Whole-genome sequences provide massive amounts of data that can illuminate many aspects of a species’biology and evolutionary history,as well as valuable information for conservation *** is particularly important for large carnivores,whose large area requirements and typical low densities make them prone to undergo rapid genetic erosion(*** of runs of homozygosity,Diez del Molino et al.,2018;Leroy et al.,2018)in the face of habitat loss and human *** such species is the jaguar(Pan-thera onca),the largest extant felid in the Americas,whose range has already declined by>50%and some of whose remaining popu-lations have been severely impacted by human-induced habitat frag-mentation(De La Torre et al.,2018).

大熊猫微卫星DNA的筛选及其应用

经ＤｐｎⅡ限制酶消化大熊猫基因组ＤＮＡ后，选取１５０－５００ｂｐ大小的片段构建了ＤＮＡ文库。用合成的（ＣＡ）（１５）探针从这一文库中克隆筛选了１０个大熊猫特异的微卫星ＤＮＡ座位。在测定ＤＮＡ序列的基础上设计并合成了１... 详细信息

经ＤｐｎⅡ限制酶消化大熊猫基因组ＤＮＡ后，选取１５０－５００ｂｐ大小的片段构建了ＤＮＡ文库。用合成的（ＣＡ）（１５）探针从这一文库中克隆筛选了１０个大熊猫特异的微卫星ＤＮＡ座位。在测定ＤＮＡ序列的基础上设计并合成了１０个座位特异的引物，以ＰＣＲ技术扩增了７份抽提自组织细胞和６份抽提自毛发的大熊猫ＤＮＡ样品。发现除座位ｇ（００７）外，其余９个座位均呈多态性，而且所有座位均为偶数碱基的长度变异。对有准确谱系记录的家系的研究结果证明，这１０个微卫星ＤＮＡ座位严格按孟德尔方式遗传。因而这些座位能有效地应用于大熊猫的亲子鉴定，从而为建立各地大熊猫谱系和制定有效的繁殖计划提供了一个有效的遗传学手段。根据这１０个微卫星座位的分析，我们澄清了两组未知的父系关系。

熊科动物是现生的陆生食肉目动物中体形最大者。熊科动物在全球分布有7种,它们被划为2～7个属。迄今为止,熊科的系统发育关系和其物种的分类划分仍然存在着很多争议。主要的原因就在于熊科物种的分化间隔时间相对较短,是一个快速的进化辐射和物种形成事件。以前的研究主要利用线粒体不同基因片段作为分子标记来重建熊科物种的进化历史,但是熊科物种之间的进化关系却因不同作者选取的线粒体片段、分析方法等方面各不相同,其结果也表现出明显的差异。因此,许多学者认为应该挖掘新的不同遗传特征的分子标记,以解决熊科物种进化方面的争议。鉴于此,我们探讨用核基因序列数据研究熊科分类和系统演化中长期悬而未决的疑难问题。

About 336-444 bp mitochondrial D-loop region and tRNA gene were sequenced for 40 individuals of the giant panda which were collected from Mabian, Meigu, Yuexi, Baoxing, Pingwu, Qingchuan, Nanping and Baishuijiang, respectively. 9 haplotypes were found in 21 founders. The results showed that the giant panda has low genetic variations, and that there is no notable genetic isolation among geographical populations. The ancestor of the living giant panda population perhaps appeared in the late Pleistocene, and unfortunately, might have suffered bottle-neck attacks. Afterwards, its genetic diversity seemed to recover to some extent.

Despite the small number of ursid species,bear phylogeny has long been a focus of study due to their conservation value,as all bear genera have been classified as endangered at either the species or subspecies *** Ursidae family represents a typical example of rapid evolutionary *** analyses with a single mitochondrial(mt)gene or a small number of mt genes either provide weak support or a large unresolved polytomy for *** revisit the contentious relationships within Ursidae by analyzing complete mt genome sequences and evaluating the performance of both entire mt genomes and constituent mtDNA genes in recovering a phylogeny of extremely recent speciation *** mitochondrial genome-based phylogeny provides strong evidence that the spectacled bear diverged first,while within the genus Ursus,the sloth bear is the sister taxon of all the other five *** latter group is divided into the brown bear/polar bear and the two black bears/sun bear *** findings resolve the previous conflicts between trees using partial mt *** ability of different categories of mt protein coding genes to recover the correct phylogeny is concordant with previous analyses for taxa with deep divergence times. This study provides a robust Ursidae phylogenetic framework for future validation by additional independent evidence,and also has significant implications for assisting in the resolution of other similarly difficult phylogenetic *** of base composition bias and utilization of the combined data of whole mitochondrial genome sequences has allowed recovery of a strongly supported phylogeny that is upheld when using multiple alternative outgroups for the Ursidae,a mammalian family that underwent a rapid radiation since the mid-to late *** remains to be seen if the reliability of mt genome analysis will hold up in studies of other difficult phylogenetic *** the whole mitochondrial DNA sequence b
