Polystyrene nanoplastics induce glycolipid metabolism disorder via NF-κB and MAPK signaling pathway in mice

Nanoplastics-induced developmental and reproductive toxicity,neurotoxicity and immunotoxicity are a focus of widespread attention.However,the effects of nanoplastics(NPs)on glycolipid metabolism and the precise underlying mechanisms are unclear at present.Here,we showed that oral administration of polystyrene nanoparticles(PS-NPs)disrupts glycolipid metabolism,with reactive oxygen species(ROS)identified as a potential key signaling molecule.After PS-NPs treatment,excessive production of ROS induced the infammatory response and activated the antioxidant pathway through nuclear factor-erythroid factor 2-related factor 2.The activation of nuclear factor-κB(NFκB)signaling pathway induced the phosphorylation of the mitogen-activated protein kinases(MAPK)signaling pathway,which induced the activation of extracellular regulated kinases(ERK)and p38.Constitutive activation of the MAPK signaling proteins induced high continued phosphorylation of insulin receptor substrate-1,in turn,leading to decreased protein kinase B(Akt)activity,which weakened the sensitivity of liver cells to insulin signals and induced insulin resistance.In parallel,phosphorylation of Akt led to loss of control of Fo XO1,a key gene of gluconeogenesis,activating transcription of glucose-6-phosphatase(G6PC)and phosphoenolpyruvate carboxykinase(PEPCK)in a manner dependent on PGC1α.Moreover,the activated ERK promoted lipid accumulation through ERK-PPARγcascades.Therefore,sterol regulatory element-binding protein-1 and levels of its downstream lipogenic enzymes,ACC-1,were up-regulated.Upon treatment with the antioxidant resveratrol,PS-NPs-induced glucose and lipid metabolic disorders were improved by inhibiting ROS-induced activation of NFκB and MAPK signaling pathway in mice.Based on above,PS-NPs exposure disrupts glycolipid metabolism in mice,with ROS identified as a potential key signaling molecule.

Effect of guanxin-V Mixture Combined with Sacubitril Valsartan on Cardiac Function after PCI in STEMI Patients

[Objectives]To observe the effect of guanxin-V Mixture combined with Sacubitril Valsartan on cardiac function in patients after PCI for acute ST-segment elevation myocardial infarction(STE-MI).[Methods]41 cases of STEMI patients(qi and yin deficiency and blood stasis and obstruction)hospitalized in Nanjing Hospital of Chinese Medicine affiliated to Nanjing University of Chinese Medicine from January 2020 to June 2021 were randomly divided into 21 cases in the treatment group and 20 cases in the control group,and the two groups were given standardized Western medicine treatment as soon as possible after PCI.The control group was treated with Sacubitril Valsartan,and the treatment group was treated with guanxin-V Mixture on the basis of treatment in the control group.The patients in the two groups were treated for 3 months,and the TCM syndrome score,left ventricular ejection fraction(LVEF),and N-Terminal Pro-Brain Natriuretic Peptide(NT-proBNP),interleukin-6(IL-6),and high-sensitivity C-reactive protein(hs-CRP)levels,and the incidence of heart failure and adverse reactions in the two groups after treatment were recorded.[Results]After the treatment,the TCM syndrome score and serum NT-proBNP,IL-6 and hs-CRP levels of the two groups significantly decreased(P0.05).[Conclusions]guanxin-V Mixture combined with Sacubitril Valsartan could significantly improve cardiac function in STEMI patients undergoing PCI,and its effect may be related to the suppression of inflammatory response.

Effect of fore/aft-loaded rotor on compressor stability under inlet circumferential distortion

A modified small perturbation stability prediction model for axial compressors with circumferential inlet distortions is established and applied to investigate the effect of fore/aft-loaded rotor on compressor stability under circumferentially distorted inlet conditions.The inlet total pressure distribution downstream of the distortion screen is measured in experiments and employed for simulations which are implemented via time-space spectral method.The stall inception prediction results via the stability model indicate that the compressor with aft-loaded rotor not only performs better in terms of stability under uniform inlet,but also maintains a larger stability margin under circumferentially distorted inlet.The experiments for compressors with fore-loaded and aft-loaded rotor are respectively carried out.The results validate the reliability of numerical simulations and the predicted conclusion that the aft-loaded rotor is beneficial for compressor stability.Besides,the ability of the developed theoretical model for compressor stability prediction under circumferential distortions is confirmed.In addition,dynamic pressure signals at rotor tip measured in experiments illustrate that the circumferential distortion has little effect on the compressor stall pattern.

A Methodology for Assessing Axial Compressor Stability with Inlet Temperature Ramp Distortion

Based on a small perturbation stability model for periodic flow,the effects of inlet total temperature ramp distortion on the axial compressor are investigated and the compressor stability is quantitatively evaluated.In the beginning,a small perturbation stability model for the periodic flow in compressors is proposed,referring to the governing equations of the Harmonic Balance Method.This stability model is validated on a single-stage low-speed compressor TA36 with uniform inlet flow.Then,the unsteady flow of TA36 with different inlet total temperature ramps and constant back pressure is simulated based on the Harmonic Balance Method.Based on these simulations,the compressor stability is analyzed using the proposed small perturbation model.Further,the Dynamic Mode Decomposition method is employed to accurately extract pressure oscillations.The two parameters of the temperature ramp,ramp rate and Strouhal number,are discussed in this paper.The results indicate the occurrence and extension of hysteresis loops in the rows,and a decrease in compressor stability with increasing ramp rate.Compressor performance is divided into two phases,stable and limit,based on the ramp rate.Furthermore,the model predictions suggest that a decrease in period length and an increase in Strouhal number lead to improved compressor stability.The DMD results imply that for compressors with inlet temperature ramp distortion,the increase of high-order modes and oscillations at the rotor tip is always the signal of decreasing stability.

Multi-cohort comprehensive analysis unveiling the clinical value and therapeutic effect of GNAL in glioma

Clinical data indicates that glioma patients have poor treatment outcomes and clinical prognosis.The role of olfactory signaling pathway-related genes(OSPRGs)in glioma has not been fully elucidated.In this study,we aimed to investigate the role and relationship between OSPRGs and glioma.Univariate and multivariate Cox regression analyses were performed to assess the relationship between OSPRGs and the overall survival of glioma based on public cohorts,and the target gene(G Protein Subunit Alpha L,GNAL)was screened.The association of GNAL expression with clinicopathological characteristics,gene mutation landscape,tumor immune microenvironment(TIME),deoxyribonucleic acid(DNA)methylation,and naris-occlusion controlled genes(NOCGs)was performed.Immunohistochemistry was used to evaluate GNAL level in glioma.Further analysis was conducted to evaluate the drug sensitivity,immunotherapy response,and functional enrichment of GNAL.GNAL was an independent prognostic factor,and patients with low GNAL expression have a poor prognosis.Expression of GNAL was closely associated with clinicopathological characteristics,DNA methylation,and several immune-related pathways.Immune infiltration analysis indicated that GNAL levels were negatively correlated with immune scores.GNAL low-expression group showed efficacy with anti-PD-1 therapy.Ten compounds with significantly different half-maximal inhibitory concentration(IC50)values between the GNAL high and low-expression groups were identified.Furthermore,its expression was associated with several immune cells,immune-related genes,and NOCGs.The expression of GNAL is closely associated with clinicopathological characteristics,TIME,and the response to therapeutic interventions,highlighting its potential as a prognostic biomarker for glioma.

Numerical Simulation on Production Trials by Using Depressurization for Typical Marine Hydrate Reservoirs:Well Type and Formation Dip

Natural gas hydrate has huge reserves and is widely distributed in marine environment.Its commercial development is of great significance for alleviating the contradiction between energy supply and demand.As an efficient research method,numerical simulation can provide valuable insights for the design and optimization of hydrate development.However,most of the current production models simplify the reservoir as a two-dimensional(2D)horizontal layered model,often ignoring the impact of formation dip angle.To improve the accuracy of production prediction and provide theoretical support for the optimization of production well design,two three-dimensional(3D)geological models with different dip angles based on the geological data from two typical sites are constructed.The vertical well,horizontal well and multilateral wells are deployed in these reservoirs with different permeabilities to perform production trial,and the sensitivity analysis of dip angles is also carried out.The short-term production behaviors in high and low permeability reservoirs with different dip angles are exhibited.The simulation results show that 1)the gas and water production behaviors for different well types in the two typical reservoirs show obviously different variation laws when the short-term depressurization is conducted in the inclined formation;2)the inclined formation will reduce the gas production and increase the water extraction,and the phenomena becomes pronounced as the dip angle increases,particularly in the low-permeability reservoirs;3)and the impact of formation dip on hydrate recovery does not change significantly with the variation of well type.

With unique physicochemical properties and biological effects,magnetic nanomaterials(MNMs)play a crucial role in the biomedical field.In particular,magnetic iron oxide nanoparticles(MIONPs)are approved by the United States Food and Drug Administration(FDA)for clinical applications at present due to their low toxicity,biocompatibility,and biodegradability.Despite the unarguable effectiveness,massive space for improving such materials'performance still needs to be filled.Recently,many efforts have been devoted to improving the preparation methods based on the materials'biosafety.Besides,researchers have successfully.regulated the performance of magnetic nanoparticles(MNPs)by changing their sizes,morphologies,compositions;or by.aggregating as-synthesized MNPs in an orderly arrangement to meet various clinical requirements.The rise of cloud computing and artificial intelligence techniques provides novel ways for fast material characterization,automated data analysis,and mechanism demonstration.In this review,we summarized the studies that focused on the preparation routes and performance regulations of high-quality MNPs,and their special properties applied in biomedical detection,diagnosis,and treatment.At the same time,the future.development of MNMs was also discussed.

Versatile oxidized variants derived from TMDs by various oxidation strategies and their applications

The physicochemical properties of transition metal dichalcogenides(TMDs)are highly related to their structures and usually stable in air.However,under certain conditions they could be transformed into different structures due to oxidation.Considering this,various materials with fascinating structures have been explored by oxidation strategies,which possess novel properties and great potential in various applications such as solar batteries,hydrogen evolution reaction(HER)catalysts,and field effect transistors(FET).In this review,we systematically summarize the atomic structures of TMD oxidized variants and the corresponding fabrication approaches.Utilizing various characterization methods,the chemical components of TMD oxidized variants are illustrated.Furthermore,we expound the promising applications of the oxidized variants.This review is expected to provide a new insight for preparing precise materials at the atomic level through corresponding oxidation strategies.

Ozone Sterilizer for Treatment and Health Care

Ozone is a green broad-spectrum bactericidal disinfectant, and a trace amount of ozone in the atmosphere makes many viruses and bacteria lose their biochemical activity and infectivity. Nature produces trace amounts of ozone in the air through lightning to purify the ecological environment. The product of ozone decomposition is oxygen, without secondary pollution. Ozone sterilizer is widely used in the epidemic prevention and control of intensive breeding farms and achieved remarkable results. If the concentration and action time of ozone can be accurately controlled, then ozone can quickly eliminate pathogens, without harming the normal cells in the human body. How to use medical ozone for epidemic prevention, treatment and health care is a subject worthy of serious study, which should arouse the attention of the experts in the field.
