Octreotide induces caspase activation and apoptosis in human hepatoma HepG2 cells

AIM: To investigate the role of octreotide on cellular proliferation and apoptosis of human hepatoma (HepG2) cells. METHODS: We studied cellular proliferation, apoptosis and the possible internal caspase-mediated apoptosis pathway involved, after treatment of HepG2 carcinoma cells with octreotide in comparison with the apoptosis caused by tumor necrosis factor-α (TNF-α). Activities of caspase-3, caspase-9, caspase-8 and caspase-2 were studied, while apoptosis was investigated through detection of DNA fragmentation and through identification of apoptotic cells with the annexin-V/propidium iodide flow cytometric method. RESULTS: After an initial increase in HepG2 cellular proliferation, a significant inhibition was observed with 10-8 mol/L octreotide, while TNF-α dose-dependently decreased proliferation. Early and late apoptosis was significantly increased with both substances. Octreotide significantly increased caspase-3, caspase-8 and caspase-2 activity. TNF-α signifi cantly increased only caspase-2. Cellular proliferation was decreased after treatment with octreotide or TNF-α alone but, in contrast to TNF-α, octreotide decreased proliferation only at concentrations of 10-8 mol/L, while lower concentrations increased proliferation. CONCLUSION: Our findings are suggestive of caspasemediated signaling pathways of octreotide antitumor activity in HepG2 cells, and indicate that measurements of serum octreotide levels may be important, at least in clinical trials, to verify optimal therapeutic drug concentrations.

Overview of Gait Synthesis for the Humanoid COMAN

This paper focuses on the developments of a generic gait synthesis for the humanoid robot COMAN. Relying on the essential Gait Pattern Generator （GPG）, the proposed synthesis offers enhanced versatilities for the locomotion under different purposes, and also provides the data storage and communication mechanisms among different modules. As an outcome, we are able to augment new abilities for COMAN by integrating new control modules and software tools at a cost of very few modifications. Moreover, foot placement optimization is introduced to the GPG to optimize the gait parameter references in order to meet the robot＇s natural dynamics and kinematics, which enhances the synthesis＇s robustness while it＇s being implemented on real robots. We have also presented a practical approach to generate pelvis motion from CoM references using a simplified three-point-mass model, as well as a straightforward but effective idea for the state estimation using the sensory feedback. Three physical experiments were studied in an increasing complexity to demonstrate the effectiveness and successful implementation of the proposed gait synthesis on a real humanoid system.

Tropospheric Scattering(Troposcatter) and Tropospheric Ducting are two different mechanisms due to in-homogenoueties in the lower part of the Earths *** common influence in propagation of microwaves is studied here in order to achieve a feasible naval communication *** not new,Troposcatter together with Tropospheric Ducting communications are regaining popularity in current military and civil applications such as isolated islands and oil extraction facilities in the open *** innovation proposed here is the application of such systems in naval communications where at least one of the stations is *** propagation together with fading models are simulated using appropriate programs and scientific simulation packets in order to predict maximum range of such communication systems in each dominant mode of *** appropriate antenna design proposals are given to overcome the high propagation loss and minimize possible interferences.

This paper advocates the use of the distributed compressed sensing(DCS)paradigm to deploy energy harvesting(EH)Internet of Thing(IoT)devices for energy *** consider networks with signal/energy models that capture the fact that both the collected signals and the harvested energy of different devices can exhibit *** provide theoretical analysis on the performance of both the classical compressive sensing(CS)approach and the proposed distributed CS(DCS)-based approach to data acquisition for EH ***,we perform an in-depth comparison of the proposed DCSbased approach against the distributed source coding(DSC)*** performance characterizations and comparisons embody the effect of various system phenomena and parameters including signal correlation,EH correlation,network size,and energy availability *** results unveil that,the proposed approach offers significant increase in data gathering capability with respect to the CS-based approach,and offers a substantial reduction of the mean-squared error distortion with respect to the DSC system.

Growth of Niobium Thin Films on Si Substrates by Pulsed Nd:YAG Laser Deposition

The growth of Nb thin films on Si（100） substrates by different laser fluences （4-15 j/cm2） was reported, The pulsed Nd：YAG laser deposition （PLD） under influence of laser fluence on ablation rate and deposition rate was discussed. X-ray diffraction （XRD） investigations of the deposited films showed an amorphous structure. The droplet density on the film surface observed by scanning electron microscopy （SEM） analyses was extremely low. It was experimentally proved that the droplets on the film surface originated from liquid phase on the target surface, Profilometric measurements of the deposited Nb films revealed a substantial asymmetry in the film thickness related to the plume deflection effect. The measured electrical resistivity of the Nb film was higher than that of high purity Nb bulk. The present investigations of ablation and deposition process of Nb thin films are related to its potential application in superconducting radio-frequency （SRF） cavities.

The pandemic of coronavirus disease 2019(COVID-19),caused by a newly identifiedβ-coronavirus(SARS-CoV-2)has emerged as a dire health problem,causing a massive crisis for global *** method of transmission was firstly thought to be animal to human ***,it has been observed that the virus is transmitted from human to human via respiratory ***,SARS-CoV-2 ribonucleic acid(RNA)has been isolated from patient stools,suggesting a possible gastrointestinal(GI)*** commonly reported clinical manifestations are fever,fatigue and dry ***,a small percentage of patients experience GI symptoms with the most common being anorexia,diarrhea,nausea and *** presence of viral RNA in stools is also common and fecal tests can be positive even after negative respiratory *** exact incidence of digestive symptoms is a matter of *** distribution of Angiotensin converting enzyme type 2 receptors in multiple organs in the body provides a possible explanation for the digestive symptoms’*** with solely GI symptoms have been reported in both adults and *** RNA has also been detected in stool and blood samples,indicating the possibility of liver damage,which has been reported in COVID-19 *** presence of chronic liver disease appears to be a risk factor for severe complications and a poorer prognosis,however data from these cases is *** aim of this review is firstly,to briefly update what is known about the origin and the transmission of SARS-CoV-2,but mainly to focus on the manifestations of the GI tract and their pathophysiological background,so that physicians on the one hand,not to underestimate or disregard digestive symptoms due to the small number of patients exhibiting exclusively this symptomatology and on the other,to have SARS-CoV-2 on their mind when the“gastroenteritis”type symptoms predominate.

目的 :对种植体与原厂基台及第三方基台连接组件形态学上的微观特征进行观察,探讨形态差异与潜在并发症之间可能的关系。方法:3种基台(1个原厂装,2个来自第三方)在规定扭矩下装配到Straumann RN种植体上,种植体基台连接单元经过一系列包埋等程序后切割形成剖面,对剖面进行打磨,对完成的试件进行扫描电镜观察。采用图像测量软件(Image j)对获得的图片进行校准测量,比较三者之间在接触长度及接触性质之间的差异,尤其是3个重要部位即肩部、内连接及螺纹处的差异。结果:3个连接单元的所有被研究的接触面之间均存在一定的尺寸差异。3个单元中,种植体外肩部的紧密接触相似,但第三方基台内部连接的啮合,特别是抗旋转单元严重欠缺,其中1个第三方基台的螺丝啮合不良。结论:与原装基台对比,第三方基台呈现出关键性的形态差异,横断面的几何学差异导致总体接触面积有很大不同。无论从质量上,还是数量上这种差异对修复体的长期稳定性均有可能产生严重影响。

Myeloid sarcoma as a differential diagnosis of small bowel obstruction

We are presenting the case of a 42-year-old ?male patient, who was hospitalized due to an acute small bowel obstruction caused by a tissue mass of the mesentery. The patient reported that he had a history of a testicular tumour. For therapy of intestinal obstruction as well as for diagnostic reasons we decided to perform ?an explorative laparotomy. On histopathological examination the immunohistological staining was positive for myeloperoxidase (MPO) and KP-1 (CD68). Staining was slightly positive for Bcl-2, CD117, CD34, but negative for CD3, CD4, CD5, CD8, CD20, CD30, CD79, Bcl-6 and S-100. This leads to the diagnosis of a myeloid sarcoma. After recovery from surgery and chemotherapy, allogenic bone marrow transplantation was performed. Most intestinal obstructions are caused by postoperative adhesions or hernias and only in rare cases caused by a myeloid sarcoma.
