Dual-specificity phosphatase 6(DUSP6): a review of its molecular characteristics and clinical relevance in cancer

Mitogen-activated protein kinases(MAPKs) are the main regulators of cellular proliferation, growth, and survival in physiological or pathological conditions. Aberrant MAPK signaling plays a pivotal role in carcinogenesis, which leads to development and progression of human cancer. Dual-specificity phosphatase 6(DUSP6), a member of the MAPK phosphatase family, interacts with specifically targeted extracellular signal-regulated kinase 1/2 via negative feedback regulation in the MAPK pathway of mammalian cells. This phosphatase functions in a dual manner, pro-oncogenic or tumor-suppressive, depending on the type of cancer. To date, the tumor-suppressive role of DUSP6 has been demonstrated in pancreatic cancer, non-small cell lung cancer, esophageal squamous cell and nasopharyngeal carcinoma, and ovarian cancer. Its pro-oncogenic role has been observed in human glioblastoma, thyroid carcinoma, breast cancer, and acute myeloid carcinoma. Both roles of DUSP6 have been documented in malignant melanoma depending on the histological subtype of the cancer. Loss-or gain-of-function effects of DUSP6 in these cancers highlights the significance of this phosphatase in carcinogenesis. Development of methods that use the DUSP6 gene as a therapeutic target for cancer treatment or as a prognostic factor for diagnosis and evaluation of cancer treatment outcome has great potential. This review focuses on molecular characteristics of the DUSP6 gene and its role in cancers in the purview of development, progression, and cancer treatment outcome.

Currently,the sentiment analysis research in the Malaysian context lacks in terms of the availability of the sentiment lexicon.Thus,this issue is addressed in this paper in order to enhance the accuracy of sentiment analysis.In this study,a new lexicon for sentiment analysis is constructed.A detailed review of existing approaches has been conducted,and a new bilingual sentiment lexicon known as MELex(Malay-English Lexicon)has been generated.Constructing MELex involves three activities:seed words selection,polarity assignment,and synonym expansions.Our approach differs from previous works in that MELex can analyze text for the two most widely used languages in Malaysia,Malay,and English,with the accuracy achieved,is 90%.It is evaluated based on the experimentation and case study approaches where the affordable housing projects in Malaysia are selected as case projects.This finding has given an implication on the ability of MELex to analyze public sentiments in the Malaysian context.The novel aspects of this paper are two-fold.Firstly,it introduces the new technique in assigning the polarity score,and second,it improves the performance over the classification of mixed language content.

Ontogenetic changes in feeding and food preferences of the dog conch Laevistrombus canarium Linnaeus 1758 (Mollusca： Gastropoda) from Merambong shoal, Malaysia

Laevistrombus canarium is one of the marine gastropod mollusks that have high commercial value,particularly in the aquaculture sector in Malaysia.This study was conducted to determine the feeding and food items of L.canarium at dif ferent ontogenetic stages(juveniles,sub-adults and adults)from Merambong shoals,Malaysia.Field observations on feeding activity were conducted,followed by detailed laboratory analysis on the stomach content.Five-minutes observations on randomly selected individuals were conducted at the fi eld sampling site and their feeding activities were recorded with reference to age stage.Various shell sizes from each ontogenetic stage were randomly collected and quickly anaesthetized with ice and preserved in 10%formalin before being transported to the laboratory for stomach content analyses.Field observations showed that L.canarium mainly grazed on epiphytes occurring on seagrass(46.67%),followed by sediment surface(40%)and epiphytes occurring on macroalgae(13.33%).Stomach content analyses showed a signifi cant dif ference(P0.05)was detected between the three main food items(diatoms,sand particles and detritus)among the ontogenetic stages.Therefore,feeding activity revealed the role of the dog conch in the marine food network.While,classifi cation of the types of food consumed by L.canarium through stomach content analysis determines the particular position of the gastropod in the food chain.Further studies are needed to provide a better insight between trophic relationships of L.canarium with marine ecosystem.

Iodine and Selenium Intake in a Sample of Women of Childbearing Age in Palmerston North, New Zealand after Mandatory Fortification of Bread with Iodised Salt

Iodine deficiency is a worldwide public health problem, which has long been observed in many parts of the world, including New Zealand (NZ). The aim of this study was to assess iodine and selenium intake among women of childbearing age in Palmerston North, New Zealand post mandatory fortification of bread with iodised salt. Fifty women of childbearing age completed a researcher-led questionnaire, including a semi-quantitative food frequency questionnaire. Iodine and selenium were analysed in 24-hour urine samples. The median urinary iodine concentration (UIC) was 65 μg/l with 30% below 50 μg/l;representing mild iodine deficiency according to the World Health Organization. The estimated median daily iodine intake (130 μg/day) was higher than the Estimated Average Requirement (100 μg/day) and higher than seen in women prior to fortification. The median excretion of selenium (32 μg/day) was slightly above level suggested as adequate (30 μg/day) and estimated median intake (57 μg/day) was higher than Estimated Average Requirement (50 μg/day). Selenium and iodine excretion were significantly correlated (Spearman’s rank order;r(50) = 0.547, p 0.001). The major contributors to iodine intake were milk (36%), bread (25%) and fish/seafood (15%). Participants had a mean intake of 2.5 slices of bread/day, which contributed approximately 14 to 20 μg of iodine. The majority of participants (74%) had iodised salt at home, but less than half (48%) used iodised salt exclusively. In conclusion, despite the mandatory fortification of bread with iodised salt in NZ, UIC of the study population indicates iodine deficiency although their estimated dietary intakes appear adequate. It is essential that government initiatives to improve iodine status are evaluated for their efficacy.

The coastal medicinal plant Vitex rotundifolia: a mini-review on its bioactive compounds and pharmacological activity

Humans have long used natural remedies like plants and herbs to treat disease.Furthermore,research has been ongoing to find alternative pharmaceutical drugs based on traditionally used plants,as natural products show fewer side effects compared to synthetic drugs.Medicinal plants have long been targeted in drug development due to their bioactive compounds like alkaloids,flavonoids,and terpenoids.This is not only the case for terrestrial plants,but marine environments also provide a larger diversity of flora and fauna with medicinal bioactive compounds.Vitex rotundifolia,also known as Beach Vitex,is a coastal plant that has been traditionally used to treat a variety of diseases including premenstrual syndrome,headaches,migraines,colds,and eye pain.There have been many review papers on V.rotundifolia,emphasizing its taxonomy,distribution,and biological activity.Our current mini-review not only summarizes the pharmacology and bioactivity of V.rotundifolia,but it also provides new information on the main bioactive compounds of V.rotundifolia such as flavonoids,phenolic acid,and terpenes and their current pharmacological activity in vitro and in vivo research.This information can be useful for developing new pharmaceutical and nutraceutical agents to treat and manage disease.
