Clinical Impact of t(14;18) in Diffuse Large B-cell Lymphoma

Objective： Recent studies have suggested that t（14;18） is present in a significant proportion of diffuse large B-cell lymphomas （DLBCLs）. However, the prognostic significance of this translocation and its relationship with BCL-2 protein expression remains controversial. Our study aimed to investigate the predictive power of t（14;18） and BCL-2 protein expression in the prognosis of DLBCLs. Methods： Biopsy specimens from 106 DLBCLs were analyzed using interphase fluorescence in situ hybridization （FISH）. Immunophenotypic analysis of CD20, CD3, CD10, BCL-6, MUM1 and BCL-2 was performed by immunohistochemistry. SPSS 13.0 software was used for statistical analysis. Results： The t（14;18） was identified in 27 of 106 cases （25.5%）. The percentages of tumor cells expressing CD10, BCL-6, MUM1 and BCL-2 were 21.7%, 26.4%, 56.6% and 73.6%, respectively. The presence of this translocation was significantly correlated with the expression of CD10 and immunophenotypic subtype （p0.001）. No association was observed between BCL-2 protein expression and the presence of t（14;18）. Multivariate analysis confirmed that both t（14;18） and BCL-2 expression were significantly associated with survival. Moreover, patients with t（14;18） had worse prognosis, compared with those with BCL-2 expression （for overall survival： hazard ratio, 4.235; 95%CI, 2.153-8.329, p0.001 vs. hazard ration, 2.743; 95%CI, 1.262-5.962, p=0.011）. Conclusions： The t（14;18） is a useful prognostic tool for the evaluation of DLBCL immunophenotype and prognosis. The prognosis of GCB （germinal centre-like B cell） DLBCL patients should be made with the consideration of the presence of this translocation, and the detection of t（14;18） should be included as a routine diagnostic test in these cases.

The efficient extraction of uranium, as the primary component of nuclear energy, holds significant implications. Drawing inspiration from the charge interaction observed in biological ion channels, we encapsulated negatively charged polystyrene sulfonate(PSS) or sodium polystyrene carboxylate(PVBA) into the nanochannels of amidoxime functionalized covalent organic framework(COF-AO) in-situ to alter the cavity environment of COF-AO. The synthesized COF-AO-PSS and COF-AO-PVBA are used for ultra-fast and highly selective uranium recovery. The negatively charged PSS/PVBA was confined in the COF-AO channel providing the driving force for uranium transport and blocking other ions, thus creating a highly selective “uranium highway”. Additionally, introducing sulfonate groups or carboxyl groups into COF-AO offers supplementary coordination environments and weak interactions with uranium. Due to charge-assisted migration and various interaction mechanisms, both COF-AO-PSS and COF-AO-PVBA exhibit faster adsorption kinetics and higher selectivity compared to COF-AO alone. Their adsorption capacities are 3.8 times and 2.4 times that of COF-AO alone respectively which highlights the necessity for constructing biomimetic ion channels in uranium adsorption processes. This work presents a bionic adsorbent based on covalent organic frameworks(COFs) for the first time, overcoming environmental and equipment limitations associated with traditional photocatalysis and electrocatalysis methods for uranium capture, opening up new avenues for designing multifunctional materials that mimic biological systems.

Combinatorial optimization(CO)on graphs is a classic topic that has been extensively studied across many scientific and industrial ***,solving CO problems on graphs through learning methods has attracted great *** deep learning methods,e.g.,graph neural networks(GNNs),have been used to effectively assist the process of solving ***,current frameworks based on GNNs are mainly designed for certain CO problems,thereby failing to consider their transferable and generalizable abilities among different COs on ***,simply using original graphs to model COs only captures the direct correlations among objects,which does not consider the mathematical logicality and properties of *** this paper,we propose a unified pre-training and adaptation framework for COs on graphs with the help of the maximum satisfiability(Max-SAT)*** first use Max-SAT to bridge different COs on graphs since they can be converted to Max-SAT problems represented by standard formulas and clauses with logical *** we further design a pre-training and domain adaptation framework to extract the transferable and generalizable features so that different COs can benefit from *** the pre-training stage,Max-SAT instances are generated to initialize the parameters of the *** the fine-tuning stage,instances from CO and Max-SAT problems are used for adaptation so that the transferable ability can be further *** experiments on several datasets show that features extracted by our framework exhibit superior transferability and Max-SAT can boost the ability to solve COs on graphs.

Effects of surface energy and substrate on modulus determination of biological films by conical indentation

Micro-/nano-indentation has become prevalent in evaluating the mechanical characteristics of biological samples,such as cells and ***,the existing contact models describing conical indentation ignore the joint effects of surface energy and substrate,and consequently cannot accurately extract the Young's modulus of biological samples deposited on *** finite element methods,we examine the conical indentation of biological films on substrates while taking surface energy into *** on the dimensional analysis,the explicit relationship between load and indentation depth is achieved for films with their moduli varying from 0.001 to 100 times that of the *** the classical Sneddon's model was employed to analyze the load-depth data,the measured modulus could reach 18 times the real modulus for films on harder substrates,but only 4%of the real modulus for films on softer ***,in micro-/nano-indentations,neglecting the contribution of surface energy would result in an overestimation of the Young's modulus of films depending on the contact *** analytical expression provided here can be utilized to precisely deduce the mechanical characteristics of biological films deposited on substrate from the load and indentation depth data of a conical indentation.

The topological pumping has been instrumental in advancing our understanding of topological phase transitions in various physical systems,which can be extended to uncover intriguing higher-order topological phases in the lower-dimensional ***,we propose a theoretical exploration of topological dipole pumping on an acoustic square superlattice by cyclically modulating intracell couplings,which shares the topological nature of an extended three-dimensional system with chiral hinge *** the multipole chiral numbers,we characterize the higher-order topological phases that arise during the *** evolution of topological phase transitions is verified by numerical simulations and shows corner states are transferred across the *** findings can inspire the construction of chiral hinge states in artificial crystals,opening up new possibilities for the design of devices allowing the unidirectional propagation of sound.

This paper introduces an improvement to electrochemical drilling process by coupling flow field and electric field in pulsating state. A novel tube with half-wedged shape at the end(HW-tube) is prepared, with both sidewall and wedged part of the HW-tube insulated. Only the flat part is utilized to provide electric field for electrochemical drilling. By rotating the HW-tube, both flow field and electric field in pulsating state are generated, alternating in different positions within the inter-electrode gap(IEG). The pulsating flow field enhances the mass transfer process, while pulsating electric field disperses material dissolution process and distribution of electrolytic byproducts. Both pulsating fields are coupled at the same frequency, further enhancing the electrochemical drilling process. Simulation results indicate that both flow field and electric field in pulsating state are generated. Compared to the traditional tube, the HW-tube significantly reduces the number of residual particles in IEG, and this number is further reduced by increasing the rotation speed. Experimental results reveal that the surface quality and dimensional uniformity of small hole are improved with HW-tube. With feed rate of 2.22 mm/min, a small hole with diameter of 1.52± 0.017 mm is drilled, resulting in a surface roughness of 0.331 μm.

OBJECTIVE To examine the effect of Pin1 on the expression and bioactivity of MMP-9 through NF-kB in human colorectal carcinoma SW480 cells. METHODS The eukaryotic expression vector of RNA interfering （shRNA） with the Pin1 gene （pGenesil-1-PIN1） was constructed in our previous experiments and was confirmed through sequencing. Cell motility was tested through the wound healing assay and the Boyden chamber assay. The protein levels and bioactivity of MMP-9 were tested by Western blotting and gelatin zymography in SW480 cells after transfection with pGenesil-l-PIN1 （SW480/ p-shRNA）. The DNA-binding activity of NF-kB in cells transfected with pGenesil-1-PIN1 was analyzed by the electrophoretic mobility shift assay （EMSA）. In addition, to determine whether NF-kB has direct interaction with the MMP-9 promoter derived from the genomic DNA of SW480 cells transfected with pGenesil- 1-PIN1, oligonucleotides containing a putative NF-kB binding site were synthesized and EMSAs were performed. RESULTS The results of the Boyden chamber assay showed that cell motility was reduced from 90.2 ± 6.5 per field （x 10 objective） to 49.6± 7.2 per field （P 〈 0.05, Student＇s t-test） for SW480 cells transfected with pGenesil-1-PIN1 （SW480/p-shRNA）. Western blotting detected low protein levels of Pin1 and MMP-9 in SW480/ p-shRNA cells. The relative protein levels of Pin1 were 0.49 ± 0.07 in SW480/p-shRNA compared with 0.94 ± 0.09 in SW480/p-Con, and MMP-9 were 0.45 ± 0.07 in SW480/p-shRNA, 0.83 ± 0.07 in SW480/p-Con （P 〈 0.05）. The results of gelatin zymography showed that silencing Pin1 markedly reduced the bioactivity of MMP-9 in SW480 cells. EMSA results revealed low DNA-binding activity of NF-kB in SW480/p-shRNA cells compared to SW480/ p-Con cells, and that NF-kB bound directly to the oligonucleotides which contained putative NF-kB binding sites in the MMP-9 promoter derived from the genomic DNA of SW480/p-shRNA cells. Thermally conductive polymer nanocomposites integrated with lightweight,excellent flexural strength,and high fracture toughness(KIc)would be of great use in many ***,achieving all of these properties simultaneously remains a great *** by natural nacre,here we demonstrate a lightweight,strong,tough,and thermally conductive boron nitride nanosheet/epoxy layered(BNNEL)*** of the layered structure and enhancing the interfacial interactions through hydrogen bonding and Si–O–B covalent bonding,the resulting nacre-inspired BNNEL nanocomposites show high fracture toughness of~4.22 MPa·m^(1/2),which is 7 folds as high as pure ***,the BNNEL nanocomposites demonstrate sufficient flexural strength(~168.90 MPa,comparable to epoxy resin),while also being lightweight(~1.23 g/cm^(3)).Additionally,the BNNEL nanocomposites display a thermal conductivity(κ)of~0.47 W/(m·K)at low boron nitride nanosheet loading of 2.08 vol.%,which is 2.7 times higher than that of pure epoxy *** developed nacre-inspired strategy of layered structure design and interfacial enhancement provides an avenue for fabricating high mechanical properties and thermally conductive polymer nanocomposites.
