Herein we report an electrochemical DNA biosensor for the rapid detection of sequence (5’ AAT GGA TTT ATC TGC TCT TCG 3’) specific for the breast cancer 1 (BRCA1) gene. The proposed electrochemical genosensor is based on short oligonucleotide DNA probe immobilized onto zinc oxide nanowires (ZnONWs) chemically synthesized onto gold electrode via hydrothermal technique. The morphology studies of the ZnONWs, performed by field emission scanning electron microscopy (FESEM), showed that the ZnO nanowires are uniform, highly dense and oriented perpendicularly to the substrate. Recognition event between the DNA probe and the target was investigated by differential pulse voltammetry (DPV) in 0.1 M acetate buffer solution (ABS), pH 7.00;as a result of the hybridization, an oxidation signal was observed at +0.8 V. The influences of pH, target concentration, and non-complimentary DNA on biosensor performance were examined. The proposed DNA biosensor has the ability to detect the target sequence in the range of concentration between 10.0 and 100.0 μM with a detection limit of 3.32 μM. The experimental results demonstrated that the prepared ZnONWs/Au electrodes are suitable platform for the immobilization of DNA.

Identification of thermal front dynamics in the northern Malacca Strait using ROMS 3D-model

The thermal front in the oceanic system is believed to have a significant effect on biological *** an era of climate change,changes in heat regulation between the atmosphere and oceanic interior can alter the characteristics of this important *** the simulation results of the 3D Regional Ocean Modelling System(ROMS),we identified the location of thermal fronts and determined their dynamic variability in the area between the southern Andaman Sea and northern Malacca *** Single Image Edge Detection(SIED)algorithm was used to detect the thermal front from model-derived *** show that a thermal front occurred every year from 2002 to 2012 with the temperature gradient at the location of the front was 0.3°C/*** to the years affected by El Ni?o and negative Indian Ocean Dipole(IOD),the normal years(e.g.,May 2003)show the presence of the thermal front at every selected depth(10,25,50,and 75 m),whereas El Ni?o and negative IOD during 2010 show the presence of the thermal front only at depth of 75 m due to greater warming,leading to the thermocline deepening and enhanced *** May 2003,the thermal front was separated by cooler SST in the southern Andaman Sea and warmer SST in the northern Malacca *** higher SST in the northern Malacca Strait was believed due to the besieged Malacca Strait,which trapped the heat and make it difficult to release while higher chlorophyll a in Malacca Strait is due to the freshwater conduit from nearby rivers(Klang,Langat,Perak,and Selangor).Furthermore,compared to the southern Andaman Sea,the chlorophyll a in the northern Malacca Strait is easier to reach the surface area due to the shallower thermocline,which allows nutrients in the area to reach the surface faster.

Antimicrobial effect of Malaysian vegetables against enteric bacteria

Objective: To investigate the antibacterial activities of green vegetables(pennywort,mint, garlic, parsley and celery) against four common enteric bacteria [Salmonella enterica(ATCC 25957)(S. enterica), Shigella flexneri(ATCC 12022)(S. flexneri),Escherichia coli(ATCC 43889)(E. coli) and Enterobacter cloacae(ATCC 13047)(E. cloacae)] as an alternative medicine for controlling food borne diarrhea disease and the synergistic effect of green vegetables against those ***: Five common vegetables(pennywort, mint, garlic, parsley and celery) were purchased and extracted. The antimicrobial activities of these extracts were tested against four common enteric bacteria(S. enterica, S. flexneri, E. coli and E. cloacae). Ten different concentrations of the extracts(from 640 to 1.25 mg/m L) were prepared and used for the study. The minimal inhibitory concentration(MIC) was determined by the broth dilution method. The antimicrobial activities were assessed by using both well diffusion and disc diffusion ***: Garlic extract showed excellent inhibitory effects on all enteric bacteria. Other plants(parsley, celery, mint and pennywort) were not effective against enteric *** MIC of garlic against S. flexneri and E. cloacae was 40 mg/m L. The MIC of S. enterica and E. coli were 20 and 10 mg/m L, respectively. The performance of the well diffusion method was better than that of the disc diffusion method with clear and sharp inhibition zones of tested bacteria against plant ***: Garlic had excellent antimicrobial effects against enteric bacteria and was recommended to be given to patients with gastroenteritis. The other vegetables(pennywort, mint, parsley and celery) showed no inhibitory effects on enteric bacteria but still can be used for its richness in vitamins and fibers. The performance of the well diffusion method was better than that of the disc diffusion method in detecting the antibacterial effects of green vegetables.
