Transcutaneous perianal sonography:A sensitive method for the detection of perianal inflammatory lesions in Crohn's disease

AIM: Pelvic magnetic resonance imaging (MRI) and endoanalultrasound which are established imaging methods forperianal inflammatory lesions in patients with Crohn'sdisease require expensive specialized equipments andexpertise. We investigated the feasibility and sensitivity oftranscutaneous perianal ultrasound (PAUS) using regularultrasound probes in the imaging of perianal inflammatorylesions. The sonographic findings were correlated to pelvicMRI-scans.METHODS: We performed PAUS in 25 patients with Crohn'sdisease and clinical signs of perianal inflammatory disease.VCChin a median of 10 d (range 0-75) these patients underwentMRI of the pelvis. Regular convex and linear high resolutionprobes were used for PAUS. The sonographic findings werecorrelated to the MRI findings by blinded investigators.RESULTS: The sonographic investigations were welltolerated by all patients. Fistulae typically presented ashypoechoic tracks. Twenty-nine fistulae were detected in22 patients. Abscesses were detected in 7 patients andpresented as hypo- or anechoic formations. Twenty-six of29 fistulae and 6 of 7 abscesses could be confirmed byMRI. Kappa statistics showed an excellent agreement(kappa>0.83) between the two imaging methods.CONCLUSION: PAUS is a simple, painless, feasible, real-time method that can be performed without specific patientpreparation which is comparable in its sensitivity to pelvicMRI in the detection of perianal fistulae and/or abscesses.PAUS can especially be recommended as a screening toolin acute perianal disorders such as perianal abscess andfor follow-up studies of perianal inflammatory disease.

Eribulin is a non-taxane synthetic analogue approved in many countries as thirdline treatment for the treatment of patients with metastatic breast cancer.In addition to its mitotic property,eribulin has non-mitotic properties including but not limited to,its ability to induce phenotypic reversal of epithelial to mesenchymal transition,vascular remodelling,reduction in immunosuppressive tumour microenvironment.Since approval,there has been a surge in studies investigating the application of eribulin as an earlier-line treatment and also in combination with other agents such as immunotherapy and targeted therapy across all breast cancer sub-types,including hormone receptor positive,HER2 positive and triple negative breast cancer,many demonstrating promising activity.This review will focus on the application of eribulin in the treatment of metastatic breast cancer across all subtypes including its role as an earlier-line agent,its toxicity profile,and potential future directions.

Recycling of solid-state batteries—challenge and opportunity for a circular economy?

The tremendous efforts made in the research field of solid-state Li-ion batteries have led to considerable advancement of this technology and the first market-ready systems can be expected in the near future.The research community is currently investigating different solid-state electrolyte classes(e.***.oxides,sulfides,halides and polymers)with a focus on further optimizing the synthesis and electrochemical performance.However,so far,the development of sustainable recycling strategies allowing for an efficient backflow of critical elements contained in these batteries into the economic cycle and thus a transition from a linear to a circular economy lags behind.In this contribution,resource aspects with respect to the chemical value of crucial materials,which are used for the synthesis of solid-state electrolytes are being discussed.Furthermore,an overview of possible approaches in relation to their challenges and opportunities for the recycling of solid-state batteries with respect to different solid-state electrolyte classes by means of pyrometallurgy,hydrometallurgy and direct recycling/dissolution-based separation processes is given.Based on these considerations and with reference to previous research,it will be shown that different solid-state electrolytes will require individually adapted recycling processes to be suitably designed for a circular economy and that further improvements and investigations will be required.

This editorial addresses catatonia,a complex neuropsychiatric syndrome characterised by a spectrum of psychomotor disturbances.The editorial seeks to clarify the ambiguous aspects of catatonia,integrating recent research findings,including global studies and diagnostic advancements.It discusses catatonia’s clinical manifestations,prevalence,and associated psychiatric and medical conditions,with particular emphasis on its frequent co-occurrence with schizophrenia and mood disorders.The prevalence of catatonia,which varies across psychiatric populations,is illustrated by a significant study conducted in Nelson Mandela Bay,South Africa.This study provides valuable insights into the effectiveness of the Bush-Francis Screening Instrument compared to the Diagnostic and Statistical Manual 5 criteria in diagnosing catatonia.The editorial evaluates treatment approaches,primarily focusing on benzodiazepines and electroconvulsive therapy,and discusses emerging therapeutic strategies.It underscores the importance of robust diagnostic frameworks and early intervention in managing catatonia,as recommended by the latest evidence-based consensus guideline.Furthermore,it suggests future research directions,particularly in exploring the neurobiological and genetic factors of catatonia,to enhance our understanding and improve treatment outcomes.This editorial succinctly aims to demystify catatonia and provide valuable insights for clinicians and researchers in mental health care.

The metabolite lactate (L-lactate) can be generated and released by diverse brain cells,including neurons,astrocytes,and oligodendrocytes (Kann,2023;Rae et al.,2024).Lactate production usually requires the degradation of glucose (D-glucose)-and glycogen in astrocytes-to pyruvate by glycolysis and subsequent conversion of pyruvate to lactate by the enzyme lactate dehydrogenase(Figure 1A;Dienel,2019;Rae et al.,2024).

Explainable artificial intelligence models for mineral prospectivity mapping

Mineral prospectivity mapping(MPM) is designed to reduce the exploration search space by combining and analyzing geological prospecting big data. Such geological big data are too large and complex for humans to effectively handle and interpret. Artificial intelligence(AI) algorithms, which are powerful tools for mining nonlinear mineralization patterns in big data obtained from mineral exploration, have demonstrated excellent performance in MPM. However, AI-driven MPM faces several challenges, including difficult interpretability, poor generalizability, and physical inconsistencies. In this study, based on previous studies, we devised a novel workflow that aims to constructing more transparent and explainable artificial intelligence(XAI) models for MPM by embedding domain knowledge throughout the AI-driven MPM, from input data to model design and model output. This newly proposed approach provides strong geological and conceptual leads that guide the entire AI-driven MPM model training process, thereby improving model interpretability and performance. Overall, the development of XAI models for MPM is capable of embedding prior and expert knowledge throughout the modeling process, presenting a valuable and promising area for future research designed to improve MPM.

Tensorial tomographic Fourier ptychography with applications to muscle tissue imaging

We report tensorial tomographic Fourier ptychography(T^(2)oFu),a nonscanning label-free tomographic microscopy method for simultaneous imaging of quantitative phase and anisotropic specimen information in 3D.Built upon Fourier ptychography,a quantitative phase imaging technique,T^(2)oFu additionally highlights the vectorial nature of light.The imaging setup consists of a standard microscope equipped with an LED matrix,a polarization generator,and a polarization-sensitive camera.Permittivity tensors of anisotropic samples are computationally recovered from polarized intensity measurements across three dimensions.We demonstrate T^(2)oFu’s efficiency through volumetric reconstructions of refractive index,birefringence,and orientation for various validation samples,as well as tissue samples from muscle fibers and diseased heart tissue.Our reconstructions of healthy muscle fibers reveal their 3D fine-filament structures with consistent orientations.Additionally,we demonstrate reconstructions of a heart tissue sample that carries important polarization information for detecting cardiac amyloidosis.
