Profiling neuroprotective potential of trehalose in animal models of neurodegenerative diseases:a systematic review

Trehalose,a unique nonreducing crystalline disaccharide,is a potential disease-modifying treatment for neurodegenerative diseases associated with protein misfolding and aggregation due to aging,intrinsic mutations,or autophagy *** systematic review summarizes the effects of trehalose on its underlying mechanisms in animal models of selected neurodegenerative disorders(tau pathology,synucleinopathy,polyglutamine tract,and motor neuron diseases).All animal studies on neurodegenerative diseases treated with trehalose published in Medline(accessed via EBSCOhost)and Scopus were *** the 2259 studies screened,29 met the eligibility *** to the SYstematic Review Center for Laboratory Animal Experiment(SYRCLE)risk of bias tool,we reported 22 out of 29 studies with a high risk of *** present findings support the purported role of trehalose in autophagic flux and protein *** review identified several other lesser-known pathways,including modifying amyloid precursor protein processing,inhibition of reactive gliosis,the integrity of the blood-brain barrier,activation of growth factors,upregulation of the downstream antioxidant signaling pathway,and protection against mitochondrial *** absence of adverse events and improvements in the outcome parameters were observed in some studies,which supports the transition to human clinical *** is possible to conclude that trehalose exerts its neuroprotective effects through both direct and indirect ***,heterogeneous methodologies and outcome measures across the studies rendered it impossible to derive a definitive *** studies on trehalose would need to clarify three important questions:1)bioavailability with oral administration,2)optimal time window to confer neuroprotective benefits,and 3)optimal dosage to confer neuroprotection.

Background: Parkinson’s disease（PD） is the second most common neurodegenerative disease. Increasing evidence has demonstrated that neuroinflammation is a prominent pathological hallmark of PD. Based on this, modulation of inflammation in the brain can be offered as one of the therapeutic approach to control neuronal damage in PD. Objective: The purpose of this study was to study the effect of the brain preconditioning with single low dose of LPS on inflammatory profile in 6-OHDA model of PD. Methods: Before administration of 6-OHDA, animals in preconditioning group was pretreated with low dose of LPS, while vehicle group rats were preconditioned with normal saline. To clarify the neuroprotective effect of single brain LPS preconditioning, motor coordination and balance were evaluated using rotarod. In addition, the effect of the LPS preconditioning on neuronal damage were observed in the substantia nigra by using nissl staining. Finally, gene and protein expressions associated with different cascades induced by LPS preconditioning including TLR4 and inflammatory signaling pathways were analyzed using RT-PCR and Western Blot. Results: Based on the behavioral assessments, preconditioned animals performed significantly better on the behavioral performance compared to the vehicle group. Furthermore, neuroprotective effect of brain LPS preconditioning were consistent with the histological observation. In addition, our result showed that the brain LPS preconditioning reduced inflammatory response in the brain of PD rat by enhancing NFκB inhibitors and anti-inflammatory cytokines. Conclusion: Generally, reduction in inflammatory response through the brain LPS preconditioning may contribute to the induction of tolerance to neuronal damage in PD. This neuroprotection parallel the reprogramming strategy that leads to the synthesis of new markers to change molecular response against brain lesions. Diabetes mellitus(DM)and Alzheimer's disease(AD)are two major health concerns that have seen a rising prevalence *** studies have indicated a possible link between DM and an increased risk of developing ***,while primarily known for its role in regulating blood sugar,also plays a vital role in protecting brain *** resistance(IR),especially prevalent in type 2 diabetes,is believed to play a significant role in AD's *** insulin signalling becomes dysfunctional,it can negatively affect various brain functions,making individuals more susceptible to AD's defining features,such as the buildup of beta-amyloid plaques and tau protein *** research suggests that addressing insulin-related issues might help reduce or even reverse the brain changes linked to *** review aims to explore the relationship between DM and AD,with a focus on the role of *** also explores the molecular mechanisms by which IR might lead to brain changes and assesses current treatments that target *** IR's role in the connection between DM and AD offers new possibilities for treatments and highlights the importance of continued research in this interdisciplinary field.

A Practical Study of Intelligent Image-Based Mobile Robot for Tracking Colored Objects

Object tracking is one of the major tasks for mobile robots in many real-world ***,artificial intelligence and automatic control techniques play an important role in enhancing the performance of mobile robot *** contrast to previous simulation studies,this paper presents a new intelligent mobile robot for accomplishing multi-tasks by tracking red-green-blue(RGB)colored objects in a real experimental ***,a practical smart controller is developed based on adaptive fuzzy logic and custom proportional-integral-derivative(PID)schemes to achieve accurate tracking results,considering robot command delay and tolerance *** design of developed controllers implies some motion rules to mimic the knowledge of experienced *** scenarios of three colored object combinations have been successfully tested and evaluated by using the developed controlled image-based robot *** PID control failed to handle some tracking scenarios in this *** proposed adaptive fuzzy PID control achieved the best accurate results with the minimum average final error of 13.8 cm to reach the colored targets,while our designed custom PID control is efficient in saving both average time and traveling distance of 6.6 s and 14.3 cm,*** promising results demonstrate the feasibility of applying our developed image-based robotic system in a colored object-tracking environment to reduce human workloads.

Gastroesophageal reflux after per-oral endoscopic myotomy:Management literature

In this editorial,we respond to a review article by Nabi et al,in which the authors discussed gastroesophageal reflux(GER)following peroral endoscopic myotomy(POEM).POEM is presently the primary therapeutic option for achalasia,which is both safe and effective.A few adverse effects were documented after POEM,including *** diagnostic criteria were not clear enough because approximately 60%of patients have a long acid exposure time,while only 10%experience reflux *** predictors of high disease incidence have been identified,including old age,female sex,obesity,and a baseline lower esophageal sphincter pressure of less than 45 *** technical steps during the procedure,such as a lengthy or full-thickness myotomy,may further enhance the *** pump inhibitors are currently the first line of *** voices are increasingly advocating for the routine combining of POEM with an endoscopic fundoplication method,such as peroral endoscopic fundoplication or transoral incisionless ***,more research is necessary to determine the safety and effectiveness of these procedures in the long term for patients who have undergone them.

Thermal performance of domed roof in air-conditioned spaces

Due to their thermal performance,domed roofs are one of the passive solutions that affect energy consumption in *** thermal performance of domed roofs has been investigated in many naturally ventilated ***,few studies have discussed their performance in conditioned ***,this study introduces a computational comparison between domed and flat roofs to investigate their impact on thermal comfort inside a conditioned *** an earlier stage,field measurements were carried out inside a Bahraini mosque to acquire its indoor air conditions during the summer period of 2021,in addition to validating the computational *** findings of this study confirm that,under mechanical cooling conditions,the flat roof offers a lower indoor temperature than the domed roof by 0.4℃and 0.1℃for open and closed doors,***,the air velocity is lower by approximately 0.01 m/s for both door *** overall PMV values of the flat roof are also lower by 0.07 and 0.01,while the PPD values are lower by 0.20,and 0.34 for open and closed doors,*** on these small differences,it can be concluded that the thermal performance of both roofing systems behaves equally in conditioned ***,the air patterns are substantially different,the overall thermal performance is *** similarity drives building designers to rethink the thermal performance of the domed roofs in air-conditioned spaces with such a hot climate,regardless of their aesthetic and acoustical behaviour.

Emerging pollutants,such as antibiotics and antibiotic-resistance genes,are becoming increasingly important sources of safety and health *** water safety,which is closely related to human health,should receive more attention than natural water body ***,minimal research has been performed on the efficacy of existing treatment processes in water treatment plants for the removal of antibiotics and antibiotic resistance *** address this research gap,this study detected and analyzed six main antibiotics and nine antibiotic resistance genes in the treatment processes of two drinking water plants in *** were collected over three months and then detected and analyzed using ultra-high-performance liquid chromatography-tandem mass spectrometry and fluorescence *** total concentrations of antibiotics and antibiotic resistance genes in the influent water of the two water plants were characterized as December>March>*** precipitation and filtration processes of the Zou Maling Water Plant and Yu Shidun Water Plant successfully removed the *** ozone-activated carbon process increased the removal rate of most antibiotics to 100%.However,a large amount of antibiotic resistance gene residues remained in the effluents of the two water *** experiments demonstrated that the existing ozone-activated carbon processes could not effectively remove antibiotic resistance *** study provides a reference for the optimization of drinking water treatment processes for antibiotics and antibiotic resistance gene removal.

Quantification of Silicon (SI) and Silicon Dioxide (SiO₂) from the Nafud Desert-Al-Qassim Region, Kingdom of Saudi Arabia Using XRD Analysis

This research was conducted in the Qassim region, Kingdom of Saudi Arabia. The goal of this research is to determine the percentage of silicon in the Rub al-Khali desert. Samples were collected from four cities located in the Al-Qassim Region of Saudi Arabia (Uyun Al-Jawa, Al-Fuwailiq, Al-Sulaibiya, and Al-Qawara), from three distinct depths (the earth’s surface, 50 cm, and 100 cm). The percentages of silicon in these places vary between the highest value for silicon dioxide, which is 74.47 m/m%, and 34.8 m/m% for silicon in Al-Qawara city at a depth of 100 cm. We used an x-ray fluorescence (XRF) instrument to evaluate the samples. There are high percentage of both silica and silicon in the Nafud desert. Studies have shown that these ratios can help investors benefit from an element of Silicon and silicon dioxide, so the sands of the Nafud desert is sufficient for extracting Silicon and silica in huge quantities. This may transform the Kingdom into a leading country in the global computer technology industry that depends on silicon extracted from the desert sands, the most important of which are microcomputer data processing devices.
