Enzyme-treated date plum leave extract ameliorates atopic dermatitis-like skin lesion in hairless mice

Objective: To evaluate the effect of different extracts of Diospyros lotus leaves in atopic dermatitis Methods: Diospyros lotus leaves were extracted in ethanol and treated with or without hydrochloric acid or α-rhamnosidase to obtain three different extracts-ethanol, acid-hydrolyzed, and enzyme-hydrolyzed leaf extracts of date plum. The myricitrin content in all samples was measured using HPLC analysis. In vitro antioxidant and anti-inflammatory activities of the extracts were determined by measuring DPPH radical scavenging activities and nitric oxide production in RAW264.7 cells, respectively. Sevenweek-old male hairless mice were used to evaluate the anti-atopic dermatitis effects of three extracts in vivo. Splenocytes and mast cells were used to further determine the anti-atopic dermatitis effects of the major compound in the ethanol leaf ***: Enzyme-hydrolyzed leaf extract showed significant in vitro antioxidant and anti-inflammatory activities, and attenuated atopic dermatitis-like skin symptoms and clinical signs more significantly than ethanol and acid-hydrolyzed leaf extracts in 1-fluoro-2,4-dinitrobenzene and house dust mite antigen-treated hairless mice. Enzyme-hydrolyzed leaf extract also suppressed the serum level of immunoglobulin E, interleukin(IL)-4, tumor necrosis factor(TNF)-α, interferon(IFN)-γ, thymic stromal lymphopoietin, and thymus and activation-regulated chemokine in mice with atopic dermatitis. Furthermore, histological analysis revealed that enzymehydrolyzed leaf extract suppressed the increased epidermal thickness, dermal infiltration of inflammatory cells, and infiltration and degranulation of mast cells more markedly than the other two extracts in atopic dermatitis-like skin lesions. In addition, this extract effectively inhibited the production of IFN-γ, IL-4, and thymus and activation-regulated chemokine compared with the other two extracts in concanavalin A-stimulated splenocytes. Objective: To investigate the antibacterial activity of SHHextracted with either water or ethanol against methicillin-resistant Staphylococcus aureus(MRSA) and combinatory antimicrobial effect with ciprofloxacin(CIP) by time kill assay and checkerboard dilution test. Methods: The antibacterial activity determined by broth dilution method indicated that the antibacterial activity of Sami-Hyanglyun-Hwan(SHH) water extract(SHHW) and SHH ethanol extract(SHHE) ranged from 250 to 2000 μg/m L and 125 to 1000 μg/m L against MRSA, respectively. Results: In the checkerboard method, the combinations of SHHE with CIP had a partial synergistic or synergistic effect against MRSA. The time-kill curves showed that a combined SHHE and CIP treatment reduced the bacterial counts dramatically after 24 h. Conclusions: The present study demonstrates the therapeutic ability of SHHE against MRSA infections.

Purpose:This study has been conducted with the aim of providing practical materials for development of an anti-hair loss agent and hair growth promotor that utilizes natural ingredients replacing medications through experimental methods testing growth factor protein expression promotion effect from the Cinnamomum ***:We extracted the active ingredients using distilled water,ethanol,methanol,ethanol-ultrasonic,methanol-ultrasonic,and supercritical extraction *** then evaluated the antioxidant activity and selected the most efficient *** study aimed to evaluate the cytotoxicity of *** extract to human hair follicle dermal papilla cells(HFDPC) using the MTT assay and the promoting effect of *** extract on protein expression of hair-growth relevant factors using western ***:Among the six *** extracts,the highest extraction yield was recovered in the methanolultrasonic extract(2.0378%),the total polyphenol was highly recovered in the ethanol-ultrasonic extract(50.248 mg/g),and the total flavonoid content was maximized in the methanol-ultrasonic extract(5.907 mg/g).The electron-donating ability was elevated in the methanol-ultrasonic extract(96.34%),and the high nitrite-scavenging activity was shown in the supercritical extract(37.59%).No cytotoxicity was observed in HFDPC treated with methanol-ultrasonic extract from *** up to 500 μg/*** treatment of methanol-ultrasonic extract from ***(100 μg/mL) into HFDPC significantly promoted the protein expression of PDGF- β(42%) and IGF(64%) compared to the control,while the protein expression of VEGF,KGF,hGH,and EGF were not significantly ***:Therefore,it is estimated that the methanol-ultrasonic extract from *** could be used as a functional antioxidant and as a hair-growth promoting material.

Isolation and Characterization of Volatile Organic Compounds-Degrading <i>Bacillus</i>Strains from Loess

Volatile organic compounds (VOCs) are harmful to human health and the environment. Recently, loess (Hwangtoh) was used as an eco-friendly interior paint formulation in Korea. It is used even more commonly as a filter carrier to remove VOCs. In this study, we isolated Bacillus strains from a loess filter. The strains that were tolerant to VOCs were labeled according to the series VOC01 to VOC35. Four strains—VOC03, VOC11, VOC18, and VOC30—were investigated for their ability to degrade cyclohexane and toluene. Strain VOC18 best degraded both VOCs, whereas VOC03 demonstrated no ability to degrade VOCs. In keeping with this, VOC18 grew best on cyclohexane or toluene as the sole carbon source. The strains were identified by their physiochemical and phylogenetic characteristics. Strain VOC18 was determined as a strain of Bacillus cereus;VOC11 and VOC30 were determined as differentiated strains of B. thuringiensis. Strain VOC03, which demonstrated high tolerance but no ability to degrade VOCs, was identified as a strain of B. megaterium.

Thermally modified organic materials commonly known as biochar have gained popularity of being used as a soil *** information, however, is available on the role of biochar in alleviating the negative impacts of saline water on soil productivity and plant growth. This study, therefore, was conducted to investigate the effects of Conocarpus biochar(BC) and organic farm residues(FR) at different application rates of 0.0%(control), 4.0% and 8.0%(weight/weight) on yield and quality of tomatoes grown on a sandy soil under drip irrigation with saline or non-saline water. The availability of P, K, Fe, Mn, Zn and Cu to plants was also investigated. The results demonstrated clearly that addition of BC or FR increased the vegetative growth, yield and quality parameters in all irrigation treatments. It was found that salt stress adversely affected soil productivity, as indicated by the lower vegetative growth and yield components of tomato plants. However, this suppressing effect on the vegetative growth and yield tended to decline with application of FR or BC, especially at the high application rate and in the presence of biochar. Under saline irrigation system, for instance, the total tomato yield increased over the control by 14.0%–43.3% with BC and by 3.9%–35.6% with FR. These could be attributed to enhancement effects of FR or BC on soil properties, as indicated by increases in soil organic matter content and nutrient availability. Therefore, biochar may be effectively used as a soil amendment for enhancing the productivity of salt-affected sandy soils under arid conditions.
