Treatment of Chronic Aplastic Anemia with Chinese Patent Medicine Pai-Neng-Da Capsule for Replacing Androgen Partially:A Clinical Multicenter Study

Objective:To evaluate the efficacy and safety of Pai-Neng-Da Capsule(panaxadiol saponins component,PNDC)in combination with the cyclosporine and androgen for patients with chronic aplastic anemia(CAA).Methods:A total of 79 CAA patients was randomly divided into 2 groups by a random number table,including PCA group[43 cases,orally PNDC 320 mg/d plus cyclosporine 5 mg/(kg·d)plus andriol 80 mg/d]and CA group[36 cases,orally cyclosporine 5 mg/(kg·d)plus andriol 160 mg/d].All patients were treated and followed-up for 6 treatment courses over 24 *** complete blood counts,score of Chinese medical(CM)symptoms were assessed and urine routine,electrocardiogram,hepatic and renal function were observed for safety *** masculinization rating scale was established according to the actual clinical manifestations to evaluate the accurate degree of masculinization in female CAA patients treated by ***:The effective rates were 88.1%(37/42)in the PCA group and 77.8%(28/36)in the CA group based on the standard for the therapeutic efficacy evaluation of *** was no significant difference in the white blood cell(WBC)counts,platelet counts and hemoglobin concentration of peripheral blood between two groups after 6 months *** masculinization score of female patient in the PCA group was significantly lower than the CA group(P<0.05).The mild abdominal distention was observed in 1 case in the PCA *** CA group,the abnormalities in the hepatic function developed in 2 cases and the renal disfunction was found in 1 ***:The PNDC possesses certain curative effects in the treatment of CAA without obvious side-effects and can partially replace andriol thereby to reduce the degree of masculinization[Registried at Chinese Clinical Trial Registry(ChicTR1900028153)].

Evolutionary mechanisms and practical significance of reproductive success and clonal diversity in unisexual vertebrate polyploids

Unisexual reproduction is generally relevant to polyploidy, and unisexual vertebrates are often considered an evolutionary “dead end” due to the accumulation of deleterious mutations and absence of genetic diversity. However, some unisexual polyploids have developed strategies to avoid genomic decay, and thus provide ideal models to unveil unexplored evolutionary mechanisms, from the reproductive success to clonal diversity creation. This article reviews the evolutionary mechanisms for overcoming meiotic barrier and generating genetic diversity in unisexual vertebrates, and summarizes recent research advancements in the polyploid Carassius complex. Gynogenetic gibel carp(Carassius gibelio) is a unique amphitriploid that has undergone a recurrent autotriploidy and has overcome the bottleneck of triploid sterility via gynogenesis. Recently, an efficient strategy in which ploidy changes, including from amphitriploid to amphitetraploid, then from amphitetraploid to novel amphitriploid, drive unisexual-sexual-unisexual reproduction transition and clonal diversity has been *** on this new discovery, multigenomic reconstruction biotechnology has been used to breed a novel strain with superior growth and stronger disease resistance. Moreover, a unique reproduction mode that combines both abilities of ameiotic oogenesis and sperm-egg fusion,termed as ameio-fusiongensis, has been discovered, and it provides an efficient approach to synthesize sterile allopolyploids. In order to avoid ecological risks upon escape and protect the sustainable property rights of the aquaculture seed industry, a controllable fertility biotechnology approach for precise breeding is being developed by integrating sterile allopolyploid synthesis and gene-editing *** review provides novel insights into the origin and evolution of unisexual vertebrates and into the attempts being made to exploit new breeding biotechnologies in aquaculture.

Spatiotemporal Characteristics of Typical Ecosystem Services and Their Spatial Responses to Driving Factors in Ecologically Fragile Areas in Upper Yellow River,China

The identification of dominant driving factors for different ecosystem services(ESs)is crucial for ecological conservation and sustainable ***,the spatial heterogeneity of the dominant driving factors affecting various ESs has not been adequately elucidated,particularly in ecologically fragile *** study employed the integrated valuation of ESs and trade-offs(InVEST)model to evaluate four ESs,namely,water yield(WY),soil conservation(SC),habitat quality(HQ),and carbon storage(CS),and then to identify the dominant driving factors of spatiotemporal differentiation of ES and further to characterize the spatial heterogeneity characteristics of the dominant driving factors in the eco-fragile areas of the upper Yellow River,China from 2000 to *** results demonstrated that WY exhibited northeast-high and northwest-low patterns in the upper Yellow River region,while high values of SC and CS were distributed in central forested areas and a high value of HQ was distributed in vast grassland *** CS,WY,and SC exhibited decreasing trends over *** most critical factors affecting WY,SC,HQ,and CS were the actual evapotranspiration,precipitation,slope,and normalized difference vegetation index,*** addition,the effects of different factors on various ESs exhibited spatial *** results could provide spatial decision support for eco-protection and rehabilitation in ecologically fragile areas.

Obesity is associated with gut dysbiosis and metabolic *** tea extract(JSTE)reduced fat accumulation and body weight in obese ***,the effects and mechanism of JSTE in preventing obesity were ***,we used different doses of JSTE(75,150 and 300 mg/(kg·day))to evaluate the effect on high-fat diet(HFD)-induced rats under 8 weeks of ***,our results showed that JSTE could significantly reduce body weight gain,blood lipid levels and fat accumulation,improve fatty damage in liver tissue(P<0.05).In addition,JSTE increased the expression of intestinal tight junction proteins(P<0.05),relieved metabolic endotoxemia(P<0.05)and chronic low-grade inflammation in HFD *** of fecal samples showed that JSTE could effectively reverse the microbial diversity and the ratio of Firmicutes to Bacteroidetes to normal levels in HFD-fed *** and Erysipelotrichaceae,which are positively related to obesity,were decreased by JSTE intervention(P<0.05).while Bifidobacteriaceae,Bacteroidaceae,Akkermansia,and Clostridium,which are negatively related to obesity,were ***,these results suggested that JSTE might effectively prevent obesity by modulating gut microbiota dysbiosis,intestinal barrier dysfunction,metabolic endotoxemia and chronic low-grade infl ammation in HFD-induced rats.

Little is known about the mechanism of climate-vegetation coverage coupled changes in the Tibetan Plateau(TP)region,which is the most climatically sensitive and ecologically fragile region with the highest terrain in the *** study,using multisource datasets(including satellite data and meteorological observations and reanalysis data)revealed the mutual feedback mechanisms between changes in climate(temperature and precipitation)and vegetation coverage in recent decades in the Hengduan Mountains Area(HMA)of the southeastern TP and their influences on climate in the downstream region,the Sichuan Basin(SCB).There is mutual facilitation between rising air temperature and increasing vegetation coverage in the HMA,which is most significant during winter,and then during spring,but insignificant during summer and *** temperature significantly enhances local vegetation coverage,and vegetation greening in turn heats the atmosphere via enhancing net heat flux from the surface to the *** atmospheric heating anomaly over the HMA thickens the atmospheric column and increases upper air *** high pressure anomaly disperses downstream via the westerly flow,expands across the SCB,and eventually increases the SCB *** effect lasts from winter to the following spring,which may cause the maximum increasing trend of the SCB temperature and vegetation coverage in *** results are helpful for estimating future trends in climate and eco-environmental variations in the HMA and SCB under warming scenarios,as well as seasonal forecasting based on the connection between the HMA eco-environment and SCB climate.

Mussel-inspired Methacrylic Gelatin-dopamine/Ag Nanoparticles/Graphene Oxide Hydrogels with Improved Adhesive and Antibacterial Properties for Applications as Wound Dressings

A novel strategy was developed to prepare the methacrylic gelatin-dopamine(GelMA-DA)/Ag nanoparticles(NPs)/graphene oxide(GO) composite hydrogels with good biocompatibility,mechanical properties,and antibacterial ***-inspired DA was utilized to modify the GelMA molecules,which imparts good adhesive performance to the ***,interfacial enhancer,not only improves mechanical properties of the hydrogels,but also provides anchor sites for loading Ag NPs through numerous oxygencontaining functional groups on the *** experimental results show that the GelMA/Ag NPs/GO hydrogels have good biocompatibility,and exhibit a swelling rate of 202±16%,the lap shear strength of 147±17 kPa,and compressive modulus of 136±53 kPa,in the case of the Ag NPs/GO content of 2 mg/*** activity of the hydrogels against both gram-negative and gram-positive bacteria is dependent on the Ag NPs/GO content derived from the release of Ag^(+).Furthermore,the GelMA/Ag NPs/GO hydrogels possess good adhesive ability,which is resistant to highly twisted state when stuck on the surface of *** results demonstrate promising potential of the GelMA-DA/Ag NPs/GO hydrogels as wound dressings for biomedical applications in clinical and emergent treatment.

A New Excess Free Energy Mixing Rule for Representing Vapor-Liquid Equilibria of Mixed Refrigerants

A suitable mixing rule is important for vapor liquid equilibrium(VLE)investigations for mixed *** this work,a new excess free energy mixing rule(MRv)was proposed at zero pressure based on the linear relationship between dimensionless parameter 1/(u-1)and *** mixing rule was explicit adopted variable liquid molar *** applicable temperature range of MRv could be extended by means of an empirical method to estimate the liquid molar volume for components at high *** mixing rules modified Huron-Vidal mixing rule(MHV1),Wong-Sandler mixing rule(WS),and MRv at two reference pressures were used to compare the VLE data in the calculation of 37 mixed *** demonstrated that MRv had a relatively similar accuracy to MHV1 and WS for component and pressure ***,the average excess Gibbs free energy using the MRv mixing rule for the 37 selected mixed refrigerants(0.0013)was much lower than those using the MHV1(0.0078)and WS(0.0809)mixing rules,which was very valuable for the design and optimization of thermodynamic systems using mixed refrigerants.

目的:探讨急性髓系白血病伴骨髓增生异常相关改变(AML-MRC)患者的遗传特征及其预后特点。方法:选取近5年到宁波市第一医院就诊的非M3 AML患者230例,其中58例初发AML-MRC患者,对其进行随访并用SPSS 25.0软件进行相关统计分析。结果:共有49例患者进行了染色体检测,29例(59.2%)染色体异常,其中7q-8例(16.3%),5q-6例(12.2%),17p异常5例(10.2%),高度异常的复杂核型(CK)(≥5个无关染色体异常)13例(26.5%),CK中包含有+8、5q-等染色体异常,单体核型(MK)12例(24.5%)。37例患者进行了基因检测,24例(64.9%)存在基因突变,其中ASXL1突变8例(21.6%),TET2突变6例(16.2%)。Kaplan-Meier分析显示,伴有高度CK(P=0.012)、5q-(P=0.038)以及TP53突变(P=0.008)的AML-MRC患者总体生存率更差。结论:AML-MRC具有独特遗传学特征,高度CK、5q-及TP53突变是影响患者预后的不良因素。
