proteoliposome and polysaccharide-Based Meningococcal Vaccine Are Immunogenic in Infants and Toddlers and primes for Memory against Serogroup C polysaccharide

Neisseria meningitidis capsular polysaccharides are the main target of the protective immune response against bacterial meningitis. They are thymus-independent type 2 (TI-2) antigens that are poorly immunogenic and not protective in young children, and their administration may impair subsequent challenge with the same polysaccharide. These problems have been addressed using three different vaccines consisting of 1) polysaccharide alone, 2) polysaccharide covalently conjugated to a carrier protein, and 3) polysaccharide with proteoliposome (pL) adsorbed onto Al(OH)3. VA-MENGOC-BC? is one of the third types of vaccine. It contains pL (detergent-extracted external membrane proteins forming vesicles) and polysaccharide (ps) from N. meningitidis serogroups B and C (psC), respectively. Nevertheless, there is a concern that to overcome the TI-2 nature of ps the covalently conjugation to a carrier is mandatory. Therefore, we evaluated the immune response induced by VA-MENGOC-BC? in infants and toddlers in order to determine whether it stimulates the response against the psC. High IgG anti psC and anti pL responses were seen following the administration of two doses in infants and toddlers, after a 3rd dose in pre-teenagers, and after confirmed carrier stages in young adults. The anti pL IgG response persisted longer than anti psC IgG response and IgM response against both antigens was maintained. An IgG1 anti pL response predominated, as well as IgG4 > IgG3 > IgG1 anti psC responses. These results suggest that non-covalent incorporation of psC onto Al(OH)3 containing?pL as adjuvant is immunogenic, primes for memory, and induces long-lasting specific antibody response. The improved psC immunogenicity of this vaccine may be due to the preferential and potent Th1 response induced in mice and human by the pL as adjuvant.

Introduction: Neisseria is a large genus of bacteria that colonize mucosal surfaces. Of the 11 species that colonize humans, only two are pathogens, N. meningitidis and N. gonorrhoeae. Both are obligate human pathogens;the last is causal agent of gonorrhea disease. Although curable with timely antibiotic treatment, an annual incidence of more than 62 million cases is estimated by the World Health Organization and there are no successful vaccines available. In contrast, several prophylactic vaccine options for Neisseria meningitidis meningitis do exist. Of note, there is trace of cross parenteral response induced between N. meningitidis and N. gonorrhoeae, and proteoliposome (pL, also named outer membrane vesicles, OMV) vaccine has shown high impact on this response. Objective: To determine effect of VAMENGOC-BC? and its derivates (AFpL1 and AFCo1) at mucosal and systemic level and possible cross response against Neisseria gonhorroeae in Balb/c and C57Bl/6 mice. Methods: We evaluated cross response against N. gonorrhoeae in mouse serum IgG and saliva IgA after mucosal immunization with VA-MENGOC-BC or its derivatives (AF, Adjuvant Finlay pL1 or AFCo (cochleate) 1). Results: Immunizations with AFpL1 or AFCo1 induce anti N. gonorrhoeae at saliva IgA and serum IgG responses similar to VA-MENGOC-BC? vaccine. Conclusions: Such data confirms a new possible window of prime-boost vaccination strategy against gonorrhea and extends our knowledge regarding the effect of pL vaccines on cross responses.

Dear Editor, plants secrete nectar to attract mutualistic animals, which predominantly function as pollinators, as in the case of flo- ral nectar, or defenders against herbivores, as in the case of extrafloral nectar （Nicolson et al., 2007）. Because nectars usu- ally represent aqueous solutions containing sugars and other nutrient metabolites （Baker and Baker, 1983）, they are sus- ceptible to infestation by microbial organisms, which can use the nectar-secreting tissues as entry sites to infect the plant. ON KARUSH-KUHN-TUCKER pOINTS FOR A SMOOTHING METHOD IN SEMI-INFINITE OpTIMIZATION

We study the smoothing method for the solution of generalized semi-infinite optimization problems from （O. Stein, G. Still： Solving semi-infinite optimization problems with interior point techniques, SIAM J. Control Optim., 42（2003）, pp. 769-788）. It is shown that Karush-Kuhn-Tucker points of the smoothed problems do not necessarily converge to a Karush-Kuhn-Tucker point of the original problem, as could be expected from results in （F. Facchinei, H. Jiang, L. Qi： A smoothing method for mathematical programs with equilibrium constraints, Math. program., 85（1999）, pp. 107-134）. Instead, they might merely converge to a Fritz John point. We give, however, different additional assumptions which guarantee convergence to Karush-Kuhn：Tucker points.

The macrophage migration inhibitory factor has been implicated in a number ofimmune and inflammatory processes. MIF presents particular opportunities for drug design anddevelopment with potential therapeutic applications. Drug design strategies taking intoconsideration of specific stereochemical and tautomeric requirements in the interaction of MIF withsubstrates and inhibitors allow several novel structures to be designed. Our investigationssuccessfully explored the tautomeric and stereochemical aspects of new compounds of the2-phenylpyruvic acid type, both experimentally, through synthesis and structural investigations andcomputationally, through molecular mechanics and quantum mechanics calculations.

In sexual reproduction different types of symbiotic relationships between insects and microbes have become established. For example, some bacteria have evolved almost exclusive vertical transmission and even define the compatibility of insect mating partners. Many strictly sexually transmitted diseases have also been described in insects. Apart from such rather specific relationships the role of opportunistic infections in the reproductive process has been widely neglected. Opportunistic microbes transmitted passively during mating might impose an energetic cost, as the immune system will need to be alert and will use resources to fight potential intruders. Through mating wounds and contaminated reproductive organs opportunistic microbes might be transferred to mating partners and even enter the body cavity. Females as the ＂receiving＂ sex are particularly likely to have evolved adaptations to avoid or reduce opportunistic infections. Males of several species show highly complex seminal fluids, which as well as containing components that influence a males＇ fertilization success, also possess antimicrobial substances. The role of antimicrobials in the reproductive process is not well understood. Some evidence hints at the protection of sperm against microbes, indicating a role for natural selection in shaping the evolution of reproductive traits. By highlighting the potential importance of microbes in sexual selection and their role in reproduction in general I will make a case for studies in sexual selection, especially the ones investigating postcopulatory processes, that should incorporate environmental, as well as genotypic variation, in reproductive traits.
