Bipolaris oryzae is the causal agent of brown spot disease in rice and produces the dark pigment melanin. We isolated and characterized T4HR1 gene encoding 1,3,6,8-tetrahydroxynaphthalene (1,3,6,8-THN) reductase, which converted 1,3,6,8-THN to scytalone in the melanin biosynthesis from B. oryzae. A sequence analysis showed that the T4HR1 gene encoded a putative protein of 268 amino acids showing 50% - 99% sequence identity to other fungal 1,3,6,8-THN reductases. Targeted disruption of the T4HR1 gene showed a different phenotype of mycelial color due to an accumulation of shunt products compared to those of wild-type on PDA plates using tricyclazole as a melanin biosynthesis inhibitor. A quantitative real-time PCR analysis showed that the expression of T4HR1 transcripts was enhanced by near-ultraviolet (NUV) irradiation and regulated by transcriptional factor BMR1, similar to three other melanin biosynthesis genes (polyketide synthase gene [PKS1], scytalone dehydratase gene [SCD1], and 1,3,8-THN reductase gene [THR1]) in the melanin biosynthesis of B. oryzae. These results suggested that common transcriptional mechanisms could regulate the enhanced gene expression of these melanin biosynthesis genes by NUV irradiation in B. oryzae.

Serous tubal intraepithelial carcinoma is a putative precursor of high-grade serous carcinoma, which is the most common histological type of ovarian or pelvic peritoneal cancer. Serous tubal intraepithelial carcinoma is commonly found in patients with breast cancer susceptibility gene mutations who undergo risk-reducing salpingo-oophorectomy. Incidental serous tubal intraepithelial carcinoma found by a non-prophylactic surgery is rare. A 33-year-old woman referred to our hospital for a diagnosis of ectopic pregnancy. She underwent a laparoscopic right salpingectomy. Pathologically, ectopic pregnancy in the ampulla of the right fallopian tube was confirmed and serous tubal intraepithelial carcinoma was observed in the fallopian tube. Subsequently, she underwent a laparoscopic hysterectomy, bilateral oophorectomy, and left salpingectomy as additional treatment. She has experienced no recurrence thus far for 37 months since the surgery.

Proliferative Brenner Tumor with Borderline Mucinous Cystadenocarcinoma of the Ovary in a 75-Year-Old Woman

We report here an extremely rare case of proliferative Brenner tumor with borderline mucinous cystadenocarcinoma of the ovary, metastasizing to the pleura in a 75-year-old woman. Massive pleural effusion were present due to metastasis and pseudo-Meigs’ syndrome. The cytological findings from pleural effusion, ascites and tumor imprint were similar, suggesting mucus-producing malignant tumors. After receiving a thoracic drainage for pleural effusion, the patient underwent an exploratory laparotomy to remove the left-ovarian tumor. Histopathological examination revealed proliferative Brenner ovarian tumor with borderline mucinous cystadenocarcinoma, and immunohistochemical examinations for the tumors, even mucus-rich tumor cells were positive for CK 7 and negative for CK 20, suggesting the tumor arising from transitional cells, not from mucus-producing ovarian surface epithelial tumor cells. She underwent four courses of chemotherapy with paclitaxel and carboplatin. The patient showed no signs of recurrence 16 months after the surgery.

Peak Mating and Breeding Period of the Humpback Whale —(Megaptera novaeangliae) in Okinawa Island, Japan

Although detailed knowledge on breeding ecology of humpback whales is required for developing effective and sustainable whale watching programs in breeding areas, the breeding ecology of this species is still poorly understood. Hence, we examine the migratory timing of humpback whales in Okinawa that is one of their breeding ground, distinguishing the reproductive status (male, female, or female with a calf), group compositions (singleton, pair, or whales more than three) and group types (singer or competitive group) in order to assess the peak period of breeding activities. A total of 1192 days of photo-identification surveys were conducted from 1991 to 2012 and a total of 7366 humpback whales were sighted during the surveys. Among them, 1284 whales were sex-determined (848 males, 147 females and 289 females with a calf), 1138 singletons, 1416 pairs and 710 groups of more than three whales were observed. Females without calves tended to occur from late January to late February, which was the beginning of the breeding season and male-female pairs were observed most frequently during this period. The peak occurrence of competitive group which was considered a mating-related behavior group, formed by females and males, was also observed during this period. These results indicated that humpback whales peak mating period in Okinawa occurred between late January and late February. Females with a calf tended to increase from mid-February toward the end of the breeding season maintaining a high sighting per unit effort (SPUE) value in late March. We, therefore, suggested that the peak time of birthing and newborn care was probably that period in Okinawa. These findings extended our knowledge on the reproductive ecology of humpback whales in Okinawan waters.
