Forensic profiling of non-volatile organic compounds in soil using ultra-performance liquid chromatography:a pilot study

Soil is of particular interest to the forensic community because it can be used as valuable associative evidence to link a suspect to a victim or a crime *** chromatography is a powerful analytical tool for organic compound ***,high-performance liquid chromatography(HPLC)has proven to be an efficient method for forensic soil analysis,especially in discriminating soils from proximity ***,ultra-performance liquid chromatography(UPLC),which is much more sensitive than HPLC,has never been explored in this *** study proposed a UPLC method for profiling non-volatile organic compounds in three Malaysian soils(red,brown and yellowish-brown soils).The three soils were analysed separately to assess the effects of individual chromatographic parameters:(a)elution programme(isocratic *** gradient programmes);(b)flow rate(0.1 vs.0.2 mL/min);(c)extraction solvent(acetonitrile ***)and(d)detection wavelength(230 vs.254 nm).The injection volume and total run time were set to 5μL and 35 min,***,each soil sample gave 24 different *** showed that the most desirable chromatographic parameters were(a)isocratic elution;(b)flow rate at 0.2 mL/min and(c)acetonitrile extraction *** proposed UPLC system is expected to be a feasible method for profiling non-volatile organic compounds in soil,and is more chemical-efficient than a comparable HPLC system.

Objective:To address the lack of research on invasive group B Streptococcus(GBS)infections in Malaysia and Southeast Asia through a comprehensive analysis of GBS isolates obtained from ***:Medical records from patients with GBS infection isolated from the sterile site,such as blood and cerebrospinal fluid from 14 July 2019 to 15 December 2020,were reviewed from six major hospitals in Peninsular *** criteria were invasive GBS,sterile sites and non-repeated GBS isolated from the same patients in the same *** isolates were re-identified for GBS and ***:A total of 118 patients were eligible,with a majority of non-pregnant adults(76.3%).Over half of the patients(62.7%)had underlying medical conditions,with diabetes as the most common disease,followed by respiratory disease,renal disease,cardiovascular disease and skin and soft tissue *** most common manifestations were sepsis,followed by soft tissue abscess,diabetic foot ulcer,wet gangrene and *** overall mortality was 7.6%.The most common serotype was serotype桋.Conclusions:Invasive GBS infection among non-pregnant adults showed a rising trend,particularly among diabetic *** study underscores the importance of reducing risk factors and highlights the necessity of developing GBS vaccination as a preventive strategy for both infants and adults.

Proapoptotic activities of Oroxylum indicum leave extract in HeLa cells

Objective: To examine the proapoptotic properties of Oroxylum indicum methanol extract on cervical cancer cells. Methods: Methylene blue assay was used to determine the IC50 value of the extract. Western blotting assays were done to analyze the expression of HPV oncoproteins (HPV18 E6 and E7) and apoptotic molecules (caspase-3 and caspase-8). Reverse transcriptase PCR assays were performed to determine genetic alteration of tumor suppressors p53 and pRb and apoptosis markers Fas and FasL. Enzyme-linked immunosorbent assay (ELISA) was done to determine the expression of cytokine levels (IL-6 and IL-12). Results: The determination of IC50 value indicated a higher anti-proliferative activity of the extract compared to cisplatin. After 24 hours of treatment, Western blot analysis showed that treated HeLa cells exhibited a significant down-regulation of HPV18 oncoproteins E6 and E7, and a significant induction of caspase-8 and caspase-3 activation level. Meanwhile, the mRNA expressions of p53, pRb, Fas and FasL were significantly upregulated in treated cells. Moreover, ELISA showed an increased IL-12 and decreased IL-6 production after Oroxylum indicum treatment. Conclusions: The methanol extract of Oroxylum indicum has an anti-proliferative activity and proapoptotic potential. It induces localized-immunity improvements by altering cytokine production in HPV-positive cervical cancer cells.

Implementation of biofloc technology(BFT)system in aquaculture industry shows high productivity,low feed conversion ratio,and an optimum culture *** study was divided into two *** first phase involved maintaining the water quality using the optimum carbon-to-nitrogen ratio by manipulating pH in culture *** second phase examined the performance of harvesting biofloc(remaining phytoplankton and suspended solids in the system)using chicken eggshell powder(CESP).This study showed that pH 7 to 8 were the best biofloc performance with high removal percentage of ammonia(>99%)with a remaining ammonia concentration of 0.016 mg L^(−1)and 0.018 mg L^(−1),*** second phase of this study was performed to determine the optimal formulation and conditions of using CESP as a bio-flocculant in harvesting excess *** use of eggshell showed a higher harvesting efficiency of more than 80%under the following treatment conditions:0.25 g L^(−1)of eggshell dosage;with rapid and slow mixing rates of 150 and 30 rpm,respectively;30 min of settling time;settling velocity of 0.39 mm s^(−1)and pH of 6 to ***,the results indicated that biofloc would be the best green technology approach for sustainable aquaculture wastewater and the CESP is an organic matrix that environmental-friendly bio-coagulant for biofloc harvesting.

The protective effects of procyanidin C-1 on bisphenol A-induced testicular dysfunction in aged mice

The number and quality of sperm are decreased due to bisphenol A(BPA) exposure, an endocrine-disrupting chemical on the male reproductive system, especially in advanced paternal age(APA). Procyanidin C-1(PCY-1), an antioxidant from grape seed(Vitis vinifera L.), has demonstrated anti-viral, anti-melanogenic and immunostimulatory effects. Therefore, this study aims to determine the effects of PCY-1 intervention on sperm parameters, testis morphological changes, serum testosterone, oestradiol, and luteinizing hormone(LH) concentrations and the expression of apoptotic(Bax and Bcl-2) and mitochondria-related(Mfn1 and Opa1) genes in BPA-exposed aged mice. Results revealed that PCY-1 intervention improves aged male fertility in BPA-exposed conditions by decreasing abnormal sperms percentage and increasing spermatogenic cell diameter and epithelial height. PCY-1 also decreased oestradiol, and increased LH and testosterone levels. The gene expression of Bax was significantly down-regulated by PCY-1 intervention. In contrast, Bcl-2 was substantially up-regulated. Expression of Mfn1 and Opa1 genes were also significantly up-regulated in the PCY intervention group. Hence, it is demonstrated that PCY-1 was able to mitigate the adverse effects of BPA on reproductive parameters of aged mice. Collectively, we postulated that PCY-1 has a potential role in protecting the ageing male reproductive system against the damaging impacts of BPA.

Antibacterial activity of zinc oxide nanoparticles(Zn O-NPs) has received significant interest worldwide particularly by the implementation of nanotechnology to synthesize particles in the nanometer region. Many microorganisms exist in the range from hundreds of nanometers to tens of micrometers. Zn O-NPs exhibit attractive antibacterial properties due to increased specific surface area as the reduced particle size leading to enhanced particle surface reactivity. Zn O is a bio-safe material that possesses photo-oxidizing and photocatalysis impacts on chemical and biological species. This review covered Zn O-NPs antibacterial activity including testing methods, impact of UV illumination, Zn O particle properties(size, concentration, morphology, and defects), particle surface modification, and minimum inhibitory concentration. Particular emphasize was given to bactericidal and bacteriostatic mechanisms with focus on generation of reactive oxygen species(ROS) including hydrogen peroxide(H2O2), OH-(hydroxyl radicals), and O2-2(peroxide). ROS has been a major factor for several mechanisms including cell wall damage due to Zn O-localized interaction, enhanced membrane permeability, internalization of NPs due to loss of proton motive force and uptake of toxic dissolved zinc *** have led to mitochondria weakness, intracellular outflow, and release in gene expression of oxidative stress which caused eventual cell growth inhibition and cell death. In some cases, enhanced antibacterial activity can be attributed to surface defects on Zn O abrasive surface texture. One functional application of the Zn O antibacterial bioactivity was discussed in food packaging industry where Zn O-NPs are used as an antibacterial agent toward foodborne diseases. Objective: To determine the etiologies of tropical acute febrile illness(TAFI) in West Pahang, Malaysia and to investigate morbidity and mortality factors in relation to TAFI. Methods: A multicenter prospective cohort study was conducted between January and June 2016 in six district hospitals throughout the western part of Pahang State in Peninsular Malaysia. A total of 336 patients answered a standardized questionnaire and blood samples were collected for laboratory confirmation of infectious etiology. Descriptive analysis and logistic regression were performed to identify factors associated with TAFI. Results: A total of 336 patients were included. The patients were mainly Malays(70.2%), males(61.3%), aged(44.6±17.4) years, with more than half(58.9%) presenting with gastrointestinal symptoms. The majority were diagnosed with dengue(35.7%) while malaria(4.5%) was the least frequent. The in-hospital mortality due to TAFI was 9.2%. Patients with meliodosis had five times higher mortality [Adjusted OR: 5.002, 95% CI:(1.233, 20.286)]. Patients with comorbidities such as cardiovascular symptoms(P<0.001) and renal replacement therapy initiation(P<0.001) were significantly associated with in-hospital mortality in all TAFI. Conclusions: The etiology of TAFI in the western Pahang includes dengue, leptospirosis, malaria and melioidosis, which carry the highest risk of in-hospital mortality. The presence of cardiovascular symptoms may be used to assess the disease severity in TAFI, but more studies are needed in the future.
