Elucidating the Degradation pathways of Human Insulin in the Solid State

While there have been significant advances in the development of peptide oral dosage forms in recent years, highlighted by the clinical and commercial success of approved peptides such as Rybelsus?, there remain several barriers in the way of broad range applicability of this approach to peptide delivery. one such barrier includes the poor physical and chemical stability inherent to their structures, which persists in the solid state although degradation typically occurs at different rates and via different pathways in comparison to the solution state. Using insulin as a model peptide, this work sought to contribute to the development of analytical techniques for investigating common insulin degradation pathways. Chemically denatured,deamidated and aggregated samples were prepared and used to benchmark circular dichroism spectroscopy, reverse phase HpLC and size exclusion chromatography methods for the investigation of unfolding, chemical modifications and covalent aggregation of the insulin molecule respectively. Solid state degraded samples were prepared by heating insulin powder at 60 °C and 75% relative humidity for 1, 3, 5 and 7 d, and the degradation profiles of the samples were evaluated and compared with those observed in solution. While no unfolding was observed to occur, significant deamidation and covalent aggregation were detected. Reductive disulfide bond cleavage using dithiothreitol allowed for separation of the insulin A-and B-chains,offering a facile yet novel means of assessing the mechanisms of deamidation and covalent aggregation occurring in the solid state.

Nd^(3+)-doped tellurite glasses are promising materials for thermometers based on the fluorescence intensity ratio(FIR)***,at high Nd^(3+)concentrations,energy transfer(ET)processes such as optical reabsorption and cross-relaxation can affect the Nd^(3+)emission,which has been little explored in the ***,the present work investigated the use of Nd^(3+)-doped tellurite glass(samples doped with Nd^(3+)at 0.2 mol%,0.5 mol%,2.0 mol%,and 4.0 mol%)in fluorescence thermometers,in the temperature range from 299 to 371 *** results indicate a strong dependence of the FIR parameters on the Nd^(3+)concentration,due to changes in the emission band profiles caused by optical reabsorption of the Nd^(3+)emissions and cross-relaxation processes.A decrease of the relative sensitivity of the ratio^(4)F_(5/2)→^(4)I_(9/2)/^(4)F_(3/2)→^(4)I_(9/2)is observed for samples doped with higher amounts of Nd^(3+).The maximum relative sensitivity at 299 K is 3.00%/K,which is the highest value among the reported Nd^(3+)ions.

The present study is a systematic effort to investigate the structure-sensitive magnetic parameters of Ce^(3+) substituted Ba-Sr hexaferrite nanocrystals chemically formulated as Ba_(0.5)Sr_(0.5)Ce_(x)Fe_(12-x)o_(19) where x=0.0-0.2 with Δx=*** hexaferrite powders were prepared using the sol-gel self-ignition route and structurally characterized by means of powder X-ray diffraction and Fourier transform infrared *** creation of the m-type hexaferrite phase within the synthesized samples was revealed from the Rietveld refinement of the X-ray *** occurrence of a secondary phase of Ceo_(2) was revealed within the hexaferrites for the substitution,x> *** refined X-ray diffraction data were utilized to compute the lattice parameters,X-ray density,and lattice parameter *** crystal structure plotted from the refined XRD data reveals the occupancy of the ions at different lattice *** XpS data of the hexaferrite were analyzed to confirm the oxidation states of the constituent *** nanocrystalline nature of the hexaferrites was revealed from the crystallite sizes calculated using Scherer's *** analysis of FTIR spectra confirms that only a fraction of Ce^(3+)accommodated in the lattice and the remaining Ce content reside in the form of the Ceo_(2) *** morphology of the hexaferrites was analyzed from the FESEm profiles of the ferrite *** magnetic behavior study was performed by analyzing the Curie temperature,hysteresis loops,and hyperfine interactions by means of susceptibility,V.S.m,and mossbauer spectroscopy,*** hexaferrites with increasing coercivity,decreasing saturation magnetization,and decreasing Curie temperature are reported in the *** substituted Ce^(3+)ions inhibit the grain growth and create lattice imperfections giving rise to hexaferrites with tuned magnetic parameters suitable for different applications.

Selection of digital fabrication technique in the construction industry--A multi-criteria decision-making approach

digital fabrication techniques,in recent decades,have provided the basis of a sustainable revolution in the construction ***,selecting the digital fabrication method in terms of manufacturability and functionality requirements is a complex *** paper presents alternatives and criteria for selection of digital fabrication techniques by adopting the multi-criteria decision-making *** alternatives considered in the study are concrete three-dimensional(3D)printing,shotcrete,smart dynamic casting,material intrusion,mesh molding,injection concrete 3D printing,and thin forming *** criteria include formwork utilization,reinforcement incorporation,geometrical complexity,material enhancement,assembly complexity,surface finish,and build *** demonstrates different multi-criteria decision-making techniques,with both subjective and objective weighting *** given ranking is based on the current condition of digital fabrication in the construction *** study reveals that in the selection of digital fabrication techniques,the criteria including reinforcement incorporation,build area,and geometrical complexity play a pivotal role,collectively accounting for nearly 70% of the overall *** the evaluated techniques,concrete 3D printing emerged as the best performer,however the shotcrete and mesh molding techniques in the second and third positions.

Compact mid-infrared dual-comb spectrometer over 3-4μm via intra-pulse difference frequency generation in LiNbo_(3) waveguides

The mid-infrared optical frequency comb is a powerful tool for gas *** this study,we demonstrate a simple midinfrared dual-comb spectrometer covering 3±4μm in Li Nb o_(3)*** on a low-power fiber laser system,the mid-infrared comb is achieved via intra-pulse difference frequency generation in the Li Nb o_(3)*** construct pre-chirp management before supercontinuum generation to control spatiotemporal alignment for pump and signal *** supercontinuum is directly coupled into a chirped periodically poled Li Nb o_(3)waveguide for the 3±4μm idler generation.A mid-infrared dual-comb spectrometer based on this approach provides a 100 mHz resolution over25 THz *** evaluate the applicability for spectroscopy,we measure the methane spectrum using the dualcomb *** measured results are consistent with the HITRAN database,in which the root mean square of the residual is 3.2%.This proposed method is expected to develop integrated and robust mid-infrared dual-comb spectrometers on chip for sensing.

Influence of High-Density Bedding plane Characteristics on Hydraulic Fracture propagation in Shale oil Reservoir

The existence of high-density bedding planes is a typical characteristic of shale oil *** the behavior of hydraulic fracturing in high-density laminated rocks is significant for promoting shale oil *** this study,a hydraulic fracturing model considering tensile failure and frictional slip of the bedding planes is established within the framework of the unified pipe-interface element method(Up-IEm).The model developed for simulating the interaction between the hydraulic fracture and the bedding plane is validated by comparison with experimental *** hydraulic fracturing patterns in sealed and unsealed bedding planes are ***,the effects of differential stress,bedding plane permeability,spacing,and the friction coefficient of the bedding plane are *** results showed that a single main fracture crossing the bedding planes is more likely to form in sealed bedding planes under high differential *** decrease in bedding plane permeability and the increase in the friction coefficient also promote the fracture propagating perpendicular to the bedding *** with high-density bedding planes has a poorer fracturing effect than that with low-density bedding planes,as the hydraulic fracture is prone to initiate and propagate along the bedding ***,higher injection pressure is needed to maintain fracture propagation along the *** increase in bedding density will lead to a smaller fracturing *** fluid seepage into the bedding planes slows shale *** is recommended that increasing the injection flow rate,selecting alternative fracturing fluids,and employing multi-well/multi-cluster fracturing may be efficient methods to improve energy production in shale oil reservoirs.

A native herbaceous community exerts a strong allelopathic effect on the woody range-expander Betula fruticosa

Biological invasions by alien and range-expanding native plant species can suppress native plants through ***,the homeland security hypothesis suggests that some native plants can resist invasion by producing allelopathic compounds that inhibit the growth of invasive *** research has focused on allelopathic interactions between individual native and invasive plant species,with less emphasis on how allelopathy helps entire native communities resist ***,limited knowledge exists about allelopathic interactions between range-expanding native species and recipient native communities,and their influence on invasion *** bridge this knowledge gap,we conducted two greenhouse competition experiments to test reciprocal allelopathic effects between a native woody range-expanding species,Betula fruticosa,and a community of four native herbaceous species(Sanguisorba officinalis,Gentiana manshurica,Sium suave and Deyeuxia angustifolia)in *** assessed whether *** and the native community differed in their competitive effects and responses,and whether these were influenced by activated carbon,which neutralizes allelochemicals in the *** carbon reduced the suppressive effects of the native community on the above-ground biomass of ***,which indicates that the native community exerted a strong allelopathic effect on *** contrast,activated carbon only marginally enhanced the suppressive effects of *** on the native community,which indicates that allelopathy is not the primary mechanism by which *** exerts its ***,these findings support the homeland security hypothesis and suggest that biotic resistance from the native herbaceous community may limit the invasion success of the woody range-expander ***.
