Immunohistochemical localization of glutathione S-transferase-pi in human colorectal polyps

AIM:To investigate the distribution of the placental form of glutathione-S-transferase (GST) in colon polyps in order to evaluate the role of GST-pi in these tissues. METHODS: Sixteen polyp tissues removed at colonoscopy were examined. Tissues were investigated histologically and ultrastructurally. GST-pi expression was also analysed immunohistochemically, using peroxidase anti-peroxidase (PAP) method and immunogold labelling method, for light and electron microscope respectively. RESULTS: All polyp tissues examined were adenoma of low, mild and high-grade dysplasia as shown in the histopathological reports. Nevertheless, the examination of the above specimens with electron microscope revealed that 3 of 9 adenoma of mild dysplasia had ultrastuctural features similar to high-grade dysplasia adenoma. GST-pi was variably expressed in adenoma, with the lowest relative levels occurring in low-gradeadenoma and the highest levels found in high-grade adenoma. GST-pi was located mainly in undifferentiated epithelial cells. GST-pi positive particles were found in the cytoplasm and especially in the nucleus adjacent to the nuclear membrane of these cells. CONCLUSION:The overexpression of GST-pi in mildgrade adenomas with significant subcellular changes and in the majority of high-grade dysplasia adenoma suggests that this might be related to the carcinogenetic proceeding. Immunohistochemical localization of GST-pi in combination with ultrastructural changes indicate that GST-pi might be a sensitive agent for the detection of preneoplastic transformations in adenoma.

Precuneus contributes to attentive control of finger movement

AIM: To examine whether precuneus subserves the attentive control of finger movement or whether it mediates themovement preparation and motor inhibition. METHODS: In the Preparation Stage, subjects were shown with a 5-number string in which each number stood for a key-pressing response, the number strings included a complexpattern (eg, 4-1-4-2-3), or a simple one (eg, 2-2-2-2-2), or a null one (ie, x-x-x-x-x). In the Execution Stage, fivereaction signs were presented one by one and subjects were required to press the corresponding key to each signsequentially (eg, in the 4-1-4-2-3 preparation example, subjects press key 4 to the first sign, press key 1 to thesecond sign, key 4 to the third sign and so on). For the null preparation pattern, five numbers, rather than thereaction signs, were shown at the same pace as in the other two conditions and subjects were to press the corre-sponding keys. RESULTS: Left medial frontal gyrus (BA 6) and precentral gyrus (BA 6) were involved in both ofthe Preparation Stage and the Execution Stage, whereas left precuneus (BA 7) was activated only in the ExecutionStage. CONCLUSION: Precuneus mediates the attentive control of finger movement, but not the movementpreparation or motor inhibition.

Active gastrointestinal bleeding:Use of hemostatic forceps beyond endoscopic submucosal dissection

Identifying behavioral determinants and stage of readiness for performing knee massage among individuals with knee osteoarthritis:An observational study

Objective Patients who experience knee osteoarthritis or chronic knee pain can alleviate their symptoms by performing self-knee *** the readiness and types of determinants needed to facilitate self-knee massage is needed to design effective,theory-informed *** primary objective of this study was to apply the transtheoretical model of behavior change to identify how factors,which include the type of knee condition and pain level,predict an individual’s readiness to adopt self-knee *** secondary objective employed the capability,opportunity and motivation-behavior(COM-B)model to identify relevant determinants that are predictive of an individual’s readiness to undertake self-knee *** An observational study design was used to recruit individuals with knee osteoarthritis(n=270)and chronic knee pain(n=130).Participants completed an online survey that assessed the transtheoretical model of behavior change stages,COM-B determinants(capability,opportunity and motivation),along with self-administered massage *** analysis of covariance and structural equation modeling were used to test the primary and secondary objective,*** Participants who had knee osteoarthritis scored higher on the action stage compared to those with chronic pain(P=0.003),and those who experienced greater level of pain scored higher in the contemplation(P<0.001)and action phases(P<0.001)of performing knee massage compared to those with milder *** COM-B structural equation model revealed self-administered knee massage to be predicted by capability(β=0.31,P=0.004)and motivation(β=0.29,P<0.001),but not opportunity(β=–0.10,P=0.39).Pain level predicted motivation(β=0.27,P<0.001),but not capability(β=0.09,P=0.07)or opportunity(β=0.01,P=0.83).Tests for mediating effects found that determinants of COM-B(motivation and capability)mediate between pain level and self-administered massage behavior(β=0.10,P=0.002).Conclusion Cl

Feeding behavior and gene expression of appetite-related neuropeptides in mice lacking for neuropeptide Y Y5 receptor subclass

Neuropeptide Y (NPY) is a potent neurotransmitter for feeding. Besides NPY, orexigenic neuropeptides such as agouti-related protein (AgRP), and anorexi- genic neuropeptides such as α-melatonin stimulating hormone (MSH) and cocaine-amphetamine-regulated transcript (CART) are also involved in central feeding regulation. During fasting, NPY and AgRP gene expressions are up-regulated and POMC and CART gene ex- pressions are down-regulated in hypothalamus. Based on the network of peptidergic neurons, the former are involved in positive feeding regulation, and the latter are involved in negative feeding, which exert these feeding-regulated peptides especially in paraventricular nucleus (PVN). To clarify the compensatory mecha- nism of knock-out of NPY system on feeding, change in gene expressions of appetite-related neuropeptides and the feeding behavior was studied in NPY Y5-KO mice. Food intake was increased in Y5-KO mice. Fasting increased the amounts of food and water intake in the KO mice more profoundly. These data indicated the compensatory phenomenon of feeding behavior in Y5-KO mice. RT-PCR and ISH suggested that the compensation of feeding is due to change in gene expressions of AgRP, CART and POMC in hypothalamus. Thus, these fi ndings indicated that the compensatory mechanism involves change in POMC/CART gene expression in arcuate nucleus (ARC). The POMC/CART gene expression is important for central compensatory regulation in feeding behavior.

Systemic treatment in EGFR-ALK NSCLC patients:second line therapy and beyond

Lung cancer is the most frequently diagnosed cancer and a leading cause of cancer mortality worldwide, with adenocarcinoma being the most common histological subtype. Deeper understanding of the pathobiology of non-small cell lung cancer(NSCLC) has led to the development of small molecules that target genetic mutations known to play critical roles in progression to metastatic disease and to influence response to targeted therapies. The principle goal of precision medicine is to define those patient populations most likely to respond to targeted therapies. However, the cancer genome landscape is composed of relatively few "mountains" [representing the most commonly mutated genes like KRAS, epidermal growth factor(EGFR), and anaplastic lymphoma kinase(ALK)] and a vast number of "hills"(representing low frequency but potentially actionable mutations). Low-frequency lesions that affect a druggable gene product allow a relatively small population of cancer patients for targeted therapy to be selected.

Hippocampus’s role in forming “task-related” associations: Flashing to the things you are looking for

Eichenbaum and colleagues observed that the same place did or did not activate the "goal-approach" cells in hippocampus depending on whether the place was the way for rats to approach specific goal. Parallel with this, the present neuroimage study revealed that, the same type of items could activate the hippocampus more when it was related to the task at hand than when it not. Participants were scanned by fMRI while they made judgments on the type of relationships contained in the word-pairs (e.g., Does the word pair, "furniture-table", contain a "category-exemplar" relationship?). Event-related analysis revealed that the forming of "task-related" association activated hippocampus more than that of "task-unrelated", even if it was the same type of items, and, this hippocampal difference was not caused by the different judgment requirements, nor by the effects of "yes" response. Consistently, the post-judgment cued-recall test exhibited a better retrieval performance for "task-related" associations than for the same type but "task-unrelated" associations. Results also showed that, the semantic re- latedness between the to-be-associated individual words (e.g., the related word pair "healthy-hospital" versus the unrelated word pair "price-way") was not enough to activate the hippocampus when it was "task-unrelated". Generally, we proposed that, through participating in forming of "task-related" asso- ciations and consolidating of episodic memory, hippocampus enabled the organism to keep the in- formation that owned great survival values in mind for future usage.

Crete with its complex geomorphological history is the island with the highest number of endemism observed in animal and plant taxa throughout the Aegean *** other groups of organisms within Crete are well-studied,the freshwater gastropod fauna still remains poorly *** and Pseudamnicola,are 2 genera of freshwater springsnails,both present on the island,inhabiting springs and other freshwater ***,we conduct a comprehensive study on the distribution of the different genetic lineages of the 2 gastropod genera in order to assess the mode of their differentiation on the island and infer the actual number of species present in the *** these aims,sequence data from the mitochondrial gene were used and analyzed within a phylogenetic *** Bythinella,our results strongly support at least 5 delineated Bythinella *** Crete,which correspond to the already described species from previous studies with the addition of a new *** analyses reveal an old time-frame of differentiation with vicariant phenomena being more likely the main drivers shaping the present-day distribution of the genus’genetic *** Pseudamnicola,our data indicate the presence of at least 2 delineated Pseudamnicola *** a differentiation more consistent to an isolation-by-distance pattern of a relatively recent *** processes followed by isolation of the populations and/or recent speciation,seem to be the underlying process for the current distribution of Pseudamnicola lineages.
