Metabolomic studies in the inborn error of metabolism alkaptonuria reveal new biotransformations in tyrosine metabolism

Alkaptonuria (AKU) is an inherited disorder of tyrosine metabolism caused by lack of active enzyme homogentisate 1,2-dioxygenase (HGD). The primary consequence of HGD deficiency is increased circulating homogentisic acid (HGA), the main agent in the pathology of AKU disease. Here we report the first metabolomic analysis of AKU homozygous Hgd knockout (Hgd−/−) mice to model the wider metabolic effects of Hgd deletion and the implication for AKU in humans. Untargeted metabolic profiling was performed on urine from Hgd−/− AKU (n = 15) and Hgd+/− non-AKU control (n = 14) mice by liquid chromatography high-resolution time-of-flight mass spectrometry (Experiment 1). The metabolites showing alteration in Hgd−/− were further investigated in AKU mice (n = 18) and patients from the UK National AKU Centre (n = 25) at baseline and after treatment with the HGA-lowering agent nitisinone (Experiment 2). A metabolic flux experiment was carried out after administration of 13C-labelled HGA to Hgd−/−(n = 4) and Hgd+/−(n = 4) mice (Experiment 3) to confirm direct association with HGA. Hgd−/− mice showed the expected increase in HGA, together with unexpected alterations in tyrosine, purine and TCA-cycle pathways. Metabolites with the greatest abundance increases in Hgd−/− were HGA and previously unreported sulfate and glucuronide HGA conjugates, these were decreased in mice and patients on nitisinone and shown to be products from HGA by the 13C-labelled HGA tracer. Our findings reveal that increased HGA in AKU undergoes further metabolism by mainly phase II biotransformations. The data advance our understanding of overall tyrosine metabolism, demonstrating how specific metabolic conditions can elucidate hitherto undiscovered pathways in biochemistry and metabolism.

background Traditional methods for monitoring mining equipment rely primarily on visual inspections,which are time-consuming,inefficient,and *** article introduces a novel approach to monitoring mission-critical systems and services in the mining industry by integrating virtual reality(VR)and digital twin(DT)***-based DTs enable remote equipment monitoring,advanced analysis of machine health,enhanced visualization,and improved decision *** This article presents an architecture for VR-based DT development,including the developmental stages,activities,and stakeholders involved.A case study on the condition monitoring of a conveyor belt using real-time synthetic vibration sensor data was conducted using the proposed *** study demonstrated the application of the methodology in remote monitoring and identified the need for further development for implementation in active mining *** article also discusses interdisciplinarity,choice of tools,computational resources,time and cost,human involvement,user acceptance,frequency of inspection,multiuser environment,potential risks,and applications beyond the mining *** The findings of this study provide a foundation for future research in the domain of VR-based DTs for remote equipment monitoring and a novel application area for VR in mining.

A note on a boundary condition for spouted beds

The boundary condition, zero solids pressure at the top of a particle bed of maximum spoutable height, Hm, is shown to eliminate any resort to empiricism in the derivation of the fluid velocity in the annulus of a spouted bed for which both viscous and inertial effects are taken into account. The same boundary condition fails when applied to a spouted bed for which the bed height H 〈 Hm, especially when H 〈 0.8Hm.

Robson。二者均与短柱金丝桃H. hookerianum Wight & Arn.相近。楚雄金丝桃为一地方特有种,目前仅在模式地点发现;漾濞金丝桃在缅甸东北部还有分布。二者主要以蒴果伸长,非卵状近球形,萼片无中脉而有别于短柱金丝桃。此外,将宽萼金丝桃H. latisepalum (N. Robson) N. Robson从亚种提升为种;将变异较大的纤枝金丝桃H. lagarocladum N. Robson划分为两个亚种,即叶较宽、具有开张习性的原亚种纤枝金丝桃H. lagarocladum ssp. lagarocladum和叶较窄、更为直立的新亚种狭叶金丝桃H. lagarocladum ssp. angustifolium N. Robson。

Inelastic Cotunneling Current and Shot Noise of an Interacting Quantum Dot with Ferromagnetic Correlations

We explore inelastic cotunneling through a strongly Coulomb-blockaded quantum dot attached to twoferromagnetic leads in the weak coupling limit using a generic quantum Langevin equation *** first developa b1och-type equation microscopically to describe the cotunneling-induced spin relaxation dynamics,and then developexplicit analytical expressions for the local magnetization,current,and its *** this basis,we predict a novelzero-bias anomaly of the differential conductance in the absence of a magnetic field for the anti-parallel configuration,and asymmetric peak splitting in a magnetic ***,for the same system with large polarization,we find a negativezero-frequency differential shot noise in the low positive bias-voltage region. All these effects are ascribed to rapidspin-reversal due to underlying spin-flip cotunneling.

The effort and cost required to convert satellite Earth Observation(EO)data into meaningful geophysical variables has prevented the systematic analysis of all available *** overcome these problems,we utilise an integrated High Performance Computing and Data environment to rapidly process,restructure and analyse the Australian Landsat data *** this approach,the EO data are assigned to a common grid framework that spans the full geospatial and temporal extent of the observations–the EO Data *** approach is pixel-based and incorporates geometric and spectral calibration and quality assurance of each Earth surface reflectance *** demonstrate the utility of the approach with rapid time-series mapping of surface water across the entire Australian continent using 27 years of continuous,25 m resolution *** preliminary analysis of the Landsat archive shows how the EO Data Cube can effectively liberate high-resolution EO data from their complex sensor-specific data structures and revolutionise our ability to measure environmental change.

Design and Simulation of IoT Systems Using the Cisco Packet Tracer

Design and implementation of Internet of Things (IoT) systems require platforms with smart things and components. Two dominant architectural approaches for developing IoT systems are mashup-based and model-based approaches. Mashup approaches use existing services and are mainly suitable for less critical, personalized applications. Web development tools are widely used in mashup approaches. Model-based techniques describe a system on a higher level of abstraction, resulting in very expressive modelling of systems. The article uses Cisco packet tracer 7.2 version, which consists of four subcategories of smart things—home, smart city, industrial and power grid, to design an IoT based control system for a fertilizer manufacturing plant. The packet tracer also consists of boards—microcontrollers (MCU-PT), and single boarded computers (SbC-PT), as well as actuators and sensors. The model facilitates flexible communication opportunities among things—machines, databases, and Human Machine Interfaces (HMIs). Implementation of the IoT system brings finer process control as the operating conditions are monitored online and are broadcasted to all stakeholders in real-time for quicker action on deviations. The model developed focuses on three process plants;steam raising, nitric acid, and ammonium nitrate plants. Key process parameters are saturated steam temperature, fuel flowrates, CO and SOb>xb> emissions, converter head temperature, NOb>xb> emissions, neutralisation temperature, solution temperature, and evaporator steam pressure. The parameters need to be monitored in order to ensure quality, safety, and efficiency. Through the Cisco packet tracer platform, a use case, physical layout, network layout, IoT layout, configuration, and simulation interface were developed.
