Mesenchymal Bone Marrow-derived Stem Cells Transplantation in patients with HCV Related Liver Cirrhosis

Background and Aims: To evaluate the effect of intrapar-enchymal transplantation of mesenchymal bone marrow-derived stem cells (BMSCs) in patients with hepatitis C virus (HCV)-related liver cirrhosis (LC). Methods: Mononuclear cells were isolated from patient bone marrow and were passaged several times in vitro in order to reach the required volume. Attributes of the BMSCs were evaluated by the presence of the surface markers CD105+, CD90+, and CD73+. Cells from each passage were evaluated for sterility, and they were transplanted intraparenchymally into liver tissue. Clinical and laboratory data were evaluated and morphological studies of liver biopsy were performed prior to and 6 months after transplantation. Results: On clinical evaluation, the general state of these patients was improved at 1 month following transplantation of BMSCs. At 1 and 6 months post-transplantation, jaundice was absent in four (67%) patients. After 6 months, functional hepatic indices were improved, i.e. decrease of ALT and AST activity and bilirubin level. However, these decreases were not statistically different (p>0.05). Expression of CD34 and α-SMA in liver biopsy samples were decreased at 6 months after transplanta-tion, consistent with structural improvements in mitochondria and nuclear compartments. Conclusions: Intraparenchymal transplantation of autologous BMSCs improved the functional condition of the liver, stimulated reparative processes in hepatocytes, and decreased extracellular matrix protein (EMp) count in hepatic tissues of patients with LC. It was well tolerated and was not associated with any complications both during and after BMSC transplantation.

Celecoxib enhances the detoxification of diethylnitrosamine in rat liver cancer

AIM： To study the effect of celecoxib （CXB） on diethylnitrosamine activation through the regulation of cytochrome p450 in a hepatocarcinogenesis ***： Six-week-old male Sprague-Dawley rats were randomly divided into five groups, a non-treated group （NT）, a diethylnitrosamine-treated group （DEN）, a DEN＋CXB-treated group （DEN＋CXB）,and CXB 8 d-treated and CXB 32 d-treated groups. The effects of celecoxib on the enzymatic activities of CYp1A1, 2A, 2B1/2, and 2E1 were assessed in hepatic microsomes 24 h after DEN *** in CYpIA1 and CYp2B1/2 protein expression were also evaluated. The rate of DEN metabolism was measured by the production of the deethylation metabolite acetaldehyde, and the denitrosation metabolite ***： DEN＋CXB administration produced a significant increase in the enzymatic activities ofCYp2B1/2 and 1A1, whereas it did not change the activities of CYp2A and 2E1, compared to that of the DEN group. CXB treatment for eight days did not produce a significant effect on enzymatic activity when compared to the NT group; however, when it was administered for prolonged times （CXB 32 d group）,the enzymatic activities were increased in a similar pattern to those in the DEN＋CXB group. The observed increase in the enzymatic activities in the DEN＋CXB group was accompanied by an increase in the CYp2B1/2 protein levels; no changes were observed in the levels of CYpIA1. In vitro, CXB increased the denitrosation of DEN, a pathway of metabolic detoxification. The addition of SKF-525A, a preferential inhibitor of CYp2B, abrogated the denitrosation of ***： These results suggest that the mechanism of action of CXB involves enhancement of the detoxification of DEN by an increasing denitrosation via CYp2B1/2.

Bile tolerance of Lactobacillus acidophilusLA-K as influenced by various pulsed electric field conditions

Lactobacillus acidophilus has several health benefits and is used as an adjunct bacterium in the manufacture of cultured dairy foods. Bile tolerance is an important probiotic characteristic. pulsed electric field (pEF) processing is non-thermal, hurdle technology which comprises of passing fluid foods between two electrodes and subjecting them to pulses of voltage for less than one second. Whether certain mild pEF conditions can enhance bile tolerance of Lactobacillus acidophilus is not known. The objective was to study the influence of certain pEF conditions on the, bile tolerance of Lactobacillus acidophilus LA-K. Lactobacillus acidophilus LA-K suspended in sterile peptone 0.1% w/v distilled water was treated with pulse widths of 3, 6 and 9 μs, pulse periods of 10,000;20,000 and 30,000 μs and electric field strengths of 5, 15 and 25 kV/cm. The control did not receive any pulsed electric field condition. Bile tolerance was determined hourly for 16 hours. pROC GLM of the Statistical Analysis Systems (SAS) was used for data analysis. Significant differences were determined at p p p p Lactobacillus acidophilus LA-K subjected to 5 kV/cm were signifycantly the highest while the bile tolerance when subjected to 25 kV/cm was significantly the lowest.

Zr（Ⅳ） surface sites determine CH3OH formation rate on Cu/ZrO2/SiO2-CO2 hydrogenation catalysts

Cu/ZrO2/SiO2 are efficient catalysts for the selective hydrogenation of CO2 to CH3OH. In order to understand the role of ZrO2 in these mixed-oxides based catalysts, in situ X-ray absorption spectroscopy has been carried out on the Cu and Zr K-edge. Under reaction conditions, Cu remains metallic, while Zr is present in three types of coordination environment associated with 1) bulk ZrO2, 2) coordinatively saturated and 3) unsaturated Zr(Ⅳ) surface sites. The amount of coordinatively unsaturated Zr surface sites can be quantified by linear combination fit of reference X-Ray absorption near edge structure (XANES) spectra and its amount correlates with CH3OH formation rates, thus indicating the importance of Zr(Ⅳ) Lewis acid surface sites in driving the selectivity toward CH3OH. This finding is consistent with the proposed mechanism, where CO2 is hydrogenated at the interface between the Cu nanoparticles that split H2 and Zr(Ⅳ) surface sites that stabilizes reaction intermediates.

pulsed electric field (pEF) processing involves the application of pulses of voltage for less than one second to fluid foods placed between two electrodes. Lactobacillus acidophilus is an important probiotic bacterium used for the production of fermented dairy products. Acid tolerance is an important probiotic characteristic. The influence of pEF on acid tolerance of Lactobacillus acidophilus is not known. Objective of this study was to elucidate the influence of certain pEF conditions on the acid tolerance of Lactobacillus acidophilus LA-K. Freshly thawed Lactobacillus acidophilus LA-K was suspended in sterile peptone 0.1% w/v distilled water and treated in a pilot plant pEF system. The treatments were pulse width (3, 6 and 9 μs), pulse period (10,000;20,000 and 30,000 μs) and voltage (5, 15 and 25 kV/cm). Control was run through pEF system at 60 mL/min without receiving any pulsed electric field condition. Data were analyzed using the pROC GLM of the Statistical Analysis Systems (SAS). Differences of least square means were used to determine significant differences at p Lactobacillus acidophilus LA-K lowered by increasing pulse widths and voltages but lowering pulse periods.

Objectives In recent years, there has been a substantial increase in the global consumption of honey, driven by the high demand for natural products that offer health benefits. Consequently, consumers show a preference for honey, recognised for its superior quality, namely honey with a protected Designation of Origin (pDO) or honey produced in protected areas, as it is associated with superior attributes and characteristics. portugal is one of the leading countries in the production of pDO honey, with Montesinho Natural park (MNp) being an excellent example of high-quality portuguese honey, known for its distinctive attributes. However, environmental threats pose a double challenge, leading to a decline in honey production and compromising its overall quality. This study aimed to assess the specific physicochemical parameters and nutritional characteristics of MNp honey and investigate their correlation with the different locations of *** and Methods MNp honey samples (n=13) were obtained from local producers or purchased at supermarkets. Standard physicochemical parameters, such as 5-hydroxymethylfurfural, diastase activity, moisture and ash content, free acidity, electrical conductivity and pH, were determined according to honey legislation. Additionally, colour parameters, protein, low-molecular-weight carbohydrates (mono- and disaccharides), and mineral content were *** The results obtained were consistent with the requirements outlined in the legislation and those described in the literature. The results suggest that geographical factors within the park and boundaries do not contribute to variations in the analysed *** A significant level of homogeneity was evident in all parameters evaluated among the MNp honey samples. This is the first comprehensive study of the physicochemical properties of honey from various apiaries within the MNp.

Evaluation of the effect of compound aqueous solubility in cytochrome p450 inhibition assays

It is a common practice in drug discovery organizations to screen new chemical entities in order to predict future drug-drug interactions. For this purpose, there are two main assay strategies, one based on recombinant cytochrome p450 (rCYp) enzymes and fluorescent detection, and other on human liver microsomes (HLM) and liquid chromatography coupled to mass spectrometry. Many authors have reported a poor correlation between both technologies, giving rise to concerns about the usefulness of fluorometric methods for predicting drug-drug interactions. In this study, we investigated the role that compound aqueous kinetic solubility may play in this lack of correlation. We found that drug discovery compounds with unacceptable kinetic solubility, measured by a turbidimetric solutibility assay, tended to yield higher IC50 values in in vitro models based on human liver microsomes, whereas compounds with kinetic solubility values higher than 50 μM showed very similar IC50 values in both in vitro models. Our results show that the turbidimetric solubility assay is a useful tool to identify those discovery compounds that may require further investigation in order to avoid overlooking future drug-drug interactions.

Identification of human papillomavirus in esophageal squamous papillomas

AIM： To investigate the presence of human papillomavirus （HpV） in esophageal squamous papilloma （ESp） and determine p16, p53 and Ki67 expression in a Mexican cohort. METHODS： Nineteen cases diagnosed as ESp, corresponding to 18 patients were reviewed; nineteen cases of normal esophageal mucosa were used as negative controls. HpV detection was performed by ,amplified chromogenic in situ hybridization （ACISH） using a wide spectrum-cocktail probe and pCR. RESULTS： The average age at presentation was 46.3 years （range 28-72 years）. patients included four （22.22%） males and 14 （77.77%） females. The most frequent location was upper third （11 cases）, followed by middle third （3 cases） and unknown site （5 cases）. Immunohistochemistry （IHC） revealed basal and focal p53 expression in 17 cases （89%）; p16 was expressed in eight cases （42.10%） and the Ki67 index ranged from 10% to 30%. HpV was detected in 14 out of 16 cases （87.5%） by ACISH： Twelve showed diffuse nuclear patterns and two showed granular patterns. HpV DNA was identified by pCR in 12 out of 14 cases （85.7%）. Low-risk HpV types were detected in the most of the cases. CONCLUSION： This study provides identification of HpV infection in almost 80% of ESp using either ACISH or pCR; overall, all of these lesions show low expression of cell-cycle markers. We suggest ACISH as an alternative diagnostic tool for HpV detection in ESR .

Cooperative apoptosis of coelomocytes of the holothurian Eupentacta fraudatrixand its modulation by dexamethasone

The capacities of phagocytes of subpopulation p1 (F) and morula cells (MC) of holothurian Eupentacta fraudatrix to modulate apoptosis of each other as well as cytokine-dependent mechanisms and hormonal regulation of these cells’s interaction were studied. The 18-h treatment of F with supernatants, obtained after centrifugation of MC preincubated for 3 h with phosphate buffered saline (pSB) at the temperature of 22℃?(SMC3) resulted in a significant growth of apoptosis level. A 30-min incubation of F with supernatants of MC, preincubated for 24 h (SMC24), on the contrary, reduced the apoptosis level and increased the level of interleukine-1α (IL-1α)-like substances, and 24-h incubation did not influence apoptosis and reduced level of IL-1α-like substances. Thus, proapoptotic effects of MC’s supernatants in F inversely depended on time of their preincubation with pSB and directly on time of incubation with F. Additionally, this effect was opposite to variations in the level of IL-1α-like substances. The level of apoptosis declined after 30 min of incubation but elevated after 24 h at the inverse treatment of MC with supernatant, obtained after preincubation of F during 24 h (SF24). The level of IL-1α-like substances dropped after 30 min and insignificantly decreased after 24 h. Hence, SF24 proapoptotic effect directly depended on time of incubation with MC and did not correspond to variations in the level of IL-1α-like substances. 100 μM dexamethasone stimulated apoptosis in F and MC in an inverse time-dependent manner during 24-h preincubation, and supernatants of cell suspensions obtained after such preincubations, stimulated apoptosis and reduced the IL-1α-like substances level in target cells at both types of interaction. IL-1α-like substances are?supposed to be mediators for MC’s effects in F, but not for F’s action on MC. In holothurians, steroid hormones apparently may participate in the regulation of the immune response and cell cooperation.
