Genetic tra.sforma.ion of cry1a.b) gene into tea.

Tea. (Tectona.gra.dis) provides one of the most highly sought a.ter timber in the world a.d is a.widely recommended species for reforesta.ion. a. such tea. is widely pla.ted in Ma.a.sia. Though no serious outbrea.s ha.e been recorded for tea. in Ma.a.sia. but insect a.ta.k rema.ns the most importa.t threa. to the timber industry. Thus, in efforts to overcome the problem, a. integra.ed pest ma.a.ement system needs to be developed. Spra.ing of commercia. Bt ha. been a.common pra.tice in a.dressing minor outbrea.s. However, one of the ma.n limita.ions of the spra.ing technique is poor covera.e, especia.ly on pla.t surfa.es. Poor covera.e, however, could be overcome by pla.ting insect resista.t trees. In a.dition, the a.proa.h of using genetic engineering in a.dressing the a.ove problem proves to be possible with the a.va.cement ma.e in genetic tra.sforma.ion of trees especia.ly in the la.t deca.e. This, together with improved knowledge on gene function following improved DNa.recombina.t techniques promises the ma.or a.va.cement in pest ma.a.ement of forest species. This report demonstra.es the possibility of tra.sferring foreign gene into tea. cells. In this study, noda. segments of tea. were subjected to pa.ticle bomba.dment. Noda. segments bomba.ded with gold pa.ticles coa.ed with pla.mid DNa.ca.rying hygromycin phosphotra.sfera.e (hpt), β glucuronida.e (gus) a.d cry1a.b) genes were then tra.sferred onto medium for shoot development. The shoots were tha. tra.sferred onto the sa.e medium supplemented with 10mg/L hygromycin for selection. Selection wa. repea.ed severa. times with six week subculture interva.s on the sa.e Hm conta.ning media. The presence of the tra.sgenes in the Hmr pla.ts wa. confirmed using PCR.

a.a.ia.hybrids offer a.grea. potentia. for pa.er industry in Southea.t a.ia.due to their fa.t growth a.d a.ility to grow on a.a.doned or ma.gina. la.ds. Breeding a.a.ia.hybrids with desira.le tra.ts ca. be a.hieved through ma.ker a.sisted selection(Ma.) breeding. To develop a.Ma. progra. requires development of linka.e ma.s a.d QTL a.a.ysis. Two ma.ping popula.ions were developed through interspecific hybridiza.ion for linka.e ma.ping a.d QTL a.a.ysis. a.l seeds per pod were cultured initia.ly to improve hybrid yield a. qua.ity a.d density of linka.e ma.ping is a.fected by the size of the ma.ping popula.ion. Progenies from two ma.ping popula.ions were field pla.ted for phenotypic a.d genotypic eva.ua.ion a. three loca.ions in Ma.a.sia.(1) Forest Resea.ch Institute Ma.a.sia.field sta.ion a. Sega.a., Johor,(2) Borneo Tree Seeds a.d Seedlings Supplies Sdn, Bhd.(BTS) field tria. site a. Bintulu, Sa.a.a., a.d(3) a.ia.rima.RCF field tria. site a. La.ca.g, Pa.a.g. During field pla.ting, misla.eling wa. reported a. Sega.a., Johor, a.d a.simila. problem wa. suspected for Bintulu, Sa.a.a.. Ea.ly screening with two isozymes effectively selected hybrid progenies, a.d these hybrids were subsequently further confirmed by using species-specific SNPs. During field pla.ting, clona. misla.eling wa. reported a.d la.er confirmed by using a.sma.l set of STMS ma.kers. a.la.ge set of SNPs were a.so used to screen a.l ra.ets in both popula.ions. a.tota. of 65.36% misla.eled ra.ets were encountered in the wood density popula.ion a.d 60.34% in the fibre length ma.ping popula.ion. No interpopula.ion pollen conta.ina.ion wa. detected beca.se a.l ra.ets found their ma.ch within the sa.e popula.ion in ***, misla.eling wa. detected a.ong ra.ets of the sa.e popula.ion.
