Tumor cell death visualization of renal cell carcinoma under the combined effect of the Gratiola officinalis extract and cyclophosphamide using fluorescent staining methods

Objective of the study:We used fluorescence imaging methods of apoptosis and necrosis in human renal carcinoma A498 tumor cells in vitro to reveal the indicated forms of cell death under the combined effect of flavonoid-containing extract of Gratiola officinalis and cytostatic(cyclo-phosphamide).Materials and methods:The dyes were propidium iodide and acridine orange,which were used in the"alive and dead"*** test helped us to identify the total number of dead cells in the forms of necrosis and apoptosis and the number of cells in which apoptosis had started,it was characterized by the appearance of apoptotic bodies or nucleus ***:We found the most pronounced cytotoxic activity at the ratio of extract of Gratiola officinalis and cyclophosphamide concentrations of 1∶*** number of living cells decreased when exposed to the ratio of extract and cytostatic concentrations of 2∶*** the ratio of concentration of the extract relative to the cytostatic increased to 3∶1,the cytostatic activity of the extract began to appear,the total number of tumor cells *** number of cells with nucleus pyknosis and the number of cells with apoptosis signs significantly increased at a 3∶1 ratio of extract and cytostatic concentrations,which confirms the presence of pro-apoptotic activity of the studied *** trend indicates the dependence of a certain form of cell death(apoptosis,necrosis)on the ratio of extract and cytostatic doses,and it also demon-strates the cytostatic and cytotoxic effects of this ***:Fluorescence methods of investigation in the"alive and dead"test allowed us to visualize the forms of cell death of human kidney carcinoma A498 by combined exposure to the fiavonoid-containing extract of Gratiola officinalis and cytostatic(cyclophosphamide)24 h after *** found that the combination with a concentration ratio of the extract and cyclophosphamide of 3:1 has the greatest effectiveness due to stimulation of t

Normothermic ex vivo lung perfusion(NEVLP)has emerged as a modernized organ preservation technique that allows for detailed assessment of donor lung function prior to *** main goal of this study was to identify potential biomarkers of lung function and/or injury during a prolonged(19 h)NEVLP procedure using in vivo solid-phase microextraction(SPME)technology followed by liquid chromatography-high resolution mass spectrometry(LC-HRMS).The use of minimally invasive in vivo SPME fibers for repeated sampling of biological tissue permits the monitoring and evaluation of biochemical changes and alterations in the metabolomic profile of the *** in vivo SPME fibers were directly introduced into the lung and were also used to extract metabolites(on-site SPME)from fresh perfusate samples collected alongside lung samplings.A subsequent goal of the study was to assess the feasibility of SPME as an in vivo method in metabolomics studies,in comparison to the traditional inlab metabolomics *** upregulated biochemical pathways involved in pro-and antiinflammatory responses,as well as lipid metabolism,were observed during extended lung perfusion,especially between the 11th and 12th hours of the procedure,in both lung and perfusate ***,several unstable and/or short-lived metabolites,such as neuroprostanes,have been extracted from lung tissue in vivo using SPME ***-site monitoring of the metabolomic profiles of both lung tissues through in vivo SPME and perfusate samples on site throughout the prolonged NEVLP procedure can be effectively performed using in vivo SPME technology.

Comprehensive DDoS Attack Classification Using Machine LearningAlgorithms

The fast development of Internet technologies ignited the growthof techniques for information security that protect data, networks, systems,and applications from various threats. There are many types of threats. Thededicated denial of service attack (DDoS) is one of the most serious andwidespread attacks on Internet resources. This attack is intended to paralyzethe victim’s system and cause the service to fail. This work is devoted tothe classification of DDoS attacks in the special network environment calledSoftware-Defined Networking (SDN) using machine learning algorithms. Theanalyzed dataset included instances of two classes: benign and *** the dataset contained twenty-two features, the feature selection techniques were required for dimensionality reduction. In these experiments, theInformation gain, the Chi-square, and the F-test were applied to decreasethe number of features to ten. The classes were also not completely balanced, so undersampling, oversampling, and synthetic minority oversampling(SMOTE) techniques were used to balance classes equally. The previousresearch works observed the classification of DDoS attacks applying variousfeature selection techniques and one or more machine learning ***, they did not pay much attention to classifying the combinations offeature selection and balancing methods with different machine learningalgorithms. This work is devoted to the classification of datasets with eightmachine learning algorithms: naïve Bayes, logistic regression, support vectormachine, k-nearest neighbors, decision tree, random forest, XGBoost, andCatBoost. In the experimental results, the Information gain and F-test featureselection methods achieved better performance with all eight ML algorithmsthan with the Chi-square technique. Furthermore, the accuracy values of theoversampled and SMOTE datasets were higher than that of the undersampledand imbalanced datasets. Highly selective/enantioselective Pt-ReOx/C catalyst for hydrogenation of L-malic acid at mild conditions

The catalyst preparation strategy was based on a strict introduction sequence of rhenium and platinum precursors and their strong interaction with carbon support resulted in the formation of 0.5 nm Pt-Re Ox species of atomic dispersion, where platinum is metallic, while monolayer rhenium is partially oxidized（Re2＋）. The reaction kinetics was studied taking into account the process of L-malic acid association leading to the formation of inactive cyclic oligomeric species. High TOFs（ca. 50 h-1）, selectivities（ca. 99%）and stability of Pt-Re Ox/C catalysts in aqueous-phase hydrogenation of L-malic acid, which are close to those of the homogeneous pincer type complexes, were revealed at mild conditions（T = 90–130 ℃）. Taking into account that（i） hydrogenation reaction occurred 2–3 orders of magnitude faster than its racemization and（ii） association of L-malic acid dominates at low temperatures and in a concentrated solution,special reaction conditions that allow obtaining chemically and optically（ee 〉 99%） pure（S）-3-hydroxy-γ-butyrolactone and（S）-1,2,4-butanetriol were found. Basing on HAADF-STEM, EDX, XPS, and kinetic studies, the structure of active species and basic reaction pathways are proposed.

Machine learning prediction of methane,ethane,and propane solubility in pure water and electrolyte solutions:Implications for stray gas migration modeling

Hydraulic fracturing is an effective technology for hydrocarbon extraction from unconventional shale and tight gas reservoirs.A potential risk of hydraulic fracturing is the upward migration of stray gas from the deep subsurface to shallow *** stray gas can dissolve in groundwater leading to chemical and biological reactions,which could negatively affect groundwater quality and contribute to atmospheric *** knowledge oflight hydrocarbon solubility in the aqueous environment is essential for the numerical modelling offlow and transport in the ***,we compiled a database containing 2129experimental data of methane,ethane,and propane solubility in pure water and various electrolyte solutions over wide ranges of operating temperature and *** machine learning algorithms,namely regression tree(RT)and boosted regression tree(BRT)tuned with a Bayesian optimization algorithm(BO)were employed to determine the solubility of *** predictions were compared with the experimental data as well as four well-established thermodynamic *** analysis shows that the BRT-BO is sufficiently accurate,and the predicted values agree well with those obtained from the thermodynamic *** coefficient of determination(R2)between experimental and predicted values is 0.99 and the mean squared error(MSE)is 9.97×10^(-8).The leverage statistical approach further confirmed the validity of the model developed.
