本文旨在研究人诱导多能干细胞分化的心肌细胞(human induced pluripotent stem cell-derived cardiomyocytes,hiPSc-cMs)的电生理特性。采用时序性短暂激活/短暂抑制wnt信号通路的方法将未分化的IMR90-4细胞系定向诱导分化为心肌细胞。用免疫荧光染色法和流式细胞术检测心肌肌钙蛋白T(cardiac troponin T,cTnT)蛋白表达,计算hiPSc-cMs的分化率。用膜片钳技术记录hiPSc-cMs的动作电位,根据动作电位的表现对心肌细胞进行分类,并进一步分析电生理特征。结果显示,hiPSc-cMs的纯度大于95%。根据动作电位的表现,hiPSc-cMs可分为心房肌样细胞、心室肌样细胞和窦房结样细胞。其中,心室肌样细胞的动作电位时程(action potential duration,APD)、动作电位幅度(action potential amplitude,APA)和最大去极化速率(dV/dt_(max))均大于心房肌样细胞和窦房结样细胞,窦房结样细胞的d V/dt_(max)低于心室肌样细胞和心房肌样细胞。以上结果表明hiPSc-cMs纯度高,并且分化出的三种不同类型的心肌细胞具有和成熟心肌细胞相似的动作电位特征。

Dear editor,Prostate cancer(Pca)remains a major healthcare burden in men globally[1].Most patients present with localized disease,and treatment is recommended based on risk classification systems like the National comprehensive cancer Network(NccN)[2].However,these methods are imprecise for estimating metastasis-free survival and prostate cancer-specific mortality and thus biomarkers that can predict tumor aggression are needed[3–5].Several studies have since characterized the molecular landscape of localized Pca in white[4,5]and Black/African-American men[6],but data is lacking in Asian *** chinese Prostate cancer Genome and Epigenome Atlas(cPGEA)reported on the genomic and epigenomic landscape of 208 Pca of men from china[7].comparative analyses between the cPGEA cohort and data from The cancer Genome Atlas(TcGA)revealed higher frequencies of Forkhead box A1(FOXA1)and chromodomain-helicase DNA-binding 1(cHD1)mutations,and lower frequencies of phosphatase and tensin homolog(PTEN)mutations and transmembrane protease serine 2-E26 transformationspecific related gene(TMPRSS2-ERG)fusion in chinese compared with white men[7].These preliminary findings highlight the presence of race-specific differences in molecular phenotypes of Pca.

目的:观察使用丝裂霉素c小梁切除术成功/合格史的患者超声乳化术后眼压(IOP)控制及滤过泡功能。方法:回顾性队列研究。收集2013-01-01/2015-12-31期间接受小梁切除术并随后进行白内障手术患者的数据。所有患者都曾成功/合格地进行过小梁切除术。他们通过透明角膜切口进行了简单的超声乳化手术。用Goldmann眼压计进行术后眼压随访,复查滤过泡情况。观察术后IOP控制、视力改善、抗青光眼药物的数量以及从小梁切除术到超声乳化术的时间,并与对照组进行比较。结果:共13例15眼患者满足了研究标准,并且在小梁切除术后进行了简单的超声乳化术。其中2眼为继发性青光眼,13眼为原发性青光眼。所有的超声乳化术均在小梁切除术后至少6mo进行(平均14.7±4.3mo)。白内障术前仅1眼(6.7%)需要抗青光眼药物治疗,超声乳化术1a后,增加到4眼(26.7%),超声乳化术2a后,进一步增加到8眼(53.3%)。术后2a所需抗青光眼药水的数量在2~4种之间。在15眼中,白内障术前(平均13.4±2.9 mmHg)与超声乳化术后1a(平均14.1±3.2 mmHg,P=0.357)和2a(平均15.1±3.3 mmHg,P=0.212)的IOP控制无显著性差异。然而,术前平均LogMAR视力为1.52±1.00,术后平均视力为0.53±0.54,超声乳化术后的视力改善是显著的(P=0.000)。结论:结果显示,在不影响眼压控制的情况下,小梁切除术后白内障患者的视力明显改善。然而,手术后,特别是白内障手术后2a,抗青光眼眼药水的使用数量增加,证明了术后滤过泡功能的降低。白内障超声乳化手术后,患者应该被告知重新开始服用抗青光眼药物的可能性。白内障和青光眼手术的时机和顺序应优化,以获得最佳的手术效果。

目的中国自2009年起开展社区卫生服务,截至2014年已建成社区卫生服务中心8 669家,拥有社区卫生服务工作者30万人。然而,至社区卫生服务中心就诊的患者数并不多,因为人们对全科医生缺乏信任,但对专科医生过度依赖。方法于2015年9—12月,采用分层随机抽样法,按社区卫生服务中心在城市和郊区的分布比例(2∶1)在中国东、中、西部地区抽取样本社区卫生服务中心,并采用不同问卷对在其工作的资深临床医生和全科医生进行调查。两套问卷内容均涉及所在社区卫生服务中心的服务人口特征、开办的临床和健康教育活动情况。158例资深临床医生和3 580例全科医生对调查进行了有效应答,问卷有效应答率分别为84%和86%。结果社区卫生服务中心的中位医生数为8例、中位护士数为13例,但仅半数医生注册为全科医生,极少数护士接受过专业的全科培训。98%的社区卫生服务中心配有听诊器,97%配有血压计,但仅43%配有检眼镜,64%配有妇科检查仪器。社区卫生服务中心倾向于有选择地向部分慢性病患者提供医疗服务。全科医生每天接诊的中位患者数为12.5例。多因素分析结果显示,在举办主体为私立医院的社区卫生服务中心、配有药剂师和护士的社区卫生服务中心,全科医生的每日接诊患者数较多。结论中国拥有规模较大的、以年轻人为主的全科医疗服务团队,并配有持续性专业培训;社区卫生服务中心配有基础医疗设备,但仅少数可为患者提供多种常见病的综合性医疗服务;社区卫生服务中心就诊患者数仍较少。

chrysin(5.7-dihydroxyflavone) is a natural flavonoid commonly found in many *** anti-cancer property of chrysin has been demonstrated although the molecular mechanisms remain to be further *** the present study,we found that,pretreatment with chrysin greatly sensitized various human cancer cells to tumor necrosis factor-alpha (TNFα)-induced *** the search of the molecular mechanisms responsible for the sensitization effect of chrysin,we discovered that such sensitization is closely associated with the inhibitory effect of chrysin on TNFα-mediared nuclear transcription factor-kappaB (NF-κB) *** with chrysin inhibited TNFα-induced degradation of Inhibitor of kappaB(lκB) protein and subsequent nuclear translocation of *** a result, chrysin suppressed the expression of NF-κB-targeted anti-apoptotic *** role of c-FUP-L was further confirmed by its ectopic expression,which significantly protecred cell death induced by combined treatment with chrysin and TNFα.Data from this study thus reveal a novel function of chrysin and enhance the value of chrysin as an anti-cancer agent.

Over the past half-century, plastic consumption has grown rapidly due to its versatility, low cost, and unrivaled functional properties. Among the diff erent implemented strategies for recycling waste plastics, pyrolysis is deemed the most economical option. currently, the wax obtained from the pyrolysis of waste plastics is mainly used as a feedstock to manufacture chemicals and fuels or added to asphalt for pavement construction, with no other applications of wax being reported. Herein, the thermal pyrolysis of three common waste polyolefin plastics: high-density polyethylene(HDPE), low-density polyethylene(LDPE), and polypropylene(PP), was conducted at 450 ℃. The waste plastics-derived waxes were characterized and studied for a potential new application: phase change materials(PcMs) for thermal energy storage(TES). Gas chromatography–mass spectrometry analysis showed that paraffin makes up most of the composition of HDPE and LDPE waxes, whereas PP wax contains a mixture of naphthene, isoparaffin, olefin, and paraffin. Diff erential scanning calorimetry(DSc) analysis indicated that HDPE and LDPE waxes have a peak melting temperature of 33.8 ℃ and 40.3 ℃, with a relatively high latent heat of 103.2 J/g and 88.3 J/g, respectively, whereas the PP wax was found to have almost negligible latent heat. Fourier transform infrared spectroscopy and DSc results revealed good chemical and thermal stability of HDPE and LDPE waxes after 100 cycles of thermal cycling. Performance evaluation of the waxes was also conducted using a thermal storage pad to understand their thermoregulation characteristics for TES applications.
