Development and characterization of 3D-printed electroconductive pHEMA-co-MAA nP-laden hydrogels for tissue engineering

Tissue engineering(TE)continues to be widely explored as a potential solution to meet critical clinical needs for diseased tissue replacement and tissue *** this study,we developed a poly(2-hydroxyethyl methacrylate-co-methacrylic acid)(pHEMA-co-MAA)based hydrogel loaded with newly synthesized conductive poly(3,4-ethylene-dioxythiophene)(PEDOT)and polypyrrole(PPy)nanoparticles(nPs),and subsequently processed these hydrogels into tissue engineered constructs via three-dimensional(3D)*** presence of the nPs was critical as they altered the rheological properties during ***,all samples exhibited suitable shear thinning properties,allowing for the development of an optimized processing window for 3D *** were 3D printed into pre-determined disk-shaped configurations of 2 and 10 mm in height and diameter,*** observed that the nPs disrupted the gel crosslinking efficiencies,leading to shorter degradation times and compressive mechanical properties ranging between 450 and 550 *** conductivity of the printed hydrogels increased along with the nP concentration to(5.10±0.37)×10^(−7)s/*** vitro studies with cortical astrocyte cell cultures demonstrated that exposure to the pHEMA-co-MAA nP hydrogels yielded high cellular viability and proliferation ***,hydrogel antimicrobial studies with staphylococcus epidermidis bacteria revealed that the developed hydrogels affected bacterial *** together,these materials show promise for various TE strategies.

Telomeric DnA breaks in human induced pluripotent stem cells trigger ATR-mediated arrest and telomerase-independent telomere damage repair

Although mechanisms of telomere protection are well-defined in differentiated cells,how stem cells sense and respond to telomere dysfunction,in particular telomeric double-strand breaks(DsBs),is poorly ***,we report the DnA damage signaling,cell cycle,and transcriptome changes in human induced pluripotent stem cells(iPsCs)in response to telomere-internal *** engineer human iPsCs with an inducible TRF1-FokI fusion protein to acutely induce DsBs at *** this model,we demonstrate that TRF1-FokI DsBs activate an ATR-dependent DnA damage response,which leads to p53-independent cell cycle arrest in *** CRIsPR–Cas9 to cripple the catalytic domain of telomerase reverse transcriptase,we show that telomerase is largely dispensable for survival and lengthening of TRF1-FokI-cleaved telomeres,which instead are effectively repaired by robust homologous recombination(HR).In contrast to HR-based telomere maintenance in mouse embryonic stem cells,where HR causes ZsCAn4-dependent extension of telomeres beyond their initial lengths,HR-based repair of telomeric breaks is sufficient to maintain iPsC telomeres at a normal length,which is compatible with sustained survival of the cells over several days of TRF1-FokI *** findings suggest a previously unappreciated role for HR in telomere maintenance in telomerase-positive iPsCs and reveal distinct iPsC-specific responses to targeted telomeric DnA damage.

Using photodynamic therapy(PDT)to treat deep-seated cancers is limited due to inefficient delivery of photosensitizers and low tissue penetration of *** nanocarriers are widely used for photosensitizer delivery,while the self-quenching of the encapsulated photosensitizers would impair the PDT ***,the generated short-lived reactive oxygen spieces(ROs)can hardly diffuse out of nanocarriers,resulting in low PDT ***,a smart nanocarrier system which can be degraded by light,followed by photosensitizer activation can potentially overcome these limitations and enhance the PDT efficacy.A light-sensitive polymer nanocarrier encapsulating photosensitizer(RB-M)was *** implantable wireless dual wavelength microLED device which delivers the two light wavelengths sequentially was developed to programmatically control the release and activation of the loaded *** transmitter coils with matching resonant frequencies allow activation of the connected LEDs to emit different wavelengths *** irradiation time,dose,and RB-M concentration were determined using an agent-based digital simulation *** vitro and in vivo validation experiments in an orthotopic rat liver hepatocellular carcinoma disease model confirmed that the nanocarrier rupture and sequential low dose light irradiation strategy resulted in successful PDT at reduced photosensitizer and irradiation dose,which is a clinically significant event that enhances treatment safety.

In this study,the characteristics and preliminary causes of tropical cyclone remote precipitation(TRP)over China during the period from 1979 to 2020 are *** indicated that approximately 72.42%of tropical cyclones(TCs)in the Western Pacific produce TRP over *** peak months for TRP are July and *** four key regions of TRP are the adjacent areas between the sichuan and shaanxi Provinces,the northern coast of the Bohai sea,the coast of the Yellow sea,and the southern coast *** typical distance between the station with TRP and the TC center ranges from 1500 to 2500 *** of these stations are situated north to 60°west of north of the *** south–west water vapor transportation on the west side of the TC is crucial to *** has a decreasing trend because of the decrease in the number of TCs that generate *** the perspective of large-scale environmental conditions,a decrease in the integrated horizontal water vapor transport in China' Mainland,the weakening of upward motion at approximately 25°–35°n,which is inconducive to convection,and an increase in low-level vertical wind shear,which is unfavorable for the development of TC in areas with high frequencies of TRP-related TCs,are the factors that result in the decreasing trend of TRP.

Platelets are small, versatile blood cells that are critical for hemostasis/thrombosis. Local platelet accumulation is a known contributor to proinflammation in various disease states. However, the anti-inflammatory/immunosuppressive potential of platelets has been poorly explored. Here, we uncovered, unexpectedly, desialylated platelets (dPLTs) down-regulated immune responses against both platelet-associated and -independent antigen challenges. Utilizing multispectral photoacoustic tomography, we tracked dPLT trafficking to gut vasculature and an exclusive Kupffer cell-mediated dPLT clearance in the liver, a process that we identified to be synergistically dependent on platelet glycoprotein Ibα and hepatic Ashwell–Morell receptor. Mechanistically, Kupffer cell clearance of dPLT potentiated a systemic immunosuppressive state with increased anti-inflammatory cytokines and circulating CD4+ regulatory T cells, abolishable by Kupffer cell depletion. Last, in a clinically relevant model of hemophilia A, presensitization with dPLT attenuated anti-factor VIII antibody production after factor VIII ( infusion. As platelet desialylation commonly occurs in daily-aged and activated platelets, these findings open new avenues toward understanding immune homeostasis and potentiate the therapeutic potential of dPLT and engineered dPLT transfusions in controlling autoimmune and alloimmune diseases.

Robust Machine Learning Mapping of sEMG signals to Future Actuator Commands in Biomechatronic Devices

A machine learning model for regression of interrupted surface Electromyography(sEMG)signals to future control-oriented signals(e.g.,robot’s joint angle and assistive torque)of an active biomechatronic device for high-level myoelectric-based hierarchical control is proposed.A Recurrent neural network(Rnn)was trained using output data,initially obtained from offline optimization of the biomechatronic(human–robot)device and shifted by the prediction *** input of the Rnn consisted of interrupted sEMG signals(to mimic signal disconnections)and previous kinematic signals of the assistive *** Rnn with a 0.1-s prediction horizon could predict the control-oriented joint angle and assistive torque with 92%and 86.5%regression accuracy,respectively,for the test *** proposed approach permits a fast,predictive,and direct estimation of control-oriented signals instead of an iterative process that optimizes assistive torque in the inverse dynamic simulation of a multibody human–robot *** with these interrupted input signals significantly improves the regression accuracy in the case of sEMG signal *** Robust Predictive Control-oriented Machine Learning(Robust-MusclenET)model can support volitional high-level myoelectric-based control of biomechatronic devices,such as exoskeletons,prostheses,and assistive/resistive *** work should study the application to prosthesis control as well as the repeatability of the high-level controller with electrode *** low-level hierarchical controller that manages the human–robot interaction,the assistance/resistance strategy,and the actuator coordination should also be studied.

The energy landscape represents a high-dimensional mapping of the configurational states of an atomic system with their respective *** isobaric conditions,enthalpy landscapes can be used to account for volumetric changes of the *** the energy or enthalpy landscape holds the key for discovering materials with targeted properties,since the landscape encapsulates the complete thermodynamic and kinetic behavior of a system,including relaxation,metastable phases,and ***,the curse of dimensionality prohibits one from enumerating and visualizing the energy landscape-the energy landscape of an n-atom system has 3n ***,we outline the emerging computational techniques that allow the exploration of complex energy landscapes of materials in three distinct categories:the classical,metaheuristic,and machine learning *** discuss the advantages and disadvantages associated with each of these methods,with a focus on the nature of problems where they can provide excellent solutions(and vice versa).Altogether,in addition to giving an overview of existing approaches,we hope the review provides an impetus to develop novel methods to explore the energy landscapes that can,in turn,provide both a fundamental understanding of the physics of materials and accelerate the discovery of novel materials.

Geochemical and mineralogical studies were performed in the La Pesca(LP)and Tesoro Altamira(TA)beach sediments,located in the Tamaulipas state,northern Gulf of *** main aim of this study is to infer the weathering history and provenance and to discriminate the tectonic environment of the beach *** sediments are composed of quartz with small amounts of accessory minerals such as plagioclase,calcite,orthoclase,microcline,and *** beach sediments are classified as fine-grained and very well sorted,however LP has coarse skewed and leptokurtic sediments,whereas TA has fine-skewed and very leptokurtic *** chemical index of weathering(CIW’)indicates intense weathering in the source *** quartz grain microtextures in the LP and TA are classified into mechanical,chemical,and mechanical/chemical *** features such as fractures,pits,percussion marks,abrasion fatigue,and V-shaped marks favor high-energy littoral,fluvial,subaqueous-marine,and aeolian *** chemical features indicate solution pits and crystalline overgrowth,which suggests a silica saturated marine *** mechanical/chemical features display adhering particles and elongated depressions suggest formation in a sub-aqueous nearshore marine *** and trace elements-based provenance discrimination diagrams indicate a felsic source derived from the Mesa Central(MC),sierra Madre Oriental(sMOr)and Oaxaquia *** major and trace element concentrations imply a passive margin setting for the northern Gulf of Mexico,which is consistent with the general geology.
