Is the pain in chronic pancreatitis of neuropathic origin? Support from EEG studies during experimental pain

AIM: To prove the hypothesis that patients with chronic pancreatitis would show increased theta activity during painful visceral stimulation. METHODS: Eight patients and 12 healthy controls underwent an experiment where the esophagus was electrically stimulated at the pain threshold using a nasal endoscope. The electroencephalogram (EEG) was recorded from 64 surface electrodes and "topographic matching pursuit" was used to extract the EEG information in the early brain activation after stimulation. RESULTS: A major difference between controls and patients were seen in delta and theta bands, whereas there were only minor differences in other frequency bands. In the theta band, the patients showed higher activity than controls persisting throughout the 450 ms of analysis with synchronous brain activation betweenthe channels. The main theta components oscillated with 4.4 Hz in the patients and 5.5 Hz in the controls. The energy in the delta (0.5-3.5 Hz) band was higher in the controls, whereas the patients only showed scattered activity in this band. CONCLUSION: The differences in the theta band indicate that neuropathic pain mechanisms are involved in chronic pancreatitis. This has important implications for the understanding and treatment of pain in these patients, which should be directed against drugs with effects on neuropathic pain disorders.

Inhibitory effects of methanolic Olea europaea and acetonic Acacia laeta on growth of Babesia and Theileria

Objective:To evaluate the antipiroplasmic activities of methanolic extract of Olea europaea(MOE)and acetonic extract of Acacia laeta(AAL)against Babesia and Theileria parasites in vitro and evaluate the chemotherapeutic effects of these extracts against Babesia(B.)microti in ***:Fluorescence assay using SYBR Green 1 nucleic acid stain was used to detect inhibitory effects of the two extracts as well as the combination effects of the two extracts with diminazene aceturate and atovaquone on four Babesia species and Theileria equi in vitro while for in vivo experiments,8-weekold female BALB/c mice were injected intraperitoneally with 1× 107 ***-iRBCs and treated orally at a dose of 150 mg/kg of both ***:The half maximal inhibitory concentration(IC50)values of AAL against ***,***,***,***,and Theileria equi were lower than those of MOE *** assay on Madin-Darby bovine kidney,mouse embryonic fibroblast(NIH/3T3),and human foreskin fibroblast cell lines showed that MOE and AAL affected only the viability of Madin-Darby bovine kidney cell line with half maximal effective concentrations(EC50)of(794.7±41.9)and(873.9±17.5)μg/mL,*** oral treatments of MOE and AAL at 150 mg/kg inhibited the growth of *** in mice by 80.4% and 64.4%,*** MOE and diminazene aceturate combination showed a higher chemotherapeutic effect than that of ***:MOE and AAL have the potential to be an alternative remedy for treating ***,the combination therapy of MOE + DA was more potent against *** infection in mice than their monotherapies.

Economic Impact of Higher Timber Truck Loads on Louisiana Bridges

Due to the limited amount of funds available for bridge inspection, maintenance and rehabilitation, the evaluation of load capacity for existing bridges is crucial to the Louisiana Department of Transportation and Development. This paper includes the development of a methodology to assess the economic impact of overweight vehicles with permits, hauling Louisiana harvest products on state bridges. The proposed higher truck loads are applied on the existing bridges and their effects are determined using deterministic load capacity evaluations as well as reliability assessments. The target rehability level is derived from bridge structures designed to satisfy AASHTO Standard Design Specifications and also satisfy safe and adequate performance levels. The amount of harvest produced is used to select a representative sample of bridges to provide specific examples of expected changes in load ratings and safety levels. The bridges include simple and continuous span behavior. Strength and serviceability criteria are investigated under current legal loads and the expected changes, due to the proposed new weights, are determined. The results are used to assess the cost of crossing a bridge and the permit fees for the proposed truck weight regulation.

Role of AXL in invasion and drug resistance of colon and breast cancer cells and its association with p53 alterations

To characterize AXL receptor tyrosine kinase (AXL) expression in relationship to tumor protein P53 (TP53 gene, p53 protein) and its role in tumor invasion and response to *** used 14 cell lines, including 3 isogenic pairs carrying mutant/knockout p53, to gain insight into the relationship between AXL and TP53. These included HCT116, HCT116.p53 mutant, RKO, and RKO.p53-/- lines (all from colon cancers) as well as breast cancer cell lines MCF7 and 1001 (MCF7-p53 mutant clone). HeLa cell line was used as a positive control for epithelial to mesenchymal transition (EMT). AXL expression was determined by Western blotting using rabbit monoclonal antibody clone C89E7. AXL siRNA silencing was performed and followed by collagen invasion assay. Cell viability analysis using the sulforhodamine B assay and the invasion assay were performed after exposure to chemotherapeutic agents (doxorubicin for breast cancer cells; 5FU or irinotecan for colon cancer cells).RESULTSWe showed that the introduction of p53 mutations or knockout increased expression levels of AXL in isogenic cells compared to the matching p53 wild-type parental cells. Overall, we found a trend for correlation between the potential EMT candidate AXL, p53 alterations, and EMT markers in colorectal and breast cancers. The expression of AXL in RKO cells, a rare colon cancer cell line with inactive Wnt signaling, suggests that the AXL oncogene might provide an alternative genetic pathway for colorectal carcinogenesis in the absence of Wnt signaling activation and TP53 mutation. AXL silencing in the TP53 mutant isogenic cell lines 1001, HCT116.p53 mutant and RKO.P53-/- was > 95% efficient and the silenced cells were less invasive compared to the parental TP53 wild-type cells. AXL silencing showed a subtle trend to restore colon cancer cell sensitivity to 5FU or irinotecan. SOX9 in biliary atresia: New insight for fibrosis progression

Brucellosis: Pathophysiology and new promising treatments with medicinal plants and natural antioxidants

Brucellosis is an old, infectious and common zoonosis whose causative agents are Gramnegative bacteria from the Brucella genus. Brucellosis is transmitted through direct contactwith infected animals or using unpasteurized dairy products of goats, pigs, camels, sheep,buffalo and cows. Brucellosis is still the most common zoonosis in the world, with mostof cases occurring in developing countries. Today, an approach to traditional medicine andmedicinal plants, especially with regards to the repeated recommendations of the World HealthOrganization, is a necessity. One-third of chemical drugs are produced by using plants andthere is a high potential to produce more drugs from plants. Medicinal plants are helpful inthe management of various conditions, especially bacterial diseases. Although there is notenough scientific evidence regarding the clinical effectiveness of herbal drugs for the treatmentof brucellosis, there is strong evidence on the antimicrobial effects of herbal drugs to preventinfection. Therefore, this article seeks to describe the antibacterial effects of some plantderived essential oils or extracts, so that they can serve as promising choices to develop newanti-Brucella medications, as suitable alternatives to conventional antibiotics for brucellosis, asmuch as possible, taking into account the benefits of these herbal drugs.

Skeletal extension rate of the reef building coral <i>Porites</i>species from Aqaba and their environmental variables

Annual skeletal extension rates of the sclera-actinian corals Porites species were investigated in 32 colonies from the northern Gulf of Aqaba fringing reef at various depths (1 - 42 m). All corals reveal clear and regular skeletal density banding patterns. Results showed that the high-density annual growth bands were formed during winter and the low-density annual growth bands during summer. The mean annual extension rates of the studied corals reveal a large inter-colony variability with values ranged between 2.36 to 20.0 mm/year. While a general trend of decreasing coral extension rate with depth was observed and best explained by a simple exponential model, the rates clustered into two groups: 10.86 ± 2.54 mm/year in water depths less than 10 m, and 5.23 ± 1.99 mm/year below 12 m. Light intensity seems to be the primary environmental factor responsible for decreasing coral extension rate with depth since the effect of other environmental parameters could be neglected from the Gulf of Aqaba. Time series record of the mean annual coral extension rate showed a slight increasing linear trend which could be linked to increase seawater temperature over the period of time represented.
