Single and double layer centrifugation improve the quality of cryopreserved bovine sperm from poor quality ejaculates

Background： Density gradient centrifugation was reported as a technique of semen preparation in assisted reproductive techniques in humans and animals. This technique was found to be efficient in improving semen quality after harmful techniques such as cryopreservation. Recently a modified technique, single layer centrifugation,was proposed as a technique providing a large amount of high quality spermatozoa, and this treatment was performed before conservation. Single layer centrifugation has been studied prevalently in stallions and in boars,but limited data were available for bulls. Occasionally bulls are known to experience a transient reduction in semen quality, thus techniques that allow improvement in semen quality could be applied in this context. The aim of this study was the evaluation of single layer and double layer centrifugation by the use of iodixanol, compared with conventional centrifugation and non-centrifuged semen, on the sperm characteristics during the cryopreservation process in bulls with normal and poor semen ***： Single layer centrifugation and double layer centrifugation both significantly increased the percentage of normal spermatozoa and decreased the percentage of non-sperm cells in poor quality samples, while both were ineffective in those of normal quality. Sperm characteristics in poor quality samples increased after single layer centrifugation and double layer centrifugation, reaching values similar to those recorded in normal samples, and this trend is maintained after equilibration and after cryopreservation. On the other hand, SLC and DLC resulted in a consistent reduction in the spermatozoa recovered, and this resulted in a reduction of the absolute amount of spermatozoa cryopreserved in the normal samples, without a clear improvement in sperm characteristics in this type of ***： These data suggested that both SLC and DLC could be performed in practice, but their application should be limited to the cas

The experimental prospects of the Bc studies of the LHCb experiment are discussed. Production rates of Bc mesons at different center-of-mass energies are estimated with the dedicated generator BCVEGPY. Theoretical estimates and experimental measurements of the Bc± inclusive production cross section at /s =1.96 TeV are compared. The possibilities of studying Bc production, Bc spectroscopy, Bc decays and CP violation in Bc decays in the LHCb experiment are evaluated.

Background: Semen evaluation is used to estimate the testicular function. In bulls, the spermatozoa present in the ejaculate are the result of a process that begun more than 2 mo earlier, bequeathing a delayed depiction of the actual function of the testis. Since testis vascularization might be critical for the gonad function, selected pulse wave Doppler ultrasound parameters were assessed in this study, for instance the peak systolic velocity, the end diastolic velocity and the resistive index of the testicular artery along the spermatic cord, the marginal portion of the testicular artery and the intratesticular branches of the testicular artery both in healthy adult and young *** between these parameters and characteristics of semen that was collected numerous times, before and after the Doppler ultrasound ***: The peak systolic velocity and the end diastolic velocity measured in the testicular artery along the spermatic cord(supratesticular artery – SA) were variable among the bulls and within individual bulls, likely due to the convoluted course of the vessel. The resistive index was found highly repeatable in the same bull. A reduction in the resistive index was found between the supratesticular artery and the marginal portion of the testicular artery(P < 0.01), and between the marginal portion of the testicular artery and the intratesticular branches of the testicular artery(P < 0.05). No differences were recorded for the pulse wave Doppler ultrasound parameters in young bulls compared with adults. A significant correlation was found between the resistive index of the marginal portion of the testicular artery and total sperm in the ejaculate(r = 0.516, P < 0.05), the immature sperm(r = 0.462, P < 0.05), the teratoid sperm(r = 0.375, P < 0.05), and the "Dag defect" sperm(r = 0.389, P < 0.05). Colorectal cancer(CRC) is the second most common cause of cancer-related death in western countries. Approximately one-quarter of newly diagnosed patients for CRC have metastases, and a further 40%-50% experience disease recurrence or develop metastases after all standard therapies. Therefore, understanding the molecular mechanisms involved in the progression of CRC and subsequently developing novel therapeutic targets is crucial to improve management of CRC and patients' long-term survival. Several tyrosine kinase receptors have been implicated in CRC development, progression and metastasis, including epidermal growth factor receptor(EGFR) and vascular EGFR. Recently, tropomyosin-related kinase B(Trk B), a tyrosine kinase receptor, has been reported in CRC and found to clearly exert several biological and clinical features, such as tumor cell growth and survival in vitro and in vivo, metastasis formation and poor prognosis. Here we review the significance of Trk B and its ligand brain derived-neurotrophic factor in CRC. We focus on their expression in CRC tumor samples, and their functional roles in CRC cell lines and in in vivo models. Finally we discuss therapeutic approaches that can lead to the development of novel therapeutic agents for treating Trk B-expressing CRC tumors.

Mineral trioxide aggregate（MTA） is considered at the present time as the gold standard for root-end filling in endodontic ***, this biocompatible material presents several drawbacks such as a long setting time and handling difficulties. The aim of this article is to present a new commercialized calcium silicate-based material named Biodentine with physical improved properties compared to MTA in a clinical application. Two endodontic microsurgeries were performed by using specific armamentarium（microsurgical instrumentation, ultrasonic tips） under high-power magnification with an operatory microscope. Biodentine was used as a root-end filling in order to seal the root canal system. The two cases were considered completely healed at 1 year and were followed for one more year. The 2-year follow-up consolidated the previous observation with absence of clinical symptoms and radiographic evidence of regeneration of the periapical tissues.

“中国的问题在于,我们有审查制度,却没有严格的审查法规。所以,在能做与不能做的事中间有一大块灰色区域。”Ian Thubron在2012年3月份的-次采访中说道。这块“灰色区域”一直以来让广告商们困惑。本文通过对中法两国广告管理体系的比较分析,重点关注具有争议性的酒类产品以凸显对比效果。本文试图探索中国广告管理体系的灰色区域,并对以下问题做出回答：形成灰色区域的不同因素有哪些?中法两国的区别是什么?这些区别能够给我们关于中国广告行业及其管理体系的怎样的启示? 基于学者Gao Zhihong的方法论,我首先比较中法两国的社会背景,其次通过比较两国广告管理体系分析体系的运营机制,然后以酒类广告为重点,通过对法律法规的比较来分析广告管控的严格度,最后通过对管理实施的比较来分析广告管理效果。我发现,中国的酒类广告管制不及法国同业严格,也不如其清晰明确。我梳理出中国广告管理体系大片灰色区域的形成因素。首先是行业的成熟度,该因素与法律文本的复杂性及其有效实施紧密相关。随着时间发展,行业很可能趋向更为清晰的法制架构,这一趋势从2011年中国颁布行业自律规范这一举措可见一斑;第二是根深蒂固的“酌情处理”文化,我们将在Guhan Yangsheng jiu这一案例中看到,这一因素严重干扰了法律管制;第三个因素是广告管理体系的结构,其决定了广告审核、监控、管理体系的效率和水准,这也是行业成熟的必然结果。只要中国广告协会继续作为非官方的政府机构,以及实施自我管控和政府管控的是同一实体,审查时就始终需要将政治议题纳入考虑,因而灰色区域将大量存在。

Carcinogenesis is a multistep process that requires the accumulation of various genetic and epigenetic aberrations to drive the progressive malignant transformation of normal human *** major hallmarks of carcinogenesis that have been described are angiogenesis and the stem cell characteristic of limitless replicative *** properties have been targeted over the past decade in the development of therapeutic treatments for colorectal cancer(CRC),one of the most commonly diagnosed and lethal cancers *** treatment of solid tumor cancers such as CRC has been challenging due to the heterogeneity of the tumor itself and the chemoresistance of the malignant ***,the same microenvironment that maintains the pool of intestinal stem cells that contribute to the continuous renewal of the intestinal epithelia also provides the necessary conditions for proliferative growth of cancer stem-like *** cancer stem-like cells are responsible for the resistance to therapy and cancer recurrence,though they represent less than 2.5%of the tumor *** stromal environment surrounding the tumor cells,referred to as the tumor niche,also supports angiogenesis,which supplies the oxygen and nutrients needed for tumor ***-angiogenic therapy,such as with bevacizumab,a monoclonal antibody against vascular-endothelial growth factor,significantly prolongs the survival of metastatic CRC ***,such treatments are not completely curative,and a large proportion of patient tumors retain chemoresistance or show *** article reviews the current knowledge regarding the molecular phenotype of CRC cancer cells,as well as discusses the mechanisms contributing to their *** personalized therapeutic approaches that are based on the interaction of the carcinogenic hallmarks,namely angiogenic and proliferative attributes,could improve survival and decrease adverse effects induced by unnecessary chemotherapy.
