Robust transcriptional indicators of immune cell death revealed by spatiotemporal transcriptome analyses

Recognition of a pathogen by the plant immune system often triggers a form of regulated cell death traditionally known as the hypersensitive response(HR).This type of cell death occurs precisely at the site of pathogen recognition,and it is restricted to a few cells.Extensive research has shed light on how plant immune receptors are mechanistically activated.However,two central key questions remain largely unresolved:how does cell death zonation take place,and what are the mechanisms that underpin this phenomenon?Consequently,bona fide transcriptional indicators of HR are lacking,which prevents deeper insight into its mechanisms before cell death becomes macroscopic and precludes early or live observation.In this study,to identify the transcriptional indicators of HR we used the paradigmatic Arabidopsis thaliana–Pseudomonas syringae pathosystem and performed a spatiotemporally resolved gene expression analysis that compared infected cells that will undergo HR upon pathogen recognition with bystander cells that will stay alive and activate immunity.Our data revealed unique and time-dependent differences in the repertoire of differentially expressed genes,expression profiles,and biological processes derived from tissue undergoing HR and that of its surroundings.Furthermore,we generated a pipeline based on concatenated pairwise comparisons between time,zone,and treatment that enabled us to define 13 robust transcriptional HR markers.Among these genes,the promoter of an uncharacterized AAA-ATPase was used to obtain a fluorescent reporter transgenic line that displays a strong spatiotemporally resolved signal specifically in cells that will later undergo pathogen-triggered cell death.This valuable set of genes can be used to define cells that are destined to die upon infection with HR-triggering bacteria,opening new avenues for specific and/or high-throughput techniques to study HR processes at a single-cell level.

shorebird populations are declining worldwide,mainly due to human disturbances and loss of coastal wetlands.However,supratidal habitats as saltpans could play a role in buffering human impact.saltpans have shown to be important as feeding or breeding sites of some shorebird species.A potential conservation strategy to increase shorebird populations in saltpans is to manipulate the cues that birds use to select optimal breeding habitat.Here it is hypothesized that shorebirds are attracted to bivalve shells due to the advantages they offer.Following this hypothesis,we supplemented a restored saltpan in 2019 and 2021 with bivalve shells,expecting an increase in the number of breeding birds’ nests.More than 75% of Kentish Plover(Charadrius alexandrinus) and Little Tern(sternula albifrons) nests were found in patches with shells in both years.The best model for both species indicates that the presence of shells is the factor that most correlates with the location of nests.The probability of choosing one place over another to settle their nest increases in areas with an abundance of shells,double in the case of the Kentish Plover and triple in the case of the Little Tern.The result of this study may constitute a valuable tool for attracting birds to restored saltpans and could contribute to the success of expensive restoration projects where time is usually a constraint.

An electrolyte destined for use in a dual-ion battery(DIB)must be stable at the inherently high potential required for anion intercalation in the graphite electrode,while also protecting the Al current collector from anodic dissolution.A higher salt concentration is needed in the electrolyte,in comparison to typical battery electrolytes,to maximize energy density,while ensuring acceptable ionic conductivity and operational safety.In recent years,studies have demonstrated that highly concentrated organic electrolytes,ionic liquids,gel polymer electrolytes(GPEs),ionogels,and water-in-salt electrolytes can potentially be used in DIBs.GPEs can help reduce the use of solvents and thus lead to a substantial change in the Coulombic efficiency,energy density,and long-term cycle life of DIBs.Furthermore,GPEs are suited to manufacture compact DIB designs without separators by virtue of their mechanical strength and electrical performance.In this review,we highlight the latest advances in the application of different electrolytes in DIBs,with particular emphasis on GPEs.

Background: Understanding the role of species identity in interactions among individuals is crucial for assessing the productivity and stability of mixed forests over time. However, there is limited knowledge concerning the variation in competitive effect and response of different species along climatic gradients. In this study, we investigated the importance of climate, tree size, and competition on the growth of three tree species: spruce(Picea abies), fir(Abies alba), and beech(Fagus sylvatica), and examined their competitive response and effect along a climatic gradient.Methods: We selected 39 plots distributed across the European mountains with records of the position and growth of 5,759 individuals. For each target species, models relating tree growth to tree size, climate and competition were proposed. Competition was modelled using a neighbourhood competition index that considered the effects of inter-and intraspecific competition on target trees. Competitive responses and effects were related to climate.Likelihood methods and information theory were used to select the best model.Results: Our findings revealed that competition had a greater impact on target species growth than tree size or climate. Climate did influence the competitive effects of neighbouring species, but it did not affect the target species? response to competition. The strength of competitive effects varied along the gradient, contingent on the identity of the interacting species. When the target species exhibited an intermediate competitive effect relative to neighbouring species, both higher inter-than intraspecific competitive effects and competition reduction occurred along the gradient. Notably, species competitive effects were most pronounced when the target species' growth was at its peak and weakest when growing conditions were far from their maximum.Conclusions: Climate modulates the effects of competition from neighbouring trees on the target tree and not the susceptibility of the target tree to competition. Tropospheric reactive bromine is important for atmospheric chemistry,regional air pollution,and global climate.Previous studies have reported measurements of atmospheric reactive bromine species in different environments,and proposed their main sources,e.***.sea-salt aerosol（ssA）,oceanic biogenic activity,polar snow/ice,and volcanoes.Typhoons and other strong cyclonic activities（e.***.hurricanes） induce abrupt changes in different earth system processes,causing widespread destructive effects.However,the role of typhoons in regulating reactive bromine abundance and sources remains unexplored.Here,we report field observations of bromine oxide（BrO）,a critical indicator of reactive bromine,on the Huaniao Island（HNI）in the East China sea in July 2018.We observed high levels of BrO below 500 m with a daytime average of9.7±4.2 pptv and a peak value of ～26 pptv under the influence of a typhoon.Our field measurements,supported by model simulations,suggest that the typhoon-induced drastic increase in wind speed amplifies the emission of ssA,significantly enhancing the activation of reactive bromine from ssA debromination.We also detected enhanced BrO mixing ratios under high NOxconditions（ppbv level） suggesting a potential pollution-induced mechanism of bromine release from ssA.such elevated levels of atmospheric bromine noticeably increase ozone destruction by as much as ～40% across the East China sea.Considering the high frequency of cyclonic activity in the northern hemisphere,reactive bromine chemistry is expected to play a more important role than previously thought in affecting coastal air quality and atmospheric oxidation capacity.We suggest that models need to consider the hitherto overlooked typhoon-and pollution-mediated increase in reactive bromine levels when assessing the synergic effects of cyclonic activities on the earth system.

Cancer and atrial fibrillation(AF)are common co-morbid conditions in older adults.Both cancer and cancer treatment increase the risk of developing new AF which increases morbidity and mortality.Heart rate and rhythm control along with anticoagulation therapy remain the mainstay of treatment of AF in older adults with both cancer and AF.Adjustments to the treatment may be necessary because of drug interactions with concurrent chemotherapy.Cancer and old age increase the risk of both,thromboembolism and bleeding.The risk of these complications is further enhanced by concomitant cancer therapy,frailty,poor nutrition status and,coexisting geriatric syndromes.Therefore,careful attention needs to be given to the risks and benefits of using anticoagulant medications.This review focuses on the management of AF in older patients with cancer,including at the end-of-life care.

Background The Inspiration4(I4)mission,the first all-civilian orbital flight mission,investigated the physiological effects of short-duration spaceflight through a multi-omic approach.Despite advances,there remains much to learn about human adaptation to spaceflight's unique challenges,including microgravity,immune system perturbations,and radiation exposure.Methods To provide a detailed genetics analysis of the mission,we collected dried blood spots pre-,during,and post-flight for DNA extraction.Telomere length was measured by quantitative PCR,while whole genome and cfDNA sequencing provided insight into genomic stability and immune adaptations.A robust bioinformatic pipeline was used for data analysis,including variant calling to assess mutational burden.Result Telomere elongation occurred during spaceflight and shortened after return to Earth.Cell-free DNA analysis revealed increased immune cell signatures post-flight.No significant clonal hematopoiesis of indeterminate potential(CHIP)or whole-genome instability was observed.The long-term gene expression changes across immune cells suggested cellular adaptations to the space environment persisting months post-flight.Conclusion Our findings provide valuable insights into the physiological consequences of short-duration spaceflight,with telomere dynamics and immune cell gene expression adapting to spaceflight and persisting after return to Earth.CHIP sequencing data will serve as a reference point for studying the early development of CHIP in astronauts,an understudied phenomenon as previous studies have focused on career astronauts.This study will serve as a reference point for future commercial and non-commercial spaceflight,low Earth orbit(LEO)missions,and deep-space exploration.

syncope is a concerning symptom that affects a large proportion of patients.It can be related to a heterogeneous group of pathologies ranging from trivial causes to diseases with a high risk of sudden death.However,benign causes are the most frequent,and identifying high-risk patients with potentially severe etiologies is crucial to establish an accurate diagnosis,initiate effective therapy,and alter the prognosis.The term cardiac syncope refers to those episodes where the cause of the cerebral hypoperfusion is directly related to a cardiac disorder,while arrhythmic syncope is cardiac syncope specifically due to rhythm disorders.Indeed,arrhythmias are the most common cause of cardiac syncope.Both bradyarrhythmia and tachyarrhythmia can cause a sudden decrease in cardiac output and produce syncope.In this review,we summarized the main guidelines in the management of patients with syncope of presumed arrhythmic origin.Therefore,we presented a thorough approach to syncope work-up through different tests depending on the clinical characteristics of the patients,risk stratification,and the management of syncope in different scenarios such as structural heart disease and channelopathies.

sterile signals generate weaker and delayed macrophage NLRP3 inflammasome responses relative to microbial signals

Inflammation is the host response to microbial infection or sterile injury that aims to eliminate the insult, repair the tissue and restore homeostasis. Macrophages and the NLRP3 inflammasome are key sentinels for both types of insult. Although it is well established that the NLRP3 inflammasome is activated by microbial products and molecules released during sterile injury, it is unclear whether the responses elicited by these different types of signals are distinct. In this study, we used lipopolysaccharide and tumor necrosis factor as prototypical microbial and sterile signal 1 stimuli, respectively, to prime the NLRP3 inflammasome. We then used the bacterial toxin nigericin and a common product released from necrotic cells, ATP, as prototypical microbial and sterile signal 2 stimuli, respectively, to trigger the assembly of the NLRP3 inflammasome complex in mouse and human macrophages. We found that NLRP3 inflammasome responses were weakest when both signal 1 and signal 2 were sterile, but responses were faster and stronger when at least one of the two signals was microbial. Ultimately, the most rapid and potent responses were elicited when both signals were microbial. Together, these data suggest that microbial versus sterile signals are distinct, both kinetically and in magnitude, in their ability to generate inflammasome-dependent responses. This hierarchy of NLRP3 responses to sterile versus microbial stimuli likely reflects the urgent need for the immune system to respond rapidly to the presence of infection to halt pathogen dissemination.

Development of a multiprimer metabarcoding approach to understanding trophic interactions in agroecosystems

To understand trophic interactions and the precise ecological role of each predatory species,it is important to know which arthropod and plant resources are used by generalist predators in agroecosystems.Molecular approaches,such as the use of high-throughput sequencing(HTs),play a key role in identifying these resources.This study develops a multiprimer metabarcoding approach for screening the most common trophic interactions of two predatory arthropods with contrasting morphologies,Rhagonycha fulva(Coleoptera:Cantharidae)and Anthocoris nemoralis(Hemiptera:Anthocoridae)collected from a peach crop.To reduce the time and cost of this metabarcoding approach,we first evaluated the effect of using two different predator-pools of different size(10 and 23 individuals of the same species).We also used our system to analyze the performance of one and two primer pairs in the same library.Our results show that the analysis of 23 individuals together with the use of two primer pairs in the same library optimize the HTs analysis.Using these best-performing conditions,we then analyzed the entire bodies of field-collected predators as well as the washing solutions used to clean the insect bodies.We were able to identify both gut content(i.e.,diet)and external pollen load(i.e.,on the insects’bodies).This study also demonstrates the importance of washing predatory insects’bodies prior to HTs analysis when the target species have a considerable size(>10 mm)and hairy structures.This metabarcoding approach has significant potential for the study of trophic links in agriculture,revealing expected and unexpected trophic relationships.
