous cell counts, and histology. The BPA only treatment group exhibited significant increases (p < 0.05) in white blood cell count, neutrophil-lymphocyte ratio, mucin concentration, NO, and malondialdehyde while gastric juice pH, bicarbonate, SOD, and GSH levels were reduced compared to control. The gastric mucosa also showed pathologies consistent with inflammation. Genistein pre-and post-treatment in BPA exposed rats significantly (p < 0.05) ameliorated these effects of BPA. However, some signs of gastric inflammation were still evident in the mucosal samples. Bisphenol A induces gastro-toxicity by increasing gastric acidity, reducing gastric juice bicarbonate level and disrupting prooxidant/antioxidant balance. Genistein pre- and post-treatment ameliorated these gastro-toxic effects of Bisphenol A via gastroprotective, antioxidant and anti-inflammatory mechanisms.

通过对16组粉煤灰再生混凝土试件的试验,考虑了粉煤灰与再生骨料的影响,经多元回归计算建立了灰水比、再生骨料取代率、粉煤灰取代率与28 d抗压强度之间的关系,并探讨了灰水比、粗骨料压碎指标、粉煤灰取代率与抗压强度的关系。对普通混凝土的鲍罗米公式进行修正,得到了适用于粉煤灰再生混凝土配合比设计的鲍罗米公式。

在正交试验的基础上研究了在不同水胶比、再生骨料掺量、粉煤灰掺量条件下对再生混凝土的各项力学性能指标的影响,对试验结果进行了极差和方差分析,得出水胶比,再生骨料掺量和粉煤灰掺量对各指标的影响规律。在此试验基础上,引用了一些国内外再生混凝土的试验结果,采用回归分析的方法,分别建立了劈裂抗拉强度、抗折强度、弹性模量与立方体抗压强度的关系。
