Metabolically healthy obesity refers to obese individuals who do not develop metabolic *** people store fat in subcutaneous adipose tissue(SAT)rather than in visceral adipose tissue(VAT).However,the molecules participating in this specific scenario remain ***18,a lipid droplet(LD)-associated protein,mediates the contact between the endoplasmic reticulum(ER)and LDs to facilitate LD growth and *** the present study,we show that the protein level of Rab18 is specifically upregulated in the SAT of obese people and ***18 adipocyte-specific knockout(Rab18 AKO)mice had a decreased volume ratio of SAT to VAT compared with wildtype *** subjected to high-fat diet(HFD),Rab18 AKO mice had increased ER stress and inflammation,reduced adiponectin,and decreased triacylglycerol(TAG)accumulation in *** contrast,TAG accumulation in VAT,brown adipose tissue(BAT)or liver of Rab18AKO mice had a moderate increase without ER stress ***18 AKO mice developed insulin resistance and systematic ***18 AKO mice maintained body temperature in response to acute and chronic cold induction with a thermogenic SAT,similar to the counterpart ***,Rab18-deficient 3T3-L1 adipocytes were more prone to palmitate-induced ER stress,indicating the involvement of Rab18 in alleviating lipid ***18 AKO mice provide a good animal model to investigate metabolic disorders such as impaired *** conclusion,our studies reveal that Rab18 is a key and specific regulator that maintains the proper functions of SAT by alleviating lipid-induced ER stress.

揭示生物体内在的调控机制是生物信息学的一项重要研究内容.各种高通量生物数据的涌现,为从基因组的尺度上重构基因调控网络提供了可能.由于单数据源仅能提供关于调控关系的片面信息且存在噪声,因此整合多种生物学数据的方法有望得到可靠性较高的调控网络.提出了一种综合ChIP-chip数据、knock out(敲除)数据和各种条件下的表达谱数据来推断调控关系的新方法.ChIP-chip数据和knock out数据能分别提供转录因子和目标基因对关系的直接物理结合和功能关系的证据,这两类数据的整合有望获得较高的识别准确率.但这两类数据的重合性通常较低,基于共调控的基因通常具有较高的表达相似性这一假设,在一定程度上降低了这两类数据重合性较低所带来的影响.算法所识别的大部分调控关系都被YEASTRACT,高质量ChIP-chip数据和文献所验证,从而证明了该方法在调控关系的预测上具有较高的准确性.与其他方法的比较,也表明了该方法具有较高的预测性能.

A synergistic Pd/Zn dual chiral catalyst system has been developed for the stereodivergent transformation of Morita–Baylis–Hillman(MBH)carbonates and unprotectedα-hydroxyketones to afford the corresponding allylation products(>50 examples).These products bearing vicinal stereocenters were obtained in high yields with excellent regio-/diastereo-/*** cyclization in an acidic medium,the allylation products could transform to the corresponding acyl-functionalizedβ,γ-disubstitutedα-methylene-γ-butyrolactones(AMGBLs).All four stereoisomers of AMGBL were readily accessed using the same starting materials with full control of the stereochemistry of the contiguous stereogenic centers,simply by switching the chiral ligand combination for the Pd/Zn system.A nucleophilic site-divergent reactivity was observed for the allylation of primary and secondaryα-hydroxyketones,suggesting that distinct enolate intermediates might have prevailed in these reactions.

Prostate cancer(PCa)is a common malignant tumor with high morbidity and mortality *** prostate cancer stem cell(PCSC)model provides novel insights into the pathogenesis of PCa and its therapeutic ***,the roles and molecular mechanisms of specific genes in mediating fate decisions of PCSCs and carcinogenesis of PCa remain to be *** this study,we have explored the expression,function,and mechanism of AZGP1P2,a pseudogene of AZGP1,in regulating the stemness and apoptosis of PCSCs and treatment resistance of docetaxel in castration-resistant prostate cancer(CRPC).We revealed that AZGP1P2 was downregulated in CRPC cell lines and PCSCs,while it was positively associated with progression-free *** of the AZGP1P2 enhanced the sensitivity of docetaxel treatment in CRPCs via inhibiting their *** pull-down associated with mass spectrometry analysis,co-immunoprecipitation assay,and RNA immunoprecipitation assay demonstrated that AZGP1P2 could bind to UBA1 and RBM15 as a“writer”of methyltransferase to form a ***1,an E1 ubiquitin-activating enzyme,contributed to RBM15 protein degradation via ubiquitination *** RNA immunoprecipitation assay displayed that RBM15 controlled the mRNA decay of TPM1 in m6A ***,a xenograft mouse model and patient-derived organoids showed that the therapeutic effect of docetaxel in CRPC was increased by AZGP1P2 in ***,these results imply that AZGP1P2 mediates the stemness and apoptosis of PCSCs and promotes docetaxel therapeutic effect by suppressing tumor growth and metastasis via UBA1/RBM15-mediated TPM1 mRNA decay in CRPC.

Large-scale structure(LSS)surveys will increasingly provide stringent constraints on our cosmological ***,the density-marked correlation function(MCF)has been introduced,offering an easily computable density-correlation *** have demonstrated that MCFs offer additional,independent constraints on cosmological models beyond the standard two-point correlation(2PCF).In this study,we apply MCFs for the first time to SDSS CMASS data,aiming to investigate the statistical information regarding clustering and anisotropy properties in the Universe and assess the performance of various weighting schemes in MCFs,and finally obtain constraints onΩ_(m).Upon analyzing the CMASS data,we observe that,by combining different weights(α=[-0.2,0,0.2,0.6]),the MCFs provide a tight and independent constraint on the cosmological parameterΩ_(m),yieldingΩ_(m)=0.293±0.006 at the 1σlevel,which represents a significant reduction in the statistical error by a factor of 3.4 compared to that from *** constraint is consistent with recent findings from the small-scale clustering of BOSS galaxies(Zhai et ***.J.948,99(2023))within the 1σ***,we also find that our estimate is lower than the Planck measurements by about 2.6σ,indicating the potential presence of new physics beyond the standard cosmological model if all the systematics are fully *** method outlined in this study can be extended to other surveys and datasets,allowing for the constraint of other cosmological ***,it serves as a valuable tool for forthcoming emulator analysis on the Chinese Space Station Telescope(CSST).

Pulse duration is considered as one of the most important characteristics of high-power femtosecond ***,pulses output from the laser system are susceptible to ambient changes and manifest the instability of pulse durations in an open *** this paper,incorporating the algorithmic framework of the improved stochastic hill-climbing search and incremental proportional-integral controller,temperature-induced fluctuations of pulse duration can be effectively compensated by an automatic feedback control in an all-fiber chirped-pulse amplification *** the experiment,sub-hundred femtosecond fluctuation of pulse duration is introduced to verify the performance robustness of the proposed pulse-duration feedback control(PDFC).The stability of pulse duration is obviously higher than the case without the feedback control,and the peak-to-peak fluctuation of pulse duration is reduced to 6.5%.Furthermore,the robust switching between different pulse durations proves the versatility of the *** expect that the proposed feedback control method could provide a novel insight into high-power femtosecond lasers widely applied in fundamental researches and industrial fields.

Adoptive transfer of FTY720-treated immature bone marrowderived dendritic cells(BMDCs) significantly reduced the spontaneous resorption rate in the CBA/J ×DBA/2 mouse model

Objective To investigate the effect of FTY720-treated immature bone marrow-derived dendritic cells（BMDCs） on the embryo resorption rate in the CBA/J× DBA/2 abortion mouse *** The dendritic cells（DCs） were derived from bone marrow of DBA/2 mice, and then co-cultured with FTY720. The abortion mouse models were established by mating female CBA/J mice with DBA/2 mice. Via the CBA/J×DBA/2 abortion mouse model, six groups were established, group A： normal pregnancy model; group B： abortion mouse model with no treatment; group C： abortion mouse model injected with DC culture medium（DCCM）; group D： abortion mouse model injected with DC; group E： abortion mouse model injected with FTY720; group F： abortion model mouse injected with FTY720-DC. The differences were compared in the embryo resorption rates of the CBA/J ×DBA/2 abortion mouse model treated with FTY720-DC or different controls observed on gestation day 12 to 14, and then the microenvironment in murine pregnancy was *** The embryo resorption rate was statistically significantly decreased in group D and group E when they compared with group B and group C（P〈0.05, respectively）.Furthermore, the embryo resorption rate in group F showed a statistically significant decrease when compared with the other groups except group A（P〈0.01）. These resultssuggest that FTY720-DCs possess a notable advantage over DCs or FTY720 in reducing the embryo resorption rate of the abortion mouse model. The percentage of Th17（IL-17＋CD4＋T cells） in peripheral blood mononuclear cell（PBMC） in the abortion mouse model was 4.35%±0.34% before treated with FTY720-DC, and was1.34%±0.28% after treated with FTY720-DC（P〈0.01）. The percentage of Tregs（CD4~＋CD25~＋Foxp3~＋T cells） in PBMC was significantly increased in group F（8.35%±1.80%） as compared with group B（2.68%±0.65%）（P〈0.01）.Conclusion Adoptive transfer of FTY720-DC can statistically significantly reduce the embryo resorption rate in the CBA/J×DBA/2 abortion mouse model.

Mechanism of Ta on the microstructure and properties of Fe-Mn damping alloys

Fe-Mn-Co-xTa alloys（x=0,1,2,3 wt.%） were fabricated using spark plasma sintering to explore the influence of Ta content on the microstructure,mechanical properties,and damping characteristics of Fe-17Mn-3Co damping *** micro structural analysis reveals the dominant presence of γ-austenite,ε-martensite,α'-martensite,and Co3Ta precipitates within the *** increments in Ta content exhibit proportional enhancements in hardness,tensile strength,and fracture elongation of the ***,these improvements in mechanical attributes are closely correlated with the generated second-phase *** the Ta content increases,there is a gradual augmentation in the formation of Co3Ta within the ***,the damping property initially experiences an ascent followed by a subsequent decline with rising Ta *** damping behavior finds its origins in multiple mechanisms,including grain boundary damping stemming from reduced grain size,interfacial damping induced by the generated Co3Ta,and dislocation *** is noteworthy that the emergence of the secondary phase wields a dual impact on damping *** the one hand,the proliferation of the second phase fosters increased interfaces for energy *** the other hand,these interfaces give rise to compressive stresses,constraining interface movement and thus impeding damping property enhancement.
