Single nucleotide polymorphisms of OCTN1, OCTN2, and DLG5 genes in Greek patients with Crohn's disease

AIM: To validate novel single nucleotide polymorphisms (SNPs) in Greek patients with Crohn's disease (CD).METHODS: A total of 120 patients with CD, 85 patients with UC, and 100 unrelated healthy controls were genotyped. Genotyping was performed by allele-specific PCR or by PCR-RFLP ***: Our results showed that the 1672T and -207C alleles were obviously over-represented in CD patients only (P＜0.01 and P＜0.05, respectively) compared to the control population. The G113A polymorphism was completely absent in our studied population. The odds ratio for the carriage of the TC haplotype was 2.21 for CD patients as compared with controls. Additionally, the frequency of the TC haplotype was increased in patients with ileocolitis or colitis, and was mainly associated with the fibrostenotic phenotype of the disease. Furthermore, when the TC haplotype was compared j.intly with the carriage of at least one mutation of the NOD2/CARD15 gene, there was an increased risk for CD, but not for UC, compared to controls. Regarding the location of the disease, the concomitant presence of the TC haplotype and NOD2/CARD15 mutations was mainly associated with ileocolitis or ileitis. CONCLUSION: Collectively, our results suggest that the 1672T variant of the OCTN1 gene and the -207C variant of the OCTN2 gene represent risk factors for CD in the Greek population.

Fleshy-fruited plants in tropical forests largely rely on vertebrate frugivores to disperse their *** this plant-animal interaction is typically considered a diffuse mutualism,it is fundamental as it provides the template on which tropical forest communities are *** applied a mutualistic network approach to investigate the relationship between small-fruited fleshy plant species and the fruit-eating bird community in an intact evergreen forest in north-east Thailand.A minimum of 53 bird species consumed fruits of 136 plant ***-avian frugivore networks were highly asymmetrical,with observed networks filling 30%of all potential *** some of the missing links in the present study might be due to undersampling,forbidden links can be attributed to size constraints,accessibility and phenological uncoupling,and although the maj.rity of missing links were unknown(58.2%),many were probably due to a given bird species being either rare or only a very occasional fruit *** most common frugivores were bulbuls,barbets and fairy-bluebirds,which were responsible for the maj.rity of fruit removal from small fleshy fruited species in our *** birds seemed to be a minor component of the plant-frugivore networks,accounting for only 3%of feeding visits to fruiting trees;they filled 2%of the overall potential *** maj.rity of interactions were generalized unspecific;however,Saurauia roxburghii *** to be dependent on flowerpeckers for dispersal,while Thick-billed Pigeons were only seen to eat figs.

Octreotide induces caspase activation and apoptosis in human hepatoma HepG2 cells

AIM: To investigate the role of octreotide on cellular proliferation and apoptosis of human hepatoma (HepG2) cells. METHODS: We studied cellular proliferation, apoptosis and the possible internal caspase-mediated apoptosis pathway involved, after treatment of HepG2 carcinoma cells with octreotide in comparison with the apoptosis caused by tumor necrosis factor-α (TNF-α). Activities of caspase-3, caspase-9, caspase-8 and caspase-2 were studied, while apoptosis was investigated through detection of DNA fragmentation and through identification of apoptotic cells with the annexin-V/propidium iodide flow cytometric method. RESULTS: After an initial increase in HepG2 cellular proliferation, a significant inhibition was observed with 10-8 mol/L octreotide, while TNF-α dose-dependently decreased proliferation. Early and late apoptosis was significantly increased with both substances. Octreotide significantly increased caspase-3, caspase-8 and caspase-2 activity. TNF-α signifi cantly increased only caspase-2. Cellular proliferation was decreased after treatment with octreotide or TNF-α alone but, in contrast to TNF-α, octreotide decreased proliferation only at concentrations of 10-8 mol/L, while lower concentrations increased proliferation. CONCLUSION: Our findings are suggestive of caspasemediated signaling pathways of octreotide antitumor activity in HepG2 cells, and indicate that measurements of serum octreotide levels may be important, at least in clinical trials, to verify optimal therapeutic drug concentrations.

Analytical Modeling to Predict the Strain Energy of Circular GLARE Fiber-Metal Laminates Under Lateral Indentation

This article deals with the strain energy calculation of thin circular clamped Glass Reinforced (GLARE) fiber-metal laminates subj.cted to static indentation by a lateral hemispherical indentor. Using one-, two- and three-parameter Ritz approximations, analytical equations of the strain energy as a function of the central plate deflection are derived. Previously published analytical formulas, concerning the load-indentation response of circular GLARE plates, are used in order to determine the Ritz parameters and the first failure load and deflection due to tensile fracture of glass-epoxy layers. In this study, the membrane and bending strain energy components of aluminum and prepreg layers are determined. Also, the elastic and plastic strain energy absorbed during the indentation loading are calculated. The derived formulas are applied successfully for GLARE 2-2/1-0.3, GLARE 3-3/2-0.4 and GLARE 3I (special lay-up) circular plates subj.cted to lateral indentation. The strain energy results converge satisfactorily in all examined cases. The predicted strain energy-indentation response is compared with published experimental data and a good agreement is found. No other solution of this problem is known to the authors.

Knowledge of an animal＇s home range is a crucial component in making informed management decisions. However, many home range studies are limited by study area size, and therefore may underestimate the size of the home range. In many cases, individuals have been shown to travel outside of the study area and utilize a larger area than estimated by the study design. In this study, data collected by multiple research groups studying bottlenose dolphins on the east coast of Florida were combined to determine how home range estimates increased with increasing study area size. Home range analyses utilized photo-identification data collected from 6 study areas throughout the St j.hns River （S j.; j.cksonville, FL, USA） and adj.cent waterways, extending a total of 253km to the southern end of Mosquito Lagoon in the Indian River Lagoon Estuarine System. Univariate kernel density estimates （KDEs） were computed for individuals with 10 or more sightings （n= 20）. Kernels were calculated for the primary study area （S j.） first, then additional kernels were calculated by combining the S j. and the next adj.cent waterway; this continued in an additive fashion until all study areas were included. The 95% and 50% KDEs calculated for the S j. alone ranged from 21 to 35 km and 4 to 19 kin, respectively. The 95% and 50% KDEs calculated for all combined study areas ranged from 116 to 217 km and 9 to 70 km, respectively. This study illustrates the degree to which home range may be underestimated by the use of limited study areas and demonstrates the benefits of conducting collaborative science.

The Study of a Simple Pine-Oil Based Laboratory Prepared and Commercial Detergents Using Conductivity Measurements

Detergents are very important substances in everyday life as they are used in laundry services. This manuscript reports the study of the commercial and laboratory prepared pine oil-based detergents employing the critical micelle concentration (CMC) phenomenon using conductivity measurements. The two samples showed the CMC values of 0.0725 g/cm3 and 0.0920 g/cm3 for laboratory and commercial samples respectively. The effect of ionic strength was investigated using NaCl and it demonstrated a drop of CMC value of about CMC by 40% (laboratory sample) and 70% (commercial sample) while the equi-molar naphthalene increased the CMC values by about 50% (laboratory sample) and 12% (commercial sample) relative to their corresponding values under distilled water. The combined effect of equi-molar NaCl and naphthalene lowered the CMC by 5% (laboratory sample) and 30% (commercial sample). These differences could signify the superiority of the laboratory sample in that it is somehow buffered against drastic changes in the CMC under different conditions. The relationship between conductivity and the CMC values does not show sufficient linearity (R2 < 0.8403) suggesting different mechanisms of interactions between NaCl and naphthalene. Overall, the results are gratifying to the small-scale manufacturer who supplied the preparation protocol for laboratory sample preparation, in two respects: they inspire some degree of confidence in his product as well as enabling the manufacturer to employ the same protocol for his quality control practices as such improve product consistency and hence profitability.

Estimating a Falsified Model

It is common econometric practice to propose a system of equations, termed the “structure,” estimate each endogenous variable in the structure via a linear regression with all of the exogenous variables as arguments, and then employ one of variety of regression techniques to recapture the coefficients in the (j.cobian) arrays of the structure. A recent literature, e.g., [1], has shown that a qualitative analysis of a model’s structural and estimated reduced form arrays can provide a robust procedure for assessing if a model’s hypothesized structure has been falsified. This paper shows that the even weaker statement of the model’s structure provided by zero restrictions on the structural arrays can be falsified, independent of the proposed nonzero entries. When this takes place, multi-stage least squares, or any procedure for estimating the structural arrays with the zero restrictions imposed, will present estimates that could not possibly have generated the data upon which the estimated reduced form is based. The examples given in the paper are based upon a Monte Carlo sampling procedure.

Regression Modeling of Individual-Patient Correlated Discrete Outcomes with Applications to Cancer Pain Ratings

Purpose: To formulate and demonstrate methods for regression modeling of probabilities and dispersions for individual-patient longitudinal outcomes taking on discrete numeric values. Methods: Three alternatives for modeling of outcome probabilities are considered. Multinomial probabilities are based on different intercepts and slopes for probabilities of different outcome values. Ordinal probabilities are based on different intercepts and the same slope for probabilities of different outcome values. Censored Poisson probabilities are based on the same intercept and slope for probabilities of different outcome values. Parameters are estimated with extended linear mixed modeling maximizing a likelihood-like function based on the multivariate normal density that accounts for within-patient correlation. Formulas are provided for gradient vectors and Hessian matrices for estimating model parameters. The likelihood-like function is also used to compute cross-validation scores for alternative models and to control an adaptive modeling process for identifying possibly nonlinear functional relationships in predictors for probabilities and dispersions. Example analyses are provided of daily pain ratings for a cancer patient over a period of 97 days. Results: The censored Poisson approach is preferable for modeling these data, and presumably other data sets of this kind, because it generates a competitive model with fewer parameters in less time than the other two approaches. The generated probabilities for this model are distinctly nonlinear in time while the dispersions are distinctly nonconstant over time, demonstrating the need for adaptive modeling of such data. The analyses also address the dependence of these daily pain ratings on time and the daily numbers of pain flares. Probabilities and dispersions change differently over time for different numbers of pain flares. Conclusions: Adaptive modeling of daily pain ratings for individual cancer patients is an effective way
